Claims
1. Use of a substance for knocking out a gene encoding a protein or inhibiting the content or activity of a protein in the production of L-lysine; wherein the protein comprises A1) or A2), as specified below: A1) a protein having the amino acid sequence of SEQ ID NO: 2; and A2) a fusion protein obtained by attaching a label to the N-terminus and / or C-terminus of A1).
2. The use according to claim 1, wherein said coding gene comprises b1), or b2), or b3), as indicated below: b1) a DNA molecule as depicted in SEQ ID NO: 1 in the sequence listing; b2) a DNA molecule that has 75% or more identity with the nucleotide sequence defined in (b1) and encodes the said protein; and b3) a DNA molecule that, under stringent conditions, hybridizes with the nucleotide sequence defined in (b1) or (b2) and codes for the specified protein.
3. Use according to paragraph 1 or 2, characterized in that the said substance contains B1) or B2): B1) a nucleic acid molecule that reduces the expression level of the protein according to claim 1; and B2) an expression cassette, recombinant vector, recombinant microorganism or transgenic cell line containing a nucleic acid molecule according to B1).
4. The use according to any one of claims 1 to 3, characterized in that said substance is a substance that mutates the codon of the glutamate residue at position 414 in SEQ ID NO: 2 into a terminator; or said substance is a substance that mutates the guanine nucleotide at position 1240 in SEQ ID NO: 1 into a thymine nucleotide.
5. A method for producing L-lysine, comprising reducing the content or activity of the protein according to claim 1 in a recipient biological cell or knocking out the gene encoding the protein according to claim 1 in a recipient biological cell to produce a recombinant biological cell; and culturing said recombinant biological cell to produce L-lysine.
6. The method according to paragraph 5, characterized in that said biological cell is a yeast, bacterium, algae, fungus, plant cell or animal cell that is capable of synthesizing L-lysine.
7. The method according to claim 6, wherein the bacterium is Corynebacterium glutamicum.
8. The method according to any one of paragraphs 5-7, characterized in that said recombinant biological cell is cultured using a medium that allows for the growth of said recombinant biological cell; and / or the said recombinant biological cell is cultured using conditions that allow the said recombinant biological cell to grow.
9. The method according to any one of claims 5-7, characterized in that said method is implemented by mutating the codon of the glutamate residue at position 414 in SEQ ID NO: 2 in said recipient biological cell into a terminator, or mutating the guanine nucleotide at position 1240 in SEQ ID NO: 1 into a thymine nucleotide, or knocking out the gene as presented in SEQ ID NO:
1.
10. The method according to claim 9, characterized in that said recombinant biological cell is cultured using a medium that allows for the growth of said recombinant biological cell; and / or the said recombinant biological cell is cultured using conditions that allow the said recombinant biological cell to grow.
11. Biological material that contains b1), or b2), or b3), or b4), or b5), as specified below: b1) a DNA molecule encoding a protein with an amino acid sequence as presented in SEQ ID NO: 6; b2) a DNA molecule that has 75% or more identity to the sequence of the DNA molecule defined in b1) and encodes a protein as set forth in SEQ ID NO: 6; b3) a DNA molecule that hybridizes under stringent conditions to the nucleotide sequence defined in (b1) or (b2) and encodes a protein as set forth in SEQ ID NO: 6; b4) an expression cassette, a recombinant vector, a recombinant microorganism, or a transgenic cell line containing a DNA molecule according to (b1), (b2), or (b3); and b5) a recombinant biological cell according to any one of paragraphs 5-9.
12. A product for obtaining L-lysine, which contains a substance according to any of paragraphs 1-4 or a biomaterial according to paragraph 11.
