Abstract

The present invention relates to methods for producing whole blood with an inactivated pathogen and to other compositions of blood products with an inactivated pathogen, as well as to kits and related compositions.

Claims

1. A kit for obtaining a whole blood composition with an inactivated pathogen, including: (a) a device for accessing blood vessels for collecting whole blood from a donor; (b) a first container suitable for mixing whole blood and a pathogen-inactivating compound (PIC), wherein the first container comprises at least a first inlet and a first outlet, and wherein the first container comprises an anticoagulant; (c) a first channel extending between the vascular access device and the first container and providing a fluid path for transferring whole blood from the vascular access device to the first container; (d) a container containing a pathogen-inactivating compound (PIC container); (e) the second channel, where the second channel: (i) is connected or designed to be connected at one end to a PIC container; (ii) is connected to the first container, designed to be connected to the first container, or designed to be connected to the first conduit, thereby providing a path for the passage of fluid between the PIC container and the first container; and (iii) further comprises a liquid supply device suitable for increasing the transfer of the PIC into the first container; and (f) an infusion line for the administration of pathogen-inactivated whole blood to the patient.

2. The kit of claim 1, wherein the PIC container is designed to be connected to the first container via a fluid passage.

3. The kit of claim 1, wherein the PIC container is designed to connect to the first container via a fluid passageway; or wherein the first container further comprises a second inlet, and the PIC container is designed to connect to the second inlet of the first container via a fluid passage; or wherein the first container further comprises a second inlet, and the PIC container is designed to be connected to the second inlet of the first container via a fluid passage.

4. A kit according to any one of paragraphs 1-3, further comprising a container containing an extinguisher (extinguisher container).

The kit of claim 4, wherein the extinguisher container is designed to connect to the first container via a fluid passageway, optionally wherein the extinguisher container is designed to connect to the first passageway or wherein the extinguisher container is designed to connect to the second inlet of the first container via a fluid passageway.

6. The kit of claim 4, wherein the extinguisher container is designed to be connected to the first container via a fluid passageway, optionally wherein the extinguisher container is designed to be connected to the second inlet of the first container via a fluid passageway.

7. A kit according to any one of paragraphs 4-6, wherein the second channel is connected at one end to a container for the damper.

8. A kit according to any one of claims 4 to 6, wherein the PIC container and the extinguisher container are the same container.

9. A kit according to any one of claims 1 to 8, wherein the second channel comprises at one end at least one adapter for connecting a container for a PIC to the second channel, optionally wherein the second channel comprises at one end a first adapter for connecting a container for a PIC to the second channel and a second adapter for connecting a container for a damper to the second channel.

10. A kit according to any one of claims 4 to 9, wherein the second channel is designed to connect to the first channel and provides a path for the passage of liquid between the extinguisher container and the first container.

11. A kit according to any one of claims 1 to 10, wherein the feeding device is further suitable for improving the transfer of the extinguisher into the first container.

The kit of any one of claims 1 to 11, further comprising a second container suitable for storing a whole blood composition with an inactivated pathogen, wherein the second container comprises at least a first inlet and a first outlet, optionally further comprising a third channel extending between the first container and the second container and providing a path for the passage of fluid for transferring a mixture containing the whole blood composition and the PIC from the first container to the second container, optionally wherein the mixture further comprises a quencher, if present.

The kit of claim 12, further comprising a third container suitable for storing the whole blood composition with the inactivated pathogen, wherein the third container comprises at least a first inlet and a first outlet, and wherein the kit further comprises a fourth channel extending between the second container and the third container and providing a fluid path for transferring the mixture containing the whole blood composition and the PIC from the second container to the third container, optionally wherein the mixture further comprises a quencher, if present.

The kit according to any one of claims 1 to 13, wherein the infusion line system is connected to the second container, optionally wherein the infusion line system comprises a second vascular access device and an infusion line channel extending between the first outlet of the second container and the second vascular access device, wherein the infusion line channel provides a path for the passage of fluid for sterile transfer of the whole blood composition with the inactivated pathogen, from the second container to the vascular access device.

15. The kit according to any one of claims 1 to 14, wherein the PIC comprises a functional group that is or that forms a reactive electrophilic group and / or wherein the PIC comprises a ligand that binds to a nucleic acid.

16. The kit according to any one of claims 4 to 15, wherein the quencher comprises a thiol group, wherein the thiol is capable of interacting with a reactive electrophilic group of PIC and / or wherein the quencher is glutathione or a pharmaceutically acceptable salt thereof.

17. A kit according to any one of claims 1 to 16, wherein the delivery device is an air delivery device; optionally, the delivery device comprises a sample collection pocket, a syringe, a rubber bulb, a flexible container or a cartridge; optionally, the delivery device is an air cushion or a sample collection bag.
