Abstract

This invention relates to a bio-bactericide formula to control corn stem rot disease caused by Dickeya zeae bacteria consisting of the rhizobacterial strain Bacillus cereus SrRG34, nutrient broth (NB) media enriched with 3% NaCL, 0.2% polysorbate 20 in the form of filtrate culture. The preparation of the bio-bactericide formula based on B. cereus SrRG34 is carried out with the following steps: growing B. cereus strain SrRG34 on Nutrient Agar (NA) media for 24 hours at 30°C; then taking one loop of B. cereus strain SrRG34 colonies is then inoculated into the Nutrient Broth (NB) medium enriched with 3% NaCL, incubated in a rotary incubator for 72 hours at a speed of 150 rpm, a temperature of 30°C; After that, the suspension of B. cereus strain SrRG34 was separated between the supernatant and pellet using a centrifuge at a speed of 3000 rpm for 15 minutes, a temperature of 5°C.The supernatant formed was filtered again using filter paper number 42 followed by filtration with a 0.22 µm syringe filter. Finally, the filtrate culture was mixed with 0.2% polysorbate 20 and homogenized using an Erlenmeyer shaker. The application of the bio-bactericide formula to control the disease was carried out by soaking the seeds for 24 hours before planting and spraying the plants, especially on the lower stem at 23 days after planting.
