Abstract

The present invention relates to a method for the preparation of 1-((3S,4R)-3-((2-((1-ethyl-1H-pyrazol-4-yl)amino)-7H-pyrrolo[2,3-d]pyrimidin-4-yl)oxy)-4-fluoropiperidin-1-yl)prop-2-en-1-one, and its intermediate compounds.A method for preparing a compound represented by Chemical Formula 1, the method comprising: (step 1) reacting a compound of Chemical Formula 1-1 and a compound of Chemical Formula 1-2 in the presence of a base to prepare a compound of Chemical Formula 1-3; (step 2) reacting a compound of Chemical Formula 1-3 and a compound of Chemical Formula 1-4 in the presence of a palladium catalyst and a base to prepare a compound of Chemical Formula 1-5; (step 3) reacting a compound of Chemical Formula 1-5 with an acid to prepare a compound of Chemical Formula 1-6; and (step 4) reacting a compound of Chemical Formula 1-6 and a compound of Chemical Formula 1-7 in the presence of a base to prepare a compound of Chemical Formula 1, wherein at least one of steps 1 to 4 further comprises a crystallization step: [Chemical Formula 1] wherein in Chemical Formulas 1-1 to 1-7, P1 signifies a protecting group.
