Abstract

The invention provides a method for long-term preservation of a pumpkin mosaic virus, comprising the following steps: firstly, a virus source to be preserved is placed in a mothproof greenhouse for isolated active propagation; secondly, after outbreak of a disease, a median serious-disease leaf is taken; the leaf and 0.02M pH 7.0 phosphate buffer (PBS) are added into a mortar for grinding according to the proportion of 1 to 5; the leaf is grinded into starchiness; thirdly, a four-layer gauze is used for filtration and filter residue is removed; fourthly, filtrate is centrifugated for 30 minutes through 4000rpm, and residue is removed; fifthly, supernatant fluid is loaded into a glass pipe and then 0.8 percent of glutamine sodium stannate, 2 percent of glucose and 2 percent of propanetriolare added into the mixture according to the volume of crude virus extract, freeze-fried and then sealed and preserved.
