Abstract

Disclosed herein is the production of antibodies against the protrusion domain (Asp201-Gly339) of the VP2 protein (NCBI Acc. No. KT281984) of Infectious Bursal Disease Virus (IBDV) in rabbits, followed by purification through protein-A affinity chromatography. The purified anti- VP2 protrusion domain antibodies (IgGs) are then conjugated with gold nanoparticles and incorporated into dip-stick and lateral flow strip-based diagnostic assays for rapid and efficient on- site detection of IBDV in poultry. These diagnostic tools comprise a conjugate pad containing the gold nanoparticle-conjugated antibodies, a nitrocellulose membrane with a test line (coated with anti-VP2 protrusion domain antibodies) and a control line (coated with VP2 protrusion domain antigen), an absorbent pad, and a sample pad for introducing triturated biological samples from IBDV-infected organs of chickens. Upon sample application, the IBDV VP2 antigen, if present, interacts with the gold-conjugated antibodies, forming an antibody-antigen complex that migrates along the nitrocellulose membrane, binding to the test line and producing a visible colorimetric signal within 10-15 minutes This invention offers a field-deployable alternative to conventional laboratory-based diagnostic methods. By leveraging the high specificity of anti-VP2 protrusion domain antibodies, this invention provides a process of detection of local IBDV strains, enabling early intervention and improved disease management in poultry farms.
