Abstract
Description
Technology Field
[0001] The present invention relates to a novel amino acid derivative and a method for preparing the same. Background Technology
[0002] Amino acids are the building blocks of proteins (peptides), and they are compounds that possess both a basic amino group (-NH2) and an acidic carboxyl group (-COOH). Most are colorless crystals and are highly soluble in water. Amino acids are representative amphoteric electrolytes and are important components of proteins. When proteins are hydrolyzed by acids or enzymes (pepsin, trypsin, erepsin), they become various types of amino acids; among these, those in which the amino group and the carboxyl group are bonded to the same carbon atom are called α-amino acids.
[0003] Most of the amino acids contained in animal bodies are α-amino acids. These amino acids combine in a specific sequence to form proteins. When amino acids combine, a water molecule is released to condense and form an -CO-NH- bond. This bond is called a peptide bond or an amide bond. The proteins we consume are hydrolyzed into smaller sizes by enzymes in the stomach and small intestine, breaking down into amino acids that are then absorbed into the body. These absorbed amino acids are rearranged in a specific sequence according to genetic information to be synthesized into the proteins that make up our bodies.
[0004] Essential amino acids cannot be synthesized in the body and must be obtained from food for survival. Since amino acids obtained from food are water-soluble, they do not require a special transport system. Essential amino acids are isoleucine, leucine, phenylalanine, lysine, methionine, threonine, tryptophan, and valine. Non-essential amino acids include alanine, asparagine, aspartic acid, glutamic acid, glutamine, glycine, proline, serine, and tyrosine. Arginine and histidine are essential during the growth period. All essential amino acids and most non-essential amino acids possess one or more asymmetric carbon atoms and exhibit photoactivity.
[0005] Amino acids have the function of forming proteins, participating in metabolism, and creating neurotransmitters. They are involved in the growth and maintenance of tissues. Furthermore, they serve as major components of hormones, enzymes, and antibodies. Each possesses its own unique function.
[0006] When RNA synthesizes proteins by linking specific amino acids according to the DNA code, the chemical structure of the amino acids controls the structure of the protein, and this form of the protein determines the protein's function.
[0007] Furthermore, amino acids are converted into energy through metabolism when necessary. In addition, glutamic acid, cysteine, and glycine form glutathione, an antioxidant. Histidine plays a role in hydrogen transfer at enzyme reaction sites and is an important amino acid for red blood cell production and maintaining healthy nerves. Tyrosine is used in the production of thyroid hormones. Methionine converts into S-adenosylmethionine and participates in DNA and neurotransmitter metabolism through methylation. Arginine is said to generate nitric oxide, which relaxes vascular muscles to lower blood pressure and inhibits platelet formation in arteries.
[0008] Amino acids such as tryptophan and tyrosine form neurotransmitters. Tryptophan produces serotonin, while tyrosine synthesizes norephedrine and adrenaline. In the brain, tryptophan induces drowsiness, while tyrosine provides more energy. In addition, glutamate and aspartic acid are excitatory neurotransmitters.
[0009] As such, amino acids perform important functions within the body, and a deficiency can affect the regulation of human metabolism. Therefore, research and development regarding the synthesis of novel amino acids effective in regulating human metabolism or neurotransmission, as well as food and pharmaceutical compositions utilizing them, are continuously being conducted.
[0010] Meanwhile, protein hydrolysis, fermentation, and chemical synthesis processes are known methods for producing amino acids. The first amino acid produced industrially was glutamic acid. Glutamic acid is produced through a microbial fermentation process using sugars such as glucose, sugarcane, and starch as a raw material for monosodium glutamate (MSG).
[0011] However, existing methods for producing amino acids using microbial fermentation had problems such as high costs, low microbial stability, and unsuitability for mass production due to the complex processes involved, including culture, filtration, and purification. Prior art literature

[0019] Another aspect of the present invention provides a method for producing an amino acid derivative according to the following reaction formula 1, comprising the steps of: adding a basic substance to a solution containing a compound of the following formula (G2); and producing an amino acid derivative of the following formula (3) by changing the pH of the solution due to the basic substance.
[0020] [Reaction Equation 1]
[0021]
[0022] In the above reaction scheme, R is selected from hydrogen, hydroxyl, C1 to C12 alkyl group, C1 to C12 alkoxy group, C2 to C12 alkenyl, C1 to C12 alkylamine group, C3 to C12 cycloalkyl group, and M is a metal ion or ammonium ion.

[0027] According to the present invention, a novel amino acid derivative having efficacy similar to glutamic acid or glutamine derived from natural products can be produced by a chemical synthesis method.
[0028] The present invention has the advantage of low manufacturing costs, mass production, and high production efficiency because it can produce amino acid derivatives in high yield under mild conditions through a simple process of adding a basic substance to a solution in which a glutamine compound is dissolved to change the pH.
[0029] Furthermore, since this invention is a chemical synthesis method, various substituents can be introduced into the chemical structure, making it highly applicable to food, cosmetics, pharmaceuticals, and the like.
[0030] The novel amino acid derivative according to the present invention is a type of glutamine derivative. Glutamine is known to have the effect of reducing stress by influencing the release of neurotransmitters in brain nerve cells, and it has the effect of relieving tension and anxiety and lowering stress responses by contributing to an increase in alpha (α) waves in the brain or reducing the secretion of cortisol (stress hormone), thereby exhibiting sedative and stabilizing effects. The amino acid compound according to the present invention is an amino acid derivative with a chemical structure similar to glutamine and exhibits equivalent efficacy to the above, so it can replace conventional glutamine derivatives in manufacturing processes for food, cosmetics, or pharmaceutical compositions.

[0043] A method for preparing an amino acid derivative according to the following reaction formula 1 comprises the steps of: adding a basic substance to a solution containing a compound of the following formula (G2); and producing an amino acid derivative of the following formula (3) by changing the pH of the solution due to the basic substance.
[0044] [Reaction Equation 1]
[0045]
[0046] In the above reaction scheme, R is selected from hydrogen, hydroxyl, C1 to C12 alkyl group, C1 to C12 alkoxy group, C2 to C12 alkenyl, C1 to C12 alkylamine group, C3 to C12 cycloalkyl group, and M is a metal ion or ammonium ion.
