Problems solved by technology

However, these solutions still need to borrow instruments for fluorescence detection, which does not meet the dual needs of the current market for fast, sensitive and convenient detection.
For example, if everyone in a high-incidence area is screened by fluorescence detection, it will be very time-consuming and energy-consuming, and it will easily cause cross-infection among the crowd

Embodiment 1

[0083] Example 1: Two-site hybridization chain reaction amplification detection and detection of novel coronavirus single-stranded nucleotide molecules its kit

specific Embodiment approach

[0085] (1) Target sequence selection:

[0086] The single-stranded nucleic acid molecule to be amplified is selected as the single-stranded ribonucleic acid of the novel coronavirus corresponding to one specific target sequence in the deoxyribonucleotide, the sequence is shown in SEQ ID NO.1, and one non-specific target sequence is selected, The sequence is shown in SEQ ID NO.4,

[0087] (2) Sequence design:

[0088] The probe pair for hybridization chain reaction with the specific target sequence is H1a 1 and H2a 1 , the H1a 1 The sequence is shown in SEQ ID NO.2, the H2a 1 The sequence is shown in SEQ ID NO.3, and the probe pair that hybridizes with the non-specific target sequence is H1b 1 and H2b1, the H1b 1 The sequence is shown in SEQ ID NO.5, the H2b 1 The sequence is shown in SEQID NO.6. The sequence information mentioned in Example 1 is shown in Table 1 below. In the H2b 1 The 5' end of the probe increases a fluorescent group (FAM); the H1a 1 Probes and H1b...

Embodiment 2

[0111] Example 2: Three-site hybridization chain reaction amplification detection and detection of novel coronavirus single-stranded nucleotide molecules its kit

[0112] All the steps in Example 2 are completely consistent with those in Example 1, and will not be described again. The core difference lies in the selection of the target sequence and the design of the probe. specifically:

[0113] One of the specific target sequences is selected, the sequence is shown in SEQ ID NO.1, and the probe pair that hybridizes with the specific target sequence is H1a 1 and H2a 1 , the H1a 1 The sequence is shown in SEQ ID NO.2, the H2a 1 The sequence is shown in SEQ ID NO.3. Two non-specific target sequences are selected, including one non-specific target sequence (1) and one non-specific target sequence (2). The non-specific target sequence (1) The sequence is shown in SEQ ID NO.4, the sequence of the non-specific target sequence (2) is shown in SEQ ID NO.8, and the probe pair f...

Abstract

The invention provides a detection method and a detection kit for amplifying an output signal based on a hybridization chain reaction. The detection method comprises the steps: target sequence selection: selecting at least one specific target sequence and at least one non-specific target sequence of a to-be-amplified single-stranded nucleic acid molecule as target sequences; and hairpin probe design: designing corresponding groups of number probe pairs which are respectively subjected to hybridization chain reaction with the selected one or more specific target sequences, designing corresponding groups of number probe pairs which are respectively subjected to hybridization chain reaction with the selected one or more non-specific target sequences, enabling the one or more specific target sequences and the one or more non-specific target sequences to be subjected to hybridization chain reaction under the action of the corresponding probe pairs so as to amplify and enlarge output signals; and detecting by using a colloidal gold test strip based on the hybridization chain reaction. The invention also provides the detection kit corresponding to the method. The detection kit has the technical effects of convenience and sensitivity in detection.

Description

technical field [0001] The invention relates to the technical field of biological detection, in particular to a detection method based on hybridization chain reaction amplified output signals and a detection kit for single-stranded nucleic acid molecules. Background technique [0002] Principle of Hybrid Chain Reaction [0003] The relevant principles of hybridization chain reaction (Hybridization Chain Reaction, HCR) were first proposed by Dirks and Pierce in 2004 (see: Triggered amplification by hybridization chainreaction R.M. Dirks and N.A.Pierce, Proc.Natl.Acad.Sci.U.S.A., 2004, 101, 15275–15278). Hybridization chain reaction is a non-enzyme-assisted in vitro isothermal signal amplification technology, which belongs to the self-assembly signal amplification technology. Compared with thermocycling amplification techniques, including polymerase chain reaction (Polymerase Chain Reaction, PCR) and ligase chain reaction (Ligase Chain Reaction, LCR), hybridization chain re...
