Abstract
The present study describes a computational approach known as relative synonymous codon usage (RSCU) which was used to enhance the expression of interferon lambda protein in a eukaryotic system. Manually designed and synthetically made Interferon Lambda (IFN-λ) gene was cloned into pET-22b expression plasmid under the control of inducible T7-lac promoter. Interferon Lambda (IFN-λ) was observed on SDS-PAGE as a protein of 22 kDa after transformation in BL21 (DE3) cells. The culture parameters were optimized and it was observed that maximum level of Interferon Lambda (IFN-λ) expression was observed by inducing transformed E. coli BL21 cells with 0.4 mM of IPTG for 4 hours in LB ampicillin medium at 127 rpm and 37C. Increasing the inducer concentration beyond 0.5 mM did not improve overall expression of Interferon Lambda (IFN-λ). After purification, this locally produced IFN-λ can be tested as a targeted treatment option for viral hepatitis. Cost effective local production of this important cytokine can help alleviate the financial burden of people suffering from these lethal diseases.
