The present invention discloses a trabectedin compound, its conjugate, and methods for their preparation and use. In particular, it discloses ligand-drug conjugates comprising a structure represented by Formula I. The ligand-drug conjugates disclosed herein exhibit excellent tumor cell proliferation inhibitory activity and / or favorable in vivo antitumor efficacy.;
