Abstract
The present invention discloses a method for preparing a two-stage controllable dual-ligand chromatographic medium, belonging to the field of chromatographic separation technology. The steps of the method for preparing the two-stage controllable dual-ligand chromatographic medium include: first activating the chromatographic matrix, then brominating it, and then coupling the first hydrophobic charge-induced ligand to obtain a single-ligand chromatographic medium; the single-ligand chromatographic medium is coupled with the second hydrophobic charge-induced ligand to obtain a dual-ligand chromatographic medium. This invention selects two types of hydrophobic charge-induced layering agents as the object for combination, these types of layering agents include various binding modes such as ion exchange, hydrophobic interaction, and hydrogen bonding. They possess unique adsorption selectivity, high capacity, salt tolerance, and relatively lower costs, forming a dual-layered medium with natural competitive advantages.
