Problems solved by technology

Although there are many methods of hydrolysis, the advantage of enzymatic degradation is that the action conditions are mild and the loss of sulfate groups can be avoided. The disadvantage is that it is not easy to obtain enzymes with high hydrolysis efficiency.
Enzymatic degradation of Porphyra laver polysaccharides has not been reported so far

Embodiment 1-1

[0044] Embodiment 1-1: The method for improving the biological activity of Porphyra laver polysaccharide comprises the following steps:

[0045] Dissolve the crude polysaccharide of Porphyra laver in acetic acid-sodium acetate buffer solution (pH 5.0), add pectinase (30,000 U / g), so that the final concentration of polysaccharide in the resulting system is 4 mg / mL, and the final concentration of pectinase is 50U / mL, the system was subjected to an enzymatic hydrolysis reaction for 2 hours at a pH of 5.0 and a temperature of 47.5°C to obtain a degradation solution;

[0046]Note: The pH of the system is controlled by the acetic acid-sodium acetate buffer solution, that is, the acetic acid-sodium acetate buffer solution with pH 5.0 is selected, and the pH of the obtained system is 5.0.

[0047] The acetic acid-sodium acetate buffer solution is a commonly used buffer solution, so those skilled in the art can prepare the acetic acid-sodium acetate buffer solution with the same pH val...

Embodiment 1-2

[0054] Change the final concentration of pectinase from "50U / mL" to "40U / mL" in Example 1-1, change the temperature during the reaction from "47.5°C" to "50°C", and the rest are the same as in Example 1- 1.

[0055] The scavenging rate of the obtained supernatant I to DPPH free radicals reaches 76.0%.

Embodiment 1-3

[0057] The temperature during the reaction in Example 1-1 was changed from "47.5°C" to "60°C", and the rest were the same as in Example 1-1.

[0058] The scavenging rate of the obtained supernatant I to DPPH free radicals reached 74.2%.

Abstract

The invention provides a method for improving the bioactivity of polysaccharides from porphyra haitanensis. The method includes dissolving crude polysaccharides from the porphyra haitanensis in buffersolution with the pH (potential of hydrogen) of 3.5-6.0, adding hydrolase into the buffer solution to allow the final polysaccharide concentration to be 2-10 mg / mL and the final hydrolase concentration to be 20-60 U / mL and then carrying out enzymatic hydrolysis reaction to obtain degradation liquid; carrying out water bath treatment on the degradation liquid to deactivate enzymes, and carrying out centrifugation to obtain supernatant I; carrying out dialysis on the supernatant I, carrying out rotary evaporation concentration on obtained dialysis solution until the volume of the dialysis solution reaches 20-30% of the original volume and carrying out freeze drying to obtain the degradation polysaccharides from the porphyra haitanensis. The invention further provides application of the degradation polysaccharides prepared from the porphyra haitanensis by the aid of the method. The application includes using the degradation polysaccharides from the porphyra haitanensis as functional foodbasic materials. The method and the application have the advantage that the in-vitro antioxidant activity, the cholic acid binding activity and the immune activity of the obtained degradation polysaccharides from the porphyra haitanensis can be obviously improved as compared with the crude polysaccharides from the porphyra haitanensis.

Description

technical field [0001] The invention belongs to the technical field of chemistry and chemical engineering, and relates to a method for enzymatically hydrolyzing crude polysaccharides of Porphyra laver to enhance its biological activity. Background technique [0002] Altar laver belongs to Rhodophyceae, Russaceae, commonly known as laver and Wucai. It is mainly planted in southern coastal areas such as Fujian and Zhejiang in my country. It is rich in protein, polysaccharides and vitamins. It is a kind of edible and medicinal value. Seaweed plants, so very popular with people. Porphyra is delicious, cheap, and rich in resources. Combining these characteristics, laver has become a research hotspot. Therefore, it has a good prospect to develop laver into functional food. Laver polysaccharide is a kind of galactan sulfate, which accounts for about 30% of the dry weight of laver. The chemical composition of laver polysaccharide extracted from different species, different seasons a...
