Abstract

FIELD: veterinary cosmetology.SUBSTANCE: grooming product for dogs suffering from alopecia X, intended for application to the skin during grooming. Description

[0001] The invention relates to the field of veterinary cosmetology, namely to the composition and method for producing a care product for dogs suffering from alopecia X, and can be used in grooming to restore the structure of the coat, moisturize, protect, nourish the skin and stimulate hair growth.

[0002] A method is known for reducing hair loss and / or stimulating hair growth and / or renewing growth (RU2535120C2, published 12 / 10 / 2014), comprising the steps of applying to the skin of a mammal in need of such treatment a safe and effective amount of a composition containing from about 0.1% to about 100% (by weight) of an additive containing:

[0003] a. from about 0.01% to about 99.98% (by weight of the additive) sodium pyruvate;

[0004] b. from about 0.01% to about 99.98% (by weight of the additive) of an antioxidant; and

[0005] c. from about 0.01% to about 99.98% (by weight of the additive) of a fatty acid mixture or a source of a fatty acid mixture comprising at least 7 unsaturated or saturated fatty acids selected from the group consisting of butyric acid, caproic acid, caprylic acid, capric acid, lauric acid, myristic acid, myristoleic acid, palmitic acid, palmitoleic acid, stearic acid, oleic acid, linoleic acid, linolenic acid, arachidic acid, gadoleic acid, pentadecanoic acid, margaric acid, margaroleic acid, behenic acid, dihomolinoleic acid, arachidonic acid and lignoceric acid.

[0006] A topical minoxidil composition is known (US2019083492A1, published 03 / 21 / 2019), including a minoxidil composition for topical use containing 1,3-propanediol as a solubilizing agent, ethyl alcohol and water, where ethyl alcohol: water: 1,3-propanediol are in a ratio of 1: 4.5: 10 by weight for a foam composition and ethyl alcohol: water: 1,3-propanediol are in a ratio of 1: 5.5: 15 by weight for a spray composition.

[0007] However, known treatments do not effectively stimulate hair follicles in dogs diagnosed with alopecia X. Despite minoxidil's proven effectiveness in stimulating hair growth, its use in high doses requires careful balancing of therapeutic effect and tolerability, which is especially relevant in veterinary dermatology, where the skin barrier in animals may be more vulnerable than in humans. High concentrations of minoxidil in veterinary cosmetic products intended for the prevention and treatment of alopecia in dogs can cause unwanted local side effects, including itching, dry skin, flaking, and a burning sensation.

[0008] The proposed invention is aimed at eliminating the said drawback and creating a care product - a composition containing active substances that neutralize the side effects of minoxidil at its high concentration and stimulate hair growth in dogs with alopecia X.

[0009] The technical result achieved by implementing the claimed invention consists in ensuring the regeneration of skin tissues and hair follicles in dogs, stimulating the anagen phase - the phase of hair growth, the possibility of using a high concentration of minoxidil and, as a consequence, the prevention of alopecia X through the use of a composition containing active substances with prolonged action and high bioavailability in the deep layers of the epidermis and dermis.

[0010] To achieve the specified technical result, a care product for dogs suffering from alopecia X is proposed, intended for application to the skin during grooming and containing an aqueous phase, minoxidil as an active component, moisturizing components, namely glycerin, propylene glycol, squalane, a composition of water and sodium hyaluronate, conditioning components, namely cetyl stearyl alcohol, behenyltrimonium methosulfate, a composition of amodimethicone bis-cetearyl, ceteareth-7 and ceteareth-25, nutritional components, namely L-arginine, hydrolyzed keratin, hydrolyzed wheat protein, a composition of water, hydrolyzed elastin and deamidated collagen, and auxiliary components, namely vitamin E, phenoxyethanol, potassium sorbate, methosulfate undecylenamidopropyltrimonium and citric acid, mixed in the following order and with the following quantitative ratios in mass percent (wt.%):

[0021] - Undecylenamidopropyltrimonium methosulfate - 0.82-15;

[0022] - Hydrolyzed wheat protein - 0.75-30;

[0023] - Composition of hydrolyzed elastin, deamidated collagen, water - 0.1-31.05;

[0024] - Hydrolyzed keratin - 0.1-71.6;

[0025] - Composition of sodium hyaluronate, water - 0.1-22.13;

[0026] - Phenoxyethanol - 0.79-1;

[0027] - Potassium sorbate - 0.16-1.

[0028] Mixing is carried out in a preheated system consisting of a water-jacketed reactor, where a water thermostat maintains the temperature. The components are introduced at specific temperature intervals, with each subsequent component added only after the previous component has completely dissolved and dispersed.

