The method comprises the following steps: hybridizing a novel rice cytoplasm male sterile line as a female parent and the original cytoplasm source donor material of the cytoplasm male sterile line as a male parent; carrying out composite crossing of a present rice variety as a backcross male parent with the above F1; continuously backcrossing 2-3 times, wherein an individual plant of normal fertile pollens from a backcross colony is used as female parent, and a backcross parent is used as a male parent; carrying out bagging selfing of the individual plant of normal fertile pollens in a BC3-4F1 colony to obtain BC3-4F2; and screening the line of normal fertile pollens and being similar to the backcross parent in comprehensive properties in all the BC3-4F2 lines.
