The method comprises the steps as follows: corn endosperm nucleus DNA is extracted with a CTAB (cetyltrimethyl ammonium bromide) method; the content of 8-OHdG in the corn endosperm nucleus DNA is detected with an ELISA (enzyme-linked immunosorbent assay) method; and through the establishment of a seed aging model and detection of the oxidation level of the 8-OHdG in the corn endosperm nucleus DNA, the oxidative damage to the corn endosperm nucleus DNA is detected, and the single corn seed vigor is detected rapidly.