[0029] Water was used in quantities of 40-90% by weight as a universal solvent, ensuring uniform distribution of active ingredients, their absorption, and achieving an optimal product consistency for comfortable application. The lower limit of 40% by weight ensured sufficient solubility of water-soluble components, while the upper limit of 90% by weight allowed for the production of light toners or sprays. This range covered all forms—from concentrates to emulsions with a high aqueous phase.

[0030] Propylene glycol was used in concentrations of 5-60 wt.% as a penetrating agent and humectant. The range is due to its dual function: at concentrations of 5-10 wt.%, it acted as a safe humectant, and at 20-60 wt.%, it acted as an effective skin penetrant for minoxidil. The upper limit of 60 wt.% was set for safety reasons - higher concentrations could cause irritation in animals with sensitive skin. Minoxidil was administered in an amount of 0.1-20 wt.% as an active hair growth stimulator. The range reflected both cosmetic dosages of 0.1-5 wt.% and therapeutic veterinary dosages of 15-20 wt.%, with a limit on the area of ​​application. The limit of 0.1 wt.% was the minimum effective for follicle activation, and 20 wt.% was the maximum safe level recorded in veterinary practice.

[0031] Minoxidil and propylene glycol were introduced at a temperature of 50-65°C, which ensured their complete dissolution, a decrease in the viscosity of the mixture and activation of penetrating properties without thermal degradation.

[0032] Behenyltrimonium methosulfate was used at 3-30 wt% as a mild cationic surfactant and conditioning agent. Concentrations of 3 wt% and above provided an antistatic effect and facilitated combing, while 30 wt% was used for pronounced conditioning in damaged coats or in mask formulations. Cetyl stearyl alcohol was added at a concentration of 1.65-20 wt%, which contributed to emulsion stabilization, improved texture, and softening of the skin and coat. A minimum concentration of 1.65 wt% was required for system stability, while a maximum concentration of 20 wt% provided a rich structure without weighing the coat down.

[0033] The introduction of behenyltrimonium methosulfate and cetyl stearyl alcohol was carried out at a temperature of 65-68°C, which ensured their melting, compatibility with the oil and aqueous phase, and prevented crystallization.

[0034] Squalane was used in an amount of 0.49-35 wt.%. The lower limit of the concentration range ensured physiological activity - restoration of the lipid barrier, the upper limit of the range - maximum saturation of the skin without the formation of a sticky film, while concentrations above 10 wt.% enhanced the penetrating ability of the composition. Vitamin E was introduced in an amount of 0.33-20 wt.% as an antioxidant and reparative agent. The minimum concentration was required to protect against oxidation of the active substances, especially minoxidil and oils, and the maximum concentration was used for severe dermatological disorders. L-arginine was used in a concentration of 0.52-15 wt.% to stimulate microcirculation and nutrition of hair follicles. The range was selected based on the minimum physiological dose, which is 0.5 wt.% and the upper limit of biological activity of 15 wt.%, at which no sensitization was observed. Glycerol was introduced in an amount of 0.62-15 wt.%. At concentrations of 1-10 wt.% provided standard moisturization, while 10-15% by weight provided a long-lasting occlusive effect. The lower limit of the range is the minimum effective concentration, while the upper limit allows for formulation without the risk of stickiness or pore clogging.

[0035] The introduction of squalane, vitamin E, L-arginine and glycerin was carried out at a temperature of 40-45°C, which maintained the activity of heat-sensitive components and prevented denaturation.

[0036] A composition of amodimethicone bis-cetearyl, ceteareth-7 and ceteareth-25 was introduced in an amount of 0.1-32.13 wt.%. The lower limit of the concentration range provided texture stabilization and a light conditioning effect, and the upper limit - deep smoothing and film protection for heavily damaged wool. Undecylenamidopropyltrimonium methosulfate was used in a concentration of 0.82-15 wt.%. The minimum concentration provided an antimicrobial effect, the maximum concentration was used in the presence of skin problems such as seborrhea, fungal infection. Hydrolyzed wheat protein was introduced in an amount of 0.75-30 wt.%. The lower limit of the concentration range provided strengthening of the wool structure, the upper limit of the range - a masking effect and regeneration of heavily damaged areas. A composition of hydrolyzed elastin, deamidated collagen and water was introduced at a concentration of 0.1-31.05 wt.%.The minimum concentration is necessary to maintain skin turgor, while the maximum concentration provides an effect similar to that of salon reconstructive products. Hydrolyzed keratin was used in an amount of 0.1-71.6 wt.%. Concentrations above 20 wt.% were used as an intensive-action mask, below 20 wt.% - as a nourishing additive, and the upper limit of 71.6 wt.% was determined by the solubility and stabilization limit of the protein. A composition of sodium hyaluronate with water was used in an amount of 0.1-22.13 wt.%. The lower limit of the concentration range provided basic hydration, concentrations above 5 wt.% produced a powerful water-reserving effect, and above 20 wt.% - a filler effect with long-term use. Phenoxyethanol was introduced at a concentration of 0.79-1 wt.% as a safe preservative with a broad spectrum of action. The concentration threshold of 1 wt.% is the upper permissible norm according to international recommendations. Potassium sorbate was used in the amount of 0.16-1 wt.%, this component enhanced the antimicrobial effect of phenoxyethanol. The concentration range was selected to ensure a broad spectrum of activity against fungi and yeast.

