Abstract
Description
Technology Field
[0001] The present invention relates to a co-culture of salivary gland organoids and neuroblastomas, a method for preparing the same, and uses. Background Technology
[0002] Sialosis (sialadenosis) is a non-inflammatory, non-neoplastic, parenchymal lesion clinically characterized by recurrent bilateral swelling of the salivary glands. It typically manifests as bilateral and symmetrical lesions and is often painless, though tenderness may be present in some cases. Known causes of sialosis include nutritional deficiencies, various types of endocrine abnormalities, or autonomic nervous system disorders; however, neurogenic sialosis also exists, resulting from nervous system abnormalities or hyperactivity caused by factors such as unclear causes or uncontrollable stress. Generally, botulinum toxin is used to treat such neurogenic sialosis.
[0003] Botulinum toxin refers to Clostridium botulinum ( Clostridium botulinum It is a neurotoxic protein produced by bacteria. Functionally, it inhibits the secretion of the neurotransmitter acetylcholine at the presynapse of the neuromuscular junction, thereby inducing flaccid paralysis of the muscles. Recently, therapeutic attempts have been made to utilize the paralyzing mechanism of botulinum toxin for various autonomic nervous system dysfunctions and indications. However, while botulinum toxin has therapeutic effects in various diseases, it is highly toxic and can be fatal even in minute amounts; therefore, precise control of concentration is essential when using it in vivo.
[0004] Currently, to confirm the potency of Botox, LD50 is obtained through intraperitoneal injection in mice.50 Measurement methods and the CBPA method using neuroblastoma cell lines are widely used, and other methods using ELISA are also being developed. However, existing mouse-based methods are scheduled to be banned in the near future due to ethical issues, and there are already countries where they have been banned. Furthermore, since the CBPA method is patented by multinational pharmaceutical companies, there is a problem of reduced price competitiveness due to royalty payments.
[0005] Accordingly, the inventors completed the present invention by manufacturing a neurogenic salivary gland disease model based on human salivary gland organoids and confirming that it can be used to screen for novel therapeutic agents and measure the responsiveness of botulinum toxin drugs. [0099] Preferred embodiments are presented below to aid in understanding the present invention. However, the following embodiments are provided merely to facilitate a better understanding of the invention and do not limit the scope of the invention.
[0100] Example 1. Suspension co-culture system of human salivary gland organoids and N2a cells
[0101] Prior to proceeding with suspension co-culture, the culture time and initial cell number at which gene expression of ChAT, which provides acetylcholine to salivary gland organoids, was high during suspension culture of N2a cells alone were confirmed via qPCR, and the culture time was 24 hours and the initial cell number was 1.0 x 10⁶ 4 The number was set at 1.0 x 10⁶ per well in a 48-well plate. Therefore, the organoids were subcultured at a rate of 1.0 x 10⁶ per well in a 48-well plate. 4 The procedure was performed with 300 μL of cells without Matrigel. The composition of the culture medium is as shown in Table 3 below.
[0102] Advanced DMEM / F12 Basal medium Glutamax 1 X HEPES 1 X Primocin 0.2% B27 vit A+ 1 X NAC 1 mM Nicotinamide 5 mM A83-01 5 μM PGE2 3 μM hRSPO3 CM 1% hNoggin CM 2% hNRG1 5 ng / mL hFGF2 5 ng / mL hFGF10 10 ng / mL Y-27632 10 μM Forskolin 10 μM
[0103] After 72 hours had elapsed, 100 μL of the above culture medium was added, and after another 72 hours had elapsed, 1.0 x 10⁶ N2a cells were added to 1 mL of culture medium. 5 100 μL of the diluted solution was added to each well for co-culture. The experiment was terminated after 24 hours, and a post-hoc analysis was performed.
[0104] Example 2. Characterization of a suspension co-culture system of human salivary gland organoids and N2a cells
[0105] 2.1. Lumen swelling
[0106] The extent of lumen expansion was compared through bright-field observation with and without co-culture of human salivary gland organoids and N2a cells (Fig. 2). As a result, it was confirmed that the lumen of the salivary gland organoid expanded only when co-cultured with N2a cells.
[0107] 2.2. Immunofluorescence staining
[0108] Immunofluorescence staining was performed to confirm that KU80, which labels the nucleus of human cells, and TUJ1, which is not expressed in salivary gland epithelial cells but is expressed only in N2a cells, were expressed in a co-culture system of human salivary gland organoids and N2a cells. Meanwhile, salivary gland epithelial cell markers such as CDH1, KRT7, KRT5, KRT14, and KRT19 were confirmed to be expressed in both the group co-cultured with N2a cells and the group not co-cultured (Fig. 3).
[0109] Example 3. Model of neurogenic salivary gland disease using a suspension co-culture system of human salivary gland organoids and N2a cells and drug efficacy test using the same
[0110] To determine the effects of treatment with or without neurotransmitters such as acetylcholine and neostigmine on the co-culture system, brightfield observation was performed on the group treated with neurotransmitters (Acetylcholine+Neostigmine) and the group with neurotransmitter blockade by treatment with Botox (GT1b+BoNT / A), and the expression levels of luminal-related genes (KRT7, KRT19) were measured.
