Abstract

The present invention relates to a plant molecular marker primer design method, specifically a whole genome SSR-specific molecular marker development method based on disease resistance sequences of common wheat, which solves the problems in existing wheat disease resistance breeding SSR markers of having excessive loci and being relatively distant from target genes. The operational steps are as follows: 1. Downloading whole genome data and establishing a local database; 2. Detecting the database using hidden Markov models of disease resistance gene families, screening the obtained sequences to obtain protein sequences containing target disease resistance structural domains, and then retrieving the local database to obtain corresponding genomic sequence fragments (scaffolds); 3. Detecting simple sequence repeat loci in each scaffold sequence, designing primers on both sides of the SSR loci, and synthesizing the primers; 4. Screening specific markers; 5. Detecting marker polymorphism. The nucleotide sequences of the SSR-specific molecular markers are the sequences shown in SEQ ID NO.1 to SEQ ID NO.78. Advantages: 1. Clear positional information and strong specificity; 2. Extremely close linkage to disease resistance candidate genes.