[0037] Citric acid was used in quantities of 0.26-10% by weight to stabilize pH in the range of 4.5-5.8. The minimum concentration is necessary for acidification, while the maximum concentration is used in acidic buffers for high water mineralization.

[0038] The introduction of a composition of amodimethicone bis-cetearyl, ceteareth-7, ceteareth-25, undecylenamidopropyltrimonium methosulfate, hydrolyzed wheat protein, a composition of water, hydrolyzed elastin and deamidated collagen, hydrolyzed keratin, a composition of water and sodium hyaluronate, phenoxyethanol, potassium sorbate and citric acid was carried out at a temperature of 25-38°C, which ensured their compatibility, stability and preservation of activity.

[0039] In this particular case, a perfume composition was added as a flavoring agent at a concentration of 0.16-5% by weight at the final stage of product production. The perfume composition included lavender essential oil and a fruity fragrance. The minimum concentration provided a scent to mask the odor of active ingredients and create a pleasant aroma. The maximum concentration provided an additional calming and antiseptic effect. Exceeding 5% by weight could cause allergies. This range corresponded to the permissible level in veterinary cosmetics. Furthermore, in this particular case of product sales, the dyes CI 14720 and CI 42090 were also added at the final stage of production in the minimum necessary quantities, sufficient for visual identification of the product without affecting the activity or stability of the composition.

[0040] Stirring during the introduction of each component was carried out at a speed of 80-300 rpm, which ensured homogeneity, emulsion and dispersion stability, minimal loss of active ingredients, and high bioavailability. Speeds below 80 rpm did not ensure effective dispersion, while speeds above 300 rpm resulted in foaming and destruction of heat-sensitive components.

[0041] The product was balanced for pH, viscosity, oxidative stability, and component compatibility. The selected composition, concentrations of active, moisturizing, conditioning, nourishing, and auxiliary ingredients, as well as the mixing method, were chosen to avoid local side effects associated with high concentrations of minoxidil. The formula was developed taking into account the physiological characteristics of canine skin, including breeds predisposed to alopecia X, and has demonstrated effectiveness with 6-24 week courses of application.

[0042] A particular case of the claimed care product for dogs suffering from alopecia X is disclosed in detail based on the composition of the composition, which represents an example of the embodiment of the invention.

[0043] To obtain 1000g of the treatment product, 460g of purified water was placed in a reactor in a water bath and heated to a temperature of 50-65°C. Then 55g of propylene glycol and 50g of minoxidil were added. The mixture was stirred and heated to 65-68°C. After reaching the specified temperature, 54.7g of behenyltrimonium methosulfate and 36.4g of cetyl stearyl alcohol were added. The mixture was stirred until smooth and cooled to 40-45°C. In the next step, 63.8g of squalane and 18.4g of vitamin E were added, after which stirring was continued. Then 12g of L-arginine and 22g of glycerin were added. The resulting mass was cooled to 25-38°C. Next, sequentially, with intermediate stirring until each component was completely dissolved, the following were added:

[0049] 29.25g composition of sodium hyaluronate and water.

[0050] At the final stage, preservatives were added:

[0051] 9g phenoxyethanol,

[0052] 1.8 g potassium sorbate.

[0053] Then they added:

[0054] 6.3g lavender essential oil,

[0055] 7g fruit flavor.

[0056] After this, 15 g of citric acid was added to adjust the pH and 0.05 g of dye to give the desired shade.

[0057] Thus, the process of preparing the composition was completed in compliance with all temperature and technological parameters.

[0058] The present invention is further illustrated by the following examples, which should not be construed as limiting. The composition in each example was prepared by mixing the specified components in a stirred reactor, adhering to the process parameters of introducing the components at a strictly specified temperature after dissolving the previous one.

