Abstract

------22 / 05 / 2569------(OCR)OCR09WP The process of producing xelolipid biosurfactant from yeast using xylose and palm oil as carbon sources begins with culturing yeast cells and transferring the prepared yeast cell starter into 250 ml shaker bottles containing liquid culture medium, xylose, palm oil, ammonium sulfate, yeast extract, urea, and sodium chloride. The pH is adjusted, the bottles are shaken, and the produced xelolipids are measured and extracted. This is then applied to a 5-liter bioreactor to increase the yield of biosurfactant.

Claims

------22 / 05 / 2569------(OCR)OCR09WP1. The process of producing xilolipid biosurfactant from yeast using xylose and palm oil as carbon sources consists of the following steps: a. Cultivating yeast cells (Meyerozymas sp.) in liquid YPD medium, 50 ml (for step b) and 400 ml (for step c) at a temperature of 28-32 degrees Celsius, shaking rate of 180-220 rpm, for 18-24 hours to prepare starter cultures. b. Transferring the prepared yeast cell starter cultures... From step (a), pour 50 ml into a 250 ml shaker bottle containing 50 ml of liquid culture medium consisting of 80-100 g xylose, 80-100 g palm oil, 15-25 g ammonium sulfate, 15-25 g yeast extract, 15-25 g urea, and 30-40 g sodium chloride in 1 liter of water. Adjust the pH to 6.

0. Start cells at OD600 = 1.

0. Shake at a rate of 180-220 rpm. Incubate at 28-32 °C for 10 days and then measure the amount of xyl lipid extract produced. C. Take the prepared material from step b.The process involves applying it to a 5-liter bioreactor. First, yeast cells are cultured in YPD liquid medium at a temperature of 28-32 degrees Celsius, shaking at a rate of 180-220 rpm for 18-24 hours to prepare a starter culture. 400 ml of the prepared yeast starter culture (from step a) is then transferred to the 5-liter bioreactor containing 4 liters of liquid medium consisting of 80-100 g of xylose, 80-100 g of palm oil, 15-25 g of ammonium sulfate, and 15-25 g of yeast extract.

1. A specific liquid culture medium formula for xylophilic compound production consisting of 80-100 g of xylose, 80-100 g of palm oil, 15-25 g of ammonium sulfate, 15-25 g of yeast extract, 15-25 g of urea, 30-40 g of sodium chloride, and 1 liter of water. Adjust the pH to 6.

0. Start with cells at an initial OD of 600 g, then incubate at 30°C in a 5-liter fermenter at a stirring speed of 180-220 rpm. Aeration rate should be 0.5-1.0 vvm for 10 days.
