The method comprises the following steps: by virtue of hydrolysis of lysozyme, forming a transparent ring in a solid culture medium comprising a color source primer, wherein the color source primer is micrococcus lysodeikticus marked by an active dye bright-red, and in a proper concentration range, the vitality of lysozyme and the diameter of the transparent ring are of a linear relationship; drawing a standard gradient curve by taking lysozyme vitality unit as vertical coordinates and the diameter of the transparent ring as horizontal ordinates; and comparing the standard gradient curve with the standard gradient curved of a standard sample according to the diameter of the transparent ring of the sample, and calculating the vitality unit of the lysozyme in the sample. The method provided by the invention can be used for qualitatively and quantitatively detect the lysozyme, can exclude inference caused by factors such as colloids, and meanwhile, further can reflect the splitting specificity of the lysozyme to the bacterial cell walls. The transparent ring cannot be formed as other antibacterial components cannot decompose the bacterial cell walls dyed by the color source primer, so that the method is intuitive, fast, sensitive, simple and correct.
