Problems solved by technology

[0005] In addition, due to the short half-life of small molecule drugs or photosensitizers in the blood, they will be quickly cleared from the blood and cannot effectively accumulate in the tumor site
Moreover, small molecule drugs often display severe toxic side effects on healthy tissues

Embodiment 1

[0047] Example 1: Preparation of nanocapsules FePtNBS@OVA

[0048] a) Dissolve 50 mg ovalbumin and 7.2 mg NaCl in 500 µL water at a stirring speed of 200 rpm. Add 250 µL to a concentration of 30 mg mL -1 Fe 3 o 4 NPs dispersion in water, 250 µL DMSO solution containing 20 mg cisplatin and 20 mg NBS.

[0049] b) 35.8 mg of surfactant P(E / B)-PEO was dissolved in 7.5 g of cyclohexane. Pour the surfactant mixture into the aqueous solution obtained in step a) at a stirring speed of 500 rpm. The emulsion was homogenized by sonication for 180 s (30 s of sonication with 10 s pauses per treatment) and cooled in ice water at 70% amplitude.

[0050] c) 10.7 mg of P(E / B)-PEO) was dissolved in 5 g of cyclohexane, and 2 mg of TDI (2,6-toluene diisocyanate) was added to this solution. The mixture was added dropwise to the miniemulsion obtained in step b) within 5 min, and allowed to react at 25 °C for 24 h. Afterwards, excess surfactant was removed from the dispersion with fresh cycl...

Embodiment 2

[0054] Embodiment 2: Preparation of ovalbumin-encapsulated Fe 3 o 4 Nanocapsule Fe@OVA of NPs.

[0055] a) Dissolve 50 mg ovalbumin and 7.2 mg NaCl in 500 µL water at a stirring speed of 200 rpm. Add 250 µL to a concentration of 30 mg mL -1 Fe 3 o 4 NPs dispersed in aqueous solution.

[0056] Steps b), c) and d) are the same as in Example 1.

Embodiment 3

[0057] Embodiment 3: Preparation of ovalbumin-encapsulated Fe 3 o 4 Nanocapsule FeNBS@OVA of NPs and photosensitizer NBS.

[0058] a) Dissolve 50 mg ovalbumin and 7.2 mg NaCl in 500 µL water at a stirring speed of 200 rpm. Add 250 µL to a concentration of 30 mg mL -1 Fe 3 o 4 NPs dispersion aqueous solution, 250 µL DMSO solution containing 20 mg NBS.

[0059] Steps b), c) and d) are the same as in Example 1.

Abstract

The invention discloses an ovalbumin nanocapsule with a synergistic effect of chemotherapy and photodynamic therapy, and a preparation method and application thereof, and belongs to the field of nanomedicines for treating cancers. The ovalbumin nanocapsule with the synergistic effect of chemotherapy and photodynamic therapy is an ovalbumin nanocapsule combining photodynamic therapy (PDT) and chemotherapy, and the purpose of synergistically enhancing cancer therapy is achieved on the basis of enhanced Fenton-like reaction. In the nanocapsule, even in a severe anoxic environment (2% O2), a photosensitizer depends on an I-type PDT mechanism and an intracellular biochemical reaction to efficiently generate high-toxicity -OH. The PDT process can mediate the escape of cis-platinum from the endosomes / lysosomes due to the effective disruption of the intracellular membrane system, thereby enhancing the targeted delivery of drugs. In addition, cis-platinum can intensify the generation of -OH through intracellular biochemical reaction. Therefore, an enhanced therapeutic effect can be achieved even under hypoxic conditions.

Description

technical field [0001] The invention relates to a class of nano-medicine in the field of cancer treatment and its preparation and application, in particular to an ovalbumin nano-capsule with synergistic effects of chemotherapy and photodynamic therapy and its preparation method and application. [0002] technical background [0003] Cancer has always been one of the major threats to human life. Various traditional cancer treatment methods including chemotherapy, radiotherapy, and new methods such as photodynamic therapy (PDT), chemodynamic therapy, etc. have been used to treat cancer. However, monotherapy often shows limited therapeutic efficacy due to tumor heterogeneity. Therefore, multimodal therapies combining different treatment methods are applied to overcome the above-mentioned problems. In particular, the strategy of PDT combined with chemotherapy showed unique advantages. The main reason is that the reactive oxygen species (ROS) generated during PDT not only direc...
