Example Embodiment

[0025] Example 1:

[0026] Material: This example uses the immature seeds of Dapanshan beech as the starting material for cultivation.

[0027] Step (1): Material extraction

[0028]From late July to late August, the immature seeds of Dapanshan were collected in the Dapanshan National Nature Reserve in Pan'an County, Zhejiang Province. Figure 1 )。 Immature seeds are green and ovoid in shape. Can be stored in a sealed bag and stored at a low temperature of 4 °C.

[0029] Step (2): Seed surface sterilization

[0030] Wipe the seed surface with 75% ethanol and remove the wrapped turquoise pseudofond with forceps. On the ultra-clean bench, soak with 70% ethanol aqueous solution and shake for 30s, rinse with sterile water 3 times for 1 min each time. After soaking and shaking with an aqueous solution of mercuric chloride with a mass fraction of 0.1%, rinse with sterile water 5 times for 1 min each time. Finally, use sterile filter paper to absorb excess water from the surface.

Abstract

The invention discloses a method for nursing and culturing immature seeds of torreya grandis, which comprises the following steps: by taking the immature seeds of the torreya grandis in the national natural reserve of the torreya grandis as a starting material, performing surface disinfection, stripping immature embryos and endosperms of the immature seeds, nursing and culturing the endosperms for 2 weeks, stripping the endosperms, and independently culturing the embryos, thereby obtaining sterile seedlings of the torreya grandis. The method has the advantages that the immature seeds of the torreya grandis are utilized, on one hand, the nutrient component advantages of endosperm are fully utilized in the early stage to promote the growth and development of embryos, on the other hand, in the later stage, in order to reduce the influence of endosperm browning on the development of the embryos, the immature embryos are independently cultured in time, and a large number of sterile seedlings of the torreya grandis can be obtained within 45 days; and the current situation that torreya grandis is extremely endangered under natural conditions due to small population quantity is improved. The method is high in operability, simple, efficient and low in pollution rate, and has important significance on protection of endangered plants in the world and rapid cultivation of seedlings.

Description

technical field [0001] The invention relates to a method for nursing and cultivating immature Torreya chinensis seeds, so as to quickly and efficiently obtain Torreya chinensis seedlings, and achieve the purpose of expanding the population of endangered species and protecting the endangered species. Background technique [0002] Torreya dapanshanica (Torreya dapanshanica) is an evergreen tree belonging to the family Taxaceae (Torreya), with a height of 5 to 8 meters and a trunk diameter of 25 cm. Immature seeds have turquoise fleshy arils, 3.5 to 4 cm long and 2 to 2.5 cm wide. Seeds (without arils) broadly ovoid or globose, with hard woody seed coats. Seeds mature from September to November. Since Torreya jiulongshanensis was named, a rare group of Torreya jiulongshanensis was found in Dapanshan National Nature Reserve, Pan'an County, Zhejiang Province. It is similar to Torreya jiulongshanensis, but through accurate comparison of leaves and seeds, it was found that this ...
