Abstract

Tricyclic compounds that modulate PI3K(alpha) activity are provided, which are useful in the treatment of various diseases, including cancer. A compound of Formula I or a pharmaceutically acceptable salt thereof, wherein: n is 0, 1, 2, 3, 4, 5, or 6; ring A is C3-14 cycloalkyl, C6-10 aryl, among others; L1 is C1-4 alkylene optionally substituted with 1, 2, 3, or 4 independently selected RG substituents; L2 is -O- and -N(RL)- linkage; RL is H, C1-6 alkyl, and C1-6 haloalkyl; R1 is H, halo, and C1-4 alkyl, among others; R2 is H, C1-6 alkyl, and C1-6 haloalkyl, among others; R3 is H, halo, and C1-6 alkyl, among others; R4 is oxo, halo, C1-6 alkyl, among others.
