Abstract
Description
Technology Field
[0001] The present invention generally relates to a class of acrylamido monomers, each of which contains (carbamoylphenyl)boronic acid and has a pKa of about 6.4 to about 7.8. Background Technology
[0002] Phenylboronic acid derivatives are known to be usable as receptors and sensors for carbohydrates and small molecules due to their ability to form cyclic boronate esters, primarily with cis-diols (see, e.g., SD Bull, et al., Acc. Chem. Res. 2013, 46 (2), 312-326; JA Peters, Coord. Chem. Rev. 2014, 268, 1-22). Recently, aryl boronic acid-containing vinyl monomers have been developed and used to prepare aryl boronic acid-containing polymers (e.g., T. Konno & K. Ishihara, Biomater. 2007, 28, 1770-1777; Z. Liu & H. He, Acc. Chem. Rev. 2017, 50, 2185-2193; WLA Brook & BS Summerlin, Chem. Rev. 2016, 116, 1375-1397; Z. Huang, et al., Biomater. Sci. 2018, 6, 2487-2495; JN Cambre & BS Summerlin, Polymer 2011, 52, 4631-4643; ZH Farooqi, et al., [Macromol. Chem. Phys. 2011, 212, 1510-1514]; [S. Roy, et al., Designed Monomers and Polymers 2009, 12, 483-495]; [D. Zhang & R. Pelton, Langmuir 2012, 28, 3112-3119]; [G. Vancoillie & R. Hoogenboom, Polym. Chem. 2016, 7, 5484-5495] see references).
[0003] These aryl boronic acid-containing polymers can form complexes with mucin, particularly epithelial mucin, by having an aryl boronic acid moiety. As such, aryl boronic acid-containing polymers have been reported to be usable in ophthalmic devices and ophthalmic compositions (see, e.g., U.S. Patents No. 7988988, 8419792, 8534031, 8631631, 8668736, 6596267, 8802075, 9244196, 10962803, 11439661; U.S. Patent Application Publication No. 2021-0077385). However, the vinyl aryl boronic acid monomers used in the manufacture of aryl boronic acid-containing polymers may have a pKa that is too high to ensure that the aryl boronic acid-containing polymers have strong interactions with mucin.
[0004] Therefore, there is still a need for arylboronic acid-containing vinyl monomers with low pKa.
[0005] In one embodiment, the present invention provides an acrylamido monomer comprising an acrylamido group and (carbamoylphenyl)boronic acid covalently bonded to the acrylamido group through bonding, wherein the acrylamido monomer has a pKa of about 6.4 to about 7.6.
[0006] In another aspect, the present invention provides a water-soluble hydrophilic copolymer comprising: (a) repeating monomer units of at least one acrylamido monomer of the present invention; (b) repeating units of at least one hydrophilic vinyl monomer; and optionally (c) repeating units of at least one hydrophilic vinyl crosslinker (provided that components (a) and (b) are present in the water-soluble hydrophilic copolymer in a total amount of at least 90 mol%).
[0007] In a further embodiment, the present invention provides an ophthalmic composition comprising: a water-soluble hydrophilic copolymer of the present invention. Specific details for implementing the invention

[0082] The water-soluble hydrophilic copolymer of the present invention may find special uses in developing ophthalmic compositions for topical application, particularly artificial tear compositions comprising compounds that lubricate and protect the surface of the eye. In the context of dry eye disorders, artificial tear compositions can prevent symptoms such as pain and discomfort, prevent friction-induced bioadhesion and tissue damage, and promote the natural healing and recovery of previously damaged tissues.
[0083] Ophthalmic compositions are typically developed to have a target viscosity to ensure they are comfortable for the user and do not cause undesirable side effects, such as blurred vision. A suitable viscosity can help ensure that ophthalmic compositions used for dry eye disorders alleviate dry eye-related symptoms and / or treat the underlying disorder. In drug delivery applications, the viscosity of an ophthalmic composition may be selected to ensure that the agent contained in the composition remains in the eye, preferably for a longer period of time.
[0084] Natural polymers or chemically modified derivatives thereof exhibiting thixotropic and viscoelastic properties are well known and have been used as lubricants and ocular surface protectants in many ophthalmic compositions in the prior art. Examples of such polymers include hydroxypropyl methylcellulose, galactomannan, e.g. guar and hydroxypropyl guar, carboxymethylcellulose, hyaluronic acid, sodium alginate, polyvinyl alcohol, etc. The shear thinning and viscoelastic profiles of these polymers play an important role when mixed with the tear film.
[0085] The copolymer of the present invention is believed to be able to form cycloboronate ester linkages with the 1,2-diol and 1,3-diol moiety present in these polymers to impart gel-forming behavior to ophthalmic compositions.
[0086] The ophthalmic composition may include one or more additional pharmaceutical active agents. In some embodiments, one or more pharmaceutical active agents may be selected from the group of phospholipids, ocular lubricants, anti-decongestants, e.g., brimonidine tartrate, tetrahydrozoline, naphazoline, cooling agents, e.g., menthol, steroids and non-steroidal anti-inflammatory drugs for relieving ocular pain and inflammation, antibiotics, antihistamines, e.g., olopatadine, antivirals for infectious conjunctivitis, antibiotics and antibacterial agents, anti-muscarinic agents for the treatment of myopia, e.g., atropine and derivatives thereof, and glaucoma drug delivery, e.g., prostaglandins and prostaglandin analogs, e.g., travoprost, and therapeutically suitable combinations thereof.

