The method includes: filling the graphite felt material in the anaerobic reactor to serve as a sludge carrier, wherein volume of the graphite felt material accounts for 30-45% of working volume of the reactor; inoculating anaerobic sludge on the graphite felt carrier, and standing for culture to enable the anaerobic sludge to be adsorbed on the graphite felt carrier; performing load improving domestication culture on the anaerobic sludge, leading the cassava alcohol wastewater which is diluted step by step into the reactor for backflow, enabling fixed sludge to fully contact with the cassava alcohol wastewater, and finishing domestication when COD removal rate and methane yield are stable; enabling the cassava alcohol wastewater to be treated to flow into the anaerobic reactor for treatment, wherein hydraulic retention time is 0.3-0.5 h.