[0059] As part of a study to evaluate the effectiveness of the developed treatment for dogs suffering from alopecia X, experiments were conducted using formulations with varying minoxidil concentrations, all other conditions being equal. The experiments lasted six months and were conducted on individuals similar in age, breed, and severity of alopecia. In the first example (Figs. 1-4), a composition containing 0.1 wt.% minoxidil was used; the detailed composition is given in Table 1. In the second example (Figs. 5-8), with a higher minoxidil concentration of 19 wt.%, the detailed composition is given in Table 2. The remaining components included in the composition, as well as the regimen and method of application of the product, remained identical: local rubbing onto bald areas of the skin twice a day, without rinsing, for the entire observation period. During the first 4-6 weeks of use, in both cases, there was a reduction in itching, decreased flaking, improvement in the general condition of the skin, and the beginning of the appearance of vellus hair.From 8-10 weeks, dogs showed more active hair growth: growth was uniform, dense, with pronounced pigmentation and shine. By the end of the six-month period, hair regeneration in dogs receiving the higher minoxidil concentration reached over 90% of normal, while in dogs receiving the 0.1% minoxidil concentration, hair growth reached approximately 60-65%. In both cases, no side effects such as itching, flaking, dryness, or burning sensation were observed.

[0060] Thus, the selected composition and quantitative ratio of the components, as well as the mixing method in a water-jacketed reactor with gradual heating and cooling, made it possible to obtain a composition with a higher concentration of minoxidil, which does not cause an unpleasant reaction on the skin of animals and significantly accelerates the process of hair regeneration. However, further increases in the minoxidil content above 40% by weight were considered undesirable due to the potential increase in the risk of skin sensitization and systemic absorption of the active substance. The obtained data indicate the advisability of using minoxidil in the range of 15 to 20% by weight, provided that it is formulated in a complex with dermatologically active excipients that ensure both the transport of the active component to the hair follicles and the protection of the skin barrier.

Claims

1. A grooming product for dogs suffering from alopecia X, intended for application to the skin during grooming, containing water, minoxidil as an active ingredient, moisturizing ingredients, namely glycerin, propylene glycol, squalane, a composition of water and sodium hyaluronate, conditioning ingredients, namely cetyl stearyl alcohol, behenyltrimonium methosulfate, a composition of amodimethicone bis-cetearyl, ceteareth-7 and ceteareth-25, nutritional ingredients, namely L-arginine, hydrolyzed keratin, hydrolyzed wheat protein, a composition of water, hydrolyzed elastin and deamidated collagen, and auxiliary ingredients, namely vitamin E, phenoxyethanol, potassium sorbate, undecylenamidopropyltrimonium methosulfate and citric acid, characterized in that all components sequentially mixed in a water-jacketed reactor at a speed of 80-300 rpm under the following conditions: Propylene glycol in an amount of 5 to 50 wt.% is added to a system preheated to a temperature of 50-65°C, containing water in an amount of 40 to 85 wt.%, mixed, and minoxidil is introduced in an amount of 0.1 to 20 wt.%, the mixture of the specified components is heated to a temperature of 65-68°C while stirring, after which behenyltrimonium methosulfate is added in an amount of 3 to 30 wt.% and cetyl stearyl alcohol in an amount of 1.65 to 20 wt.%, mixed until homogeneous, then the mixture is cooled to a temperature of 40-45°C, squalane is added in an amount of 0.49 to 35 wt.% and vitamin E in an amount of 0.33 to 20 wt.%, mixed, then L-arginine is introduced in an amount of 0.52 to 15 wt.% and glycerin in an amount of 0.62 to 15 wt.%, after which the mixture is cooled to a temperature of 25-38°C and sequentially, waiting for each component to dissolve, the following are added: a composition of amodimethicone bis-cetearyl, ceteareth-7 and ceteareth-25 in a total amount of 0.1 to 32.13 wt. %, undecylenamidopropyltrimonium methosulfate in an amount of 0.82 to 15 wt. %, hydrolyzed wheat protein in an amount from 0.75 to 30 wt.%, a composition of water, hydrolyzed elastin and deamidated collagen in an amount of 0.1 to 31.05 wt. %, hydrolyzed keratin in an amount from 0.1 to 45 wt. %, a composition of water and sodium hyaluronate in an amount of 0.1 to 22.13 wt. %, then the preservative phenoxyethanol is introduced in an amount of 0.79 to 1 wt.%, the preservative potassium sorbate in an amount of 0.16 to 1 wt.%, the pH regulator - citric acid in an amount of 0.26 to 10 wt.%.

2. The product according to item 1, characterized in that the pH of the finished product is from 4.5 to 5.