[0111] Observation with a bright-field microscope confirmed that even though the organoid's lumen expanded due to co-culture with N2a cells and neurotransmitters, the lumen contracted when the neurotransmitters were blocked (Fig. 4). In addition, an examination of the expression of lumen-related genes revealed that the expression levels of KRT7 and KRT19 increased when neurotransmitters were supplied and decreased when the neurotransmitters were blocked (Fig. 5).
[0112] In other words, it was confirmed that a neurogenic salivary gland disease model can be effectively prepared by treating a co-culture system of human salivary gland organoids and N2a cells with neurotransmitters. This appears to be because the treatment with acetylcholine and neostigmine increased the production of acetylcholine in the neuroblastoma, thereby mimicking stimulation by the parasympathetic nervous system. Furthermore, the efficacy of the drug against the disease was evaluated by treating the aforementioned neurogenic salivary gland disease model with Botox.
[0113] Example 4. Mouse salivary gland organoid-based neurosialadenopathy model and Botox potency test using the same
[0114] 4.1. Determination of concentrations of acetylcholine, neostigmine, GT1b, and Botox that do not affect mouse salivary gland organoids
[0115] Mouse salivary gland organoids were obtained using the same method as in Experimental Example 3 above, and the composition of the culture medium used is as shown in Table 4 below.
[0116] Advanced DMEM / F12 Basal medium Glutamax 1 X HEPES 1 X P / S 1 X Primocin 1 X B27 1 X NAC 1.25 mm A83-01 0.5 μM mRSPO1 1 % hNRG1 37.5 ng / ml mFGF1 79.15 ng / ml mFGF7 18.8 ng / ml Y-27632 10 μM
[0117] To prevent contamination by mesenchymal cells, the effect of Botox on organoids was confirmed using mouse salivary gland organoids from passage 2 onwards through observation of cell morphology and measurement of cell viability (Fig. 6).
[0118] As a result of treating mouse salivary gland organoids with various concentrations of acetylcholine, it was confirmed that increasing the concentration of acetylcholine up to 100 nM had no effect on the morphology and growth of the organoids (Fig. 7). Even when neostigmine was treated at 0, 50, 100, and 200 μM, no significant changes were observed in the morphology and growth of the organoids (Fig. 8). In addition, it was confirmed that treatment with 25 μg / mL of GT1b and various concentrations of botox had no effect on the organoids (Fig. 9).
[0119] 4.2. Co-culture system of mouse salivary gland organoids and N2a cells
[0120] N2a cells were cultured and subcultured using the same method as in Experimental Example 4 above, and the composition of the culture medium used is as shown in Table 5 below.
[0121] EMEM Basal medium P / S 1X FBS 10%
[0122] Co-culture was carried out by mixing 4 μL of organoid culture medium containing 300 cells constituting mouse salivary gland organoids and 4 μL of organoid culture medium containing 3,000 N2a cells with 32 μL of Matrigel to form a 40 μL dome and dispensing it into a 24-well plate (Fig. 10).
[0123] Figure 11 shows the salivary gland organoid (black arrow) and N2a cells (red arrow) observed under a bright-field microscope 7 days after the start of co-culture. Single-cell transcriptome analysis revealed that the Tubb3 gene was not expressed in epithelial cells but was expressed in N2a cells, confirming that the gene could serve as a good indicator for distinguishing between salivary gland organoids and N2a cells (Figure 12). Based on this, immunofluorescence staining of KRT5 (basal cells), KRT7 (lumen cells), AQP5 (adenocyte cells), ACTA2 (myoepithelial cells), and TUBB3 confirmed that a co-culture system between salivary gland organoids and N2a cells had been established (Figure 13).
[0124] 4.3. Botox Potency Test Using a Co-culture System of Mouse Salivary Gland Organoids and N2a Cells
[0125] On day 6 of co-culture of salivary gland organoids and N2a cells, the experiment was conducted by dividing the cells into three groups: Untreated (-Stim-Botox), Stimulated (+Stim-Botox), and Stimulated+Botox (+Stim+Botox). +Stim was indicated when 1 nM of acetylcholine and 200 μM of neostigmine were treated simultaneously, and +Botox was indicated when 25 μg / mL of GT1b and 200 U / mL of Botox were treated simultaneously.
[0126] After 24 hours, the co-cultured wells were dissolved in Trizol, and mRNA was reverse transcribed into cDNA to perform qPCR. In the case of the ChAT gene, which is required for the synthesis of acetylcholine and is mainly expressed in N2a cells, the expression level did not change even after 24 hours had passed since treatment with Botox, whereas the AchE gene, which is required for the degradation of acetylcholine, increased with treatment with acetylcholine and neostigmine (+Stim) and decreased again with treatment with Botox (+Botox) (Fig. 14).
[0127] In addition, Krt5 (basal cells), Krt7 (lumen cells), Aqp5 (aqueous cells), and Bhlha15 (aqueous cells) expressed in salivary gland organoids were increased by treatment with acetylcholine and neostigmine, and decreased again by treatment with Botox (+Botox), and a similar trend was observed in Bhlha15 (Fig. 15).
