[0090] Experimental Example 1: Confirmation of the Antioxidant Effect of a Complex of Red-Spotted Houttuynia Cordata Flowers, Leaves, and Stems
[0091] The antioxidant efficacy of the extracts of Comparative Examples 1 to 3 and the complexes of Examples 1 to 13 presented in Table 1 was confirmed by DPPH (2,2-diphenyl-1-picryl hydrezyl) radical scavenging activity.
[0092] Specifically, the extracts of Comparative Examples 1 to 3 and the complexes of Examples 1 to 13, each with a concentration of 5 μg / ml, were mixed with a 0.1 mM DPPH solution diluted in 4 ml of ethanol. The mixture was left at room temperature for 30 minutes, after which the absorbance was measured at 517 nm. For the untreated group, a DPPH solution without any mixture was used, and for the control group, ascorbic acid, which is used as a conventional antioxidant, was used at the same concentration.

[0101] The wrinkle and elasticity-improving effects of red houttuynia cordata were evaluated based on elastase inhibitory activity.
[0102] Elastase is an enzyme that breaks down elastin, a protein that maintains skin elasticity; inhibiting the activity of elastase can enhance skin elasticity and, consequently, lead to wrinkle improvement.
[0103] Elastase at a concentration of 0.5 U / ml (stock concentration) and N-succinyl-(L-Ala)3-p-nitroanilide at a concentration of 2 mM (stock concentration) were each dissolved in 50 mM tris-HCl buffer. Red Houttuynia cordata was mixed at different concentrations in 80 μL of tris-HCl buffer.
[0104] The above red Houttuynia cordata, 30 μL of elastase, and 30 μL of N-succinyl-(L-Ala)3-p-nitroanilide were mixed and reacted for 10 minutes. Afterward, the absorbance was measured at 410 nm. Ursolic acid was used as a positive control.

These results indicate that red houttuynia cordata has a skin elasticity-enhancing effect and, consequently, a wrinkle-improving effect.
[0107] Experimental Example 4: Confirmation of the whitening and pigmentation improvement effects of Red Houttuynia cordata
[0108] The whitening and pigmentation improvement effects of red Houttuynia cordata were evaluated based on tyrosinase enzyme inhibitory activity. Specifically, tyrosinase inhibitory activity was confirmed by measuring DOPA chrome, which is produced as a result of tyrosinase action, using a colorimetric method.
[0109] 110 μL of 0.1 M phosphate buffer (pH 6.5) was mixed with 40 μL of 1.5 mM L-tyrosine solution and 10 μL of red Houttuynia cordata at different concentrations. 10 μL of mushroom tyrosinase at a concentration of 2000 unit / mL was added, and the mixture was reacted at 37°C for 10 minutes. Subsequently, the absorbance was measured at 490 nm. Kojic acid at the same concentration was used as a positive control.
