Description

Abstract This disclosure provides methods and compositions for preserving bacteria.

Claims

1. A lyophilized composition comprising bacteria belonging to the class Clostridia, a disaccharide, a nutrient, an antioxidant, sodium metabisulfite, and a buffer, wherein the pH of the buffer is 6.7, 6.8, 6.9, 7.0, 7.1, 7.2, or 7.3.

2. The composition of claim 1, wherein the nutrient is yeast extract, Luria-Bertani broth, or plant peptone.

3. The composition of claim 1 or 2, wherein the antioxidant is inulin, riboflavin, or cysteine.

4. The composition of any one of claims 1-3, wherein the disaccharide is trehalose or sucrose.

5. The composition of any one of claims 1-4, wherein the disaccharide is trehalose.

6. The composition of any one of claims 1-4, wherein the disaccharide is sucrose.

7. The composition of any one of claims 1-6, wherein the concentration of sodium metabisulfite is 0.05%.

8. The composition of any one of claims 1-7, wherein the bacteria belong to one or more of Clostridium clusters IV, XIVa, XVI, XVII, or XVIII.

9. The composition of any one of claims 1-8, wherein the bacteria comprise one or more bacterial strains belonging to species selected from the group consisting of Clostridium bolteae, Anaerotruncus colihominis, Drancourtella massiliensis, Sellimonas intestinalis, Clostridium symbiosum, Blautia producta, Dorea longicatena, and Flavonifractor plautii.

A method for preserving bacteria belonging to the class Clostridia, the method comprising adding bacteria belonging to the class Clostridia to a composition comprising a disaccharide, a nutrient, sodium metabisulfite and a buffer, wherein the pH of the buffer is 6.7, 6.8, 6.9, 7.0, 7.1, 7.2, or 7.3; and subjecting the composition comprising the bacteria to a lyophilization cycle.

11. The method of claim 10, wherein the nutrient is yeast extract, Luria-Bertani broth, or plant peptone.

12. The method of claim 10 or 11, wherein the antioxidant is inulin, riboflavin, or cysteine.

13. The method of any one of claims 10-12, wherein the disaccharide is trehalose or sucrose.

14. The method of any one of claims 10-13, wherein the bacteria belong to one or more of Clostridium clusters IV, XIVa, XVI, XVII, or XVIII.

15. The method of any one of claims 10-14, wherein the bacteria comprise one or more bacterial strains belonging to species selected from the group consisting of Clostridium bolteae, Anaerotruncus colihominis, Drancourtella massiliensis, Sellimonas intestinalis, Clostridium symbiosum, Blautia producta, Dorea longicatena, and Flavonifractor plautii.
