Abstract
Description
Synthesis method of 1H-furo[3,2-b]imidazo[4,5-d]pyridine compound Technical Field
[0001] The present invention relates to the technical field of drug synthesis, in particular to a method for synthesizing a 1H-furo[3,2-b]imidazo[4,5-d]pyridine compound. Background Art
[0002] 1H-furo[3,2-b]imidazo[4,5-d]pyridine compounds are highly selective JAK1 and TYK2 inhibitors that selectively inhibit the activity of one or more protein kinases relative to other related kinases. Therefore, they are expected to be useful in treating diseases mediated by the selectively inhibited one or more kinases while avoiding the adverse side effects associated with the inhibition of other kinases. For example, they can be used to treat disorders related to JAK1 / TYK2 activity, such as autoimmune diseases or disorders, or inflammatory diseases or disorders, as well as cancer or tumor diseases or disorders.
[0003] Existing synthesis methods for such compounds are costly and complex, making them unsuitable for industrial scale production (WO 2018 / 067422 A1, WO 2020 / 244348 A1, WO 2020 / 244349 A1). Consequently, there is a need to develop a method for synthesizing 1H-furo[3,2-b]imidazo[4,5-d]pyridine compounds that is both high in yield and low in impurities, easily controllable, cost-effective, and simple to operate, making it suitable for industrial scale production.

[0285] Loss on drying:
[0286] Take the mixed sample of Compound 1 to be tested (if the sample is relatively large crystals, quickly crush it into particles approximately 2 mm in diameter). Accurately weigh 1-2 grams and place it in a flat, stoppered weighing bottle that has been dried under the same conditions as the sample. Tare the bottle first. After adding the sample, cap the bottle and weigh the total weight. Gently shake the bottle horizontally to spread the sample as evenly as possible, with a thickness of no more than 5 mm. Place the weighing bottle containing the sample in a constant temperature, reduced pressure drying oven (with the bottle cap removed and placed in a desiccator) and dry at 50°C and -0.09 to -0.1 MPa for 4 hours. Immediately cap the oven-dried sample after drying, transfer it to a desiccator and cool it to room temperature before weighing.
