OCR10KL (28 / 04 / 2569) 1. The process of preparing probiotic fermented tea that can lower cholesterol levels in test tubes involves two steps: 1.

1. Preparing the tea for fermentation: a. Boil 3-5 liters of distilled water, then add 12-20 grams of tea powder. Boil until the tea begins to darken, taking 1.00-1.30 hours, and then let it rest for about 10-20 minutes. b. Add 165-280 grams of glucose to the boiled tea in step a and stir until completely dissolved. Let it cool. c. Pour the tea from step a...

1. B. Pour the mixture into 250-500 ml Baffled flasks, 100 ml per flask.

2. Sterilize the tea from step c using an autoclave at 105-110 degrees Celsius for 10-20 minutes.

3. Let the sterilized tea stand at room temperature until it cools down to room temperature, then use it in the next step of fermented tea production. 1.

2. Preparation of Probiotic Fermented Teaa. Prepare the Enterococcus faecalis TISTR 2932 culture by streaking it onto deMann-Rogosa-Sharpe (MRS) medium. Incubate at 35-37°C in a candle jar for 48-72 hours. b. Select individual colonies from step a and incubate them in 5-10 ml of deMann-Rogosa-Sharpe (MRS) medium in a screw-top tube. The bacterial suspension was incubated in 25 x 250 mm culture media at 35-37°C for 48-72 hours without shaking. The suspension was then centrifuged at 6,000-8,000 rpm for 10-15 minutes, and the liquid portion was discarded. The bacterial cells in step c were washed twice with a 0.85% w / v sodium chloride (NaCl) solution. The turbidity of the bacterial cells in step d was calibrated to match McFarland standard No. 0.5 using a 0.85% sodium chloride (NaCl) solution.85% by weight / volume (w / v). e. Pour 20% by volume (v / v) of the bacterial suspension from step e into a 500 ml Baffled flask containing 100-200 ml of sterile tea and mix thoroughly. d. Incubate the mixture from step f at 35-37°C for 14-20 days without shaking. e. Transfer the mixture from step b into a 1.5 ml plastic centrifuge tube and centrifuge to separate the cells from the fermented tea at 10,000 rpm for 30 minutes at 4-10°C.
