Abstract

The invention relates to cells designed for adoptive therapy, including t cells. The invention also relates to methods and compositions for engineering and producing the cells, compositions containing the cells, and a method for administering them to subjects. In some embodiments, the cells, such as t cells, contain genetically modified antigen receptors that bind specifically to antigens, such as a chimeric antigen receptor (car). In some embodiments, the cells, such as a car-expressing t cell, contain an agent capable of reducing an inhibitory effect by repressing and / or disrupting a gene in a manipulated cell, such as a gene involved in inhibiting the immune response. In some embodiments, the characteristics of the cells and the methods allow for increased or improved activity, efficacy, and / or persistence.
