Abstract
The invention relates to a culture method for reducing the brown change rate of apple explants. The culture method is characterized in that a collection and culture moment of apple explants is determined by using a total phenol content continuous testing method, and then the brown change rate of the apple explants can be reduced; when apples start to sprout, stems of 1-2cm are collected every other 7 days, the stems are grinded with liquid nitrogen, a sample is weighed, and the total phenol content of twigs is tested by using a foline-phenol method; the operation is implemented till the changeamplitude of total phenol contents of two contiguous times of testing is 0-10%. The technique method provided by the invention is simple in operation technique, low in brown change rate of explants in bottles, that is, the brown change rate is only 10% or lower, low in contamination rate, that is, the contamination rate is 8% or lower, and high in survival rate, that is, the survival rate of theapple explants can be up to 90% or greater. The technique method provided by the invention is wide in application range and is applicable to most apple varieties.