[0135] Example 1
[0136] (4-(2-acrylamidoethyl)carbamoyl)-3-chlorophenyl)boronic acid (CPBA) is prepared according to the procedure shown in Reaction Scheme 1.
[0137] [Reaction Equation 1]
[0138]
[0139] 3-3-chloro-4-carboxyphenylboronic acid (0.51 g, 2.52 mmol) and N-hydroxysuccinimide, NHS (0.32 g, 2.78 mmol) are loaded into a 40 mL vial equipped with a magnetic stirrer and a rubber diaphragm. Ethyl acetate (15 mL) is added through the diaphragm using a syringe. The contents of the vial are cooled to 0°C in an ice bath. 2 mL of diisopropyl-carbodiimide, DIC (0.35 g, 2.77 mmol) dissolved in ethyl acetate is added dropwise to the cooled solution of 3-chloro-4-carboxyphenylboronic acid and NHS in ethyl acetate. After 30 minutes, the ice bath is removed, and the reaction mixture is left to be stirred overnight at room temperature. The formed urea precipitate is removed by filtration, and the ethyl acetate is evaporated. The active ester was redissolved in DMF (10 mL), 2-(aminoethyl)-acrylamide xHCl (0.45 g, 2.99 mmol) was added, and the mixture was stirred for 10 minutes until all solids were dissolved. The contents of the reaction flask were cooled to 0°C in an ice bath. N,N-diisopropylethylamine, DIPEA (0.93 g, 7.20 mmol) combined with 2 mL of DMF was added dropwise to the vial containing the NHS-ester and 2-(aminoethyl)acrylamide xHCl. After 10 minutes, the ice bath was removed, and the reaction mixture was left to be stirred at room temperature for 5 hours. TLC showed the consumption of the NHS-ester. DMF was removed overnight using a high-vacuum pump, and the crude reaction mixture was purified by silica gel column chromatography (1.5 in × 12 in; CHCl3 → CHCl3:MeOH 100 → 90:10). The solvent was removed by rotary evaporation, and the final product was purified by crystallization from a MeOH / water mixture. A white solid was produced through crystallization at room temperature over 3 days, and this It was collected by filtering through a funnel. This was transferred to a vial and dried overnight under vacuum with an oil pump to obtain 0.372 g (1.25 mmol, 50% yield) of the final product as a white solid.

[0141] Example 2
[0142] In current Systane products, the mechanism of action relies on the formation of a hydroxypropyl-gua (HP-gua) gel using borates to extend the retention of demulcants, such as polyethylene glycol and propylene glycol, in the eye. This helps reduce the symptoms of dry eye by protecting the ocular surface. The challenge is that the current retention time of the demulcants in the eye is not long enough for patients to experience.
[0143] The water-soluble hydrophilic copolymer of the present invention is believed to be capable of cross-linking with mucin present in membrane-bound mucin primarily through reversible cyclo-boronate-ester bonds formed between one substituted (carbamoylphenyl)boronic acid of the hydrophilic copolymer and one of the cis-diol moiety of mucin, respectively, and secondarily through non-covalent interactions such as hydrogen bonds formed between the hydrophilic copolymer and mucin. As such, the water-soluble hydrophilic copolymer of the present invention can act as an active ingredient / lubricant and will effectively bind to membrane-bound mucin.
[0144] In addition, when a solution of the water-soluble hydrophilic polymer of the present invention is mixed with a solution of mucin of the eye, the interaction between the water-soluble hydrophilic polymer of the present invention and the mucin present in the membrane-bound mucin is believed to provide an improved viscosity compared to the sum of the viscosities of the two solutions. This improved viscosity may be a quantitative measure of the polymer-mucin interaction and is indicated in this application as a polymer-mucin interaction synergy.
[0145] The mucin selected for evaluating the interaction between the water-soluble hydrophilic copolymer and the mucin is porcine mucin due to its similarity to human eye mucin. Porcine stomach mucin, type III, is ordered from Sigma-Aldrich (catalog number M1778). The obtained mucin is purified by ultrafiltration before use for evaluating these polymer-mucin interactions.
[0146] In this application, the polymer-mucin-interaction synergy (indicated as PMIS) of the water-soluble hydrophilic copolymer of the present invention is calculated according to Formula 1:
[0147] [Equation 1]
[0148]
[0149] Here, is the viscosity of a first copolymer solution containing 1.0 wt% of a water-soluble hydrophilic copolymer in a phosphate buffered brine, and is the viscosity of a first mucin solution containing 0.6 wt% of porcine gastric mucin type III in phosphate buffered saline, and is the viscosity of the copolymer-mucin solution obtained by mixing the second copolymer solution with an equal volume of the second mucin solution, wherein the second copolymer solution contains 2.0 wt% of a water-soluble hydrophilic copolymer in phosphate buffered brine and the second mucin solution contains 1.2 wt% of porcine gastric mucin type III in phosphate buffered brine.
[0150] analyze
[0151] A stock solution (2 wt%) of the water-soluble hydrophilic copolymer of the present invention is prepared by dissolving freeze-dried MUC2 in DPBS using a speed mixer (1500~2000 RPM / 10 min / 2~3 times). A stock solution (2 wt%) of the water-soluble hydrophilic copolymer of the present invention is prepared by reconstituting and homogenizing the water-soluble hydrophilic copolymer in DPBS using a speed mixer (1500~2000 RPM / 10 min / 2~3 times).
[0152] A 1.2 wt% MUC2 solution is prepared by diluting a 2 wt% MUC2 stock solution with DPBS.
[0153] Equal volumes (1 mL) of 1.2 wt% MUC2 solution and DPBS are mixed in a 20 mL scintillation vial to form a 0.6 wt% MUC2 solution. The prepared 0.6 wt% MUC2 solution is tested for viscosity according to the procedure described below.
[0154] An equal volume (1 mL) of a 2 wt% hydrophilic copolymer solution and DPBS are mixed in a 20 mL scintillation vial to form a 1 wt% copolymer solution. The prepared 1 wt% copolymer solution is tested for viscosity according to the procedure described below.
[0155] Add 2 mL of 2.0% copolymer solution to a 20 mL scintillation vial equipped with a stirring bar. Place the vial on a stirring plate set to 500–600 RPM. Dropwise add 2 mL of 1.2% MUC2 solution over 10–20 seconds. Verify that the mixed solution exhibits free flow as a uniform viscous solution free of clusters or solidified gels. Mix the solution at 500–600 RPM for 30 minutes, then reduce the mixing speed to 150–200 RPM for 20–30 minutes to eliminate air bubbles.
[0156] Perform viscosity measurements on a DVNext Wells-Brookfield Cone / Plate Rheometer as follows. Place approximately 0.5 mL of sample into the sample cup. All viscosities are measured at room temperature using a size 40 spindle.

