Abstract

The subject of the application is a method for assessing the quality of tomato seeds, which consists in subjecting tomato seeds to a purification pre-treatment in such a way that the seeds are freeze-dried by known methods, ground and extracted by shaking in formic acid at a concentration of 1.9 to 2.1 mM, preferably 2 mM, and sonication, then centrifuged and the supernatant obtained in this way is collected and the remaining material is subjected to another purification pre-treatment at least once, then the obtained supernatants are combined, mixed and diluted with ammonium acetate at a concentration of 49.9 to 50.1 mM, preferably 50 mM and pH from 4.49 to 4.51, preferably 4.5, in an amount of 1.99 to 2.01 ml, preferably 2 ml of ammonium acetate per 2 ml of supernatant, and the supernatant samples prepared in this way are subjected to purification on hybrid SPE columns in this way the supernatant is passed from 1 to 10 times, preferably 3 times, through SPE columns in portions of 0.5 ml to 1.21 ml, preferably 1.2 ml,washes the hybrid SPE columns successively with ammonium acetate and methanol by passing the solution through the hybrid SPE column bed and elutes the nucleotides from the hybrid SPE column bed with a solution of methanol, water and ammonium acetate in a ratio of 1.9 / 6.9 / 0.9 to 2.1 / 7.1 / 1.1, preferably 2 / 7 / 1, wherein the hybrid cleaning is carried out at a temperature of 18 to 30°C, preferably 20°C, maintaining the liquid flow at a level not higher than 5 ml / min, preferably 3 to 4 ml / min, then the thus obtained and collected eluate of the nucleotide fraction is evaporated in a stream of gaseous nitrogen and the remaining precipitate is dissolved in a solution of acetonitrile and water in a ratio of 5% to 7% acetonitrile, preferably 6% acetonitrile in water, using 50 to 250 µl, preferably 100 µl of this solvent and the precipitate thus obtained are subjected to high-pressure liquid chromatography analysis in a UHPLC device, and then the obtained analytical graphs are analyzed.,
