Abstract
The present disclosure relates to a method for preparing a lateral flow assay (LFA) biosensor strip, specifically designed for the detection of Porphyromonas gingivalis (21), a bacterium associated with periodontal disease. The method involves assembling a biosensor strip comprising a backing card (14), a sample pad (10), a conjugate pad (11), a nitrocellulose membrane pad (12), and an absorbent pad (13) in fluid communication. A unique aspect of this method is the preparation of a conjugate reagent solution by conjugating polyclonal anti-kgp antibodies against gingipains kgp with gold nanoparticles, which is then applied onto the conjugate pad (11). Additionally, test (T) and control (C) reagent solutions are prepared and applied onto designated regions on the membrane pad (12), facilitating the detection of Porphyromonas gingivalis (21) through specific immunological reactions. The conjugate and membrane pads (12) are subjected to specific incubation conditions to enhance the binding efficiency of the reagents. This method emphasizes the importance of precise assembly and preparation techniques to ensure the functionality and reliability of the LFA biosensor strip for rapid, accurate detection of periodontal disease markers, offering a valuable tool for both clinical and field settings.