[0158] Copolymer synthesis
[0159] A desired amount of CPBA is dissolved in DMSO to prepare a stock solution of CPBA (4 wt%); and a desired amount of Vazo-56, MBA, or MPC is dissolved in deionized water to prepare a stock solution of Vazo-56 (1 wt%), MBA (1 wt%), and MPC (17 wt%).
[0160] The stock solution is directly combined in a jacketed reactor equipped with an overhead stirrer, a condenser, a thermocouple, and a nitrogen gas dispersion frit tube to satisfy the target amount as shown in Table 1. Subsequently, the remainder is filled with water to produce a final 5% solid solution ([Vazo-56] = 2,2'-azobis(2-methylpropionamidine) dihydrochloride).

[0164] Oxygen is removed from the polymerizable composition by purging with nitrogen. The frit of the gas dispersion tube is left over the polymerizable composition, and a nitrogen flow of approximately 50 mL / min is maintained throughout the polymerization. The temperature of the polymerizable composition is increased from 16°C to 56°C over 2 hours. After 10 hours, the temperature is lowered from 56°C to 20°C over 2 hours. The crude polymer solution is purified by ultrafiltration using a 1 M Dalton cutoff membrane to remove residual monomers, low MW polymers, and any residual solvent. For further characterization and testing, the purified polymer is freeze-dried.
[0165] The purified polymer is characterized. The results are provided in Table 2.

[0185] The polymer-mucin-interaction synergy (PMIS) of the water-soluble hydrophilic copolymer prepared in Example 3 is determined according to the procedure described in Example 2. The results are reported in Tables 6 and 7.
[0186] [Table 6]
[0187]
[0188] [Table 7]
[0189]
[0190] Table 6 shows a comparison of the strength of polymer-mucin interactions of the copolymer of the present invention (having repeating monomer units of CPBA with a pKa of about 7.2) and the strength of the control copolymer (having repeating monomer units of APBA with a pKa of about 8.1). The water-soluble hydrophilic copolymer of the present invention has stronger mucin interactions than the control copolymer.
[0191] Table 7 shows DMA (N H-결합 공여체 - N H-결합 수용체= 0 -1=-1) is HEAm (N H-결합 공여체 - N H-결합 수용체 It is shown that replacing with =2-2=0) significantly enhances polymer-mucin interactions. Viscosity increases from 46 cP (DMA) to 141 cP (HEAm), corresponding to PMIS of 426% and 1739%, respectively. These results may be due to the formation of hydrogen bonds between the HEAm-containing copolymer and mucin caused by the presence of H-bond donors in HEAm. On the other hand, the addition of MPC (20 mol%) to either copolymer reduces PMIS, i.e., polymer-mucin interactions (Table 7). While 2-methacryloyloxyethylphosphorylcholine (MPC) can weaken mucin interactions, HEAm appears to help restore interaction strength.
