Abstract

------22 / 05 / 2569------(OCR)OCR09WP This invention relates to the process of producing glycolipid biosurfactants using yeast, molasses, and palm oil, with the potential for further industrial applications. The production process involves the following steps: culturing yeast cells under optimal conditions, transferring the prepared starter culture into shaker bottles containing liquid culture medium composed of molasses, palm oil, ammonium sulfate, yeast extract, urea, and sodium chloride, adjusting the pH, and then extracting the produced glycolipids. These extracts are then applied to a 5-liter bioreactor, and adjusting the conditions to achieve an increased yield of glycolipid biosurfactants. The aim of this invention is to improve the high-potential yeast-based production process of glycolipid biosurfactants using molasses and palm oil to reduce the cost of glycolipid production through bioprocesses.

Claims

------22 / 05 / 2569------(OCR)OCR09WP1. The process of producing glycolipid-based biosurfactants by yeast using molasses and palm oil consists of the following steps: a. Cultivating yeast cells (Meyerozymas sp.) in liquid YPD medium, 50 ml (for step b) and 400 ml (for step c) at a temperature of 28-32 degrees Celsius, shaking rate of 180-220 rpm, for 18-24 hours to prepare starter cultures. b. Transferring the prepared yeast cell starter cultures. From step (a), pour 50 ml into a 250 ml shaker bottle containing 50 ml of liquid culture medium consisting of 80-100 g molasses, 80-100 g palm oil, 15-25 g ammonium sulfate, 15-25 g yeast extract, 15-25 g urea, and 30-40 g sodium chloride in 1 liter of water. Adjust the pH to 6.

0. Start cells at OD600 = 1.

0. Shake at a rate of 180-220 rpm. Incubate at 28-32 °C for 10 days and then measure the produced glycolipid extract. C. Take the prepared material from step b.It was applied to a 5-liter bioreactor by culturing yeast cells in YPD liquid medium at a temperature of 28-32 degrees Celsius, shaking at a rate of 180-220 rpm for 18-24 hours to prepare starter cultures. 400 ml of the prepared yeast cell starter culture was then transferred to the 5-liter bioreactor containing 4 liters of liquid medium consisting of 80-100 g of molasses, 80-100 g of palm oil, 15-25 g of ammonium sulfate, 15-25 g of yeast extract, 15-25 g of urea, and sodium chloride.

1. Add 30-40 grams of sodium chloride to 1 liter of water, adjust the pH to 6.0, start the cells with an OD of 600 equal to 1.0, incubate at 30 degrees Celsius for 10 days in a 5-liter fermenter at a stirring speed of 180-220 rpm, and an aeration rate of 0.5-1.0 vvm. Then measure the produced glycolipid extract.

2. A specific liquid culture medium formula for glycolipid production consists of 80-100 grams of molasses, 80-100 grams of palm oil, 15-25 grams of ammonium sulfate, 15-25 grams of yeast extract, 15-25 grams of urea, 30-40 grams of sodium chloride, and 1 liter of water.
