AI Technical Summary

Technical Problem

Current skin rejuvenation methods, including laser and chemical resurfacing, do not effectively address textural and pigmentation issues, and regenerative medicine-based products like MSC-derived exosomes have unclear molecular mechanisms for skin regeneration.

Method used

Engineered exosomes with KGF, EGF, FGF-2, and PDGF-BB peptides anchored to their membrane are developed to enhance fibroblast proliferation, upregulate collagen and fibronectin, downregulate matrix metalloproteinases, and reduce melanin synthesis, formulated for topical application.

Benefits of technology

The engineered exosomes enhance skin rejuvenation by improving skin texture, reducing wrinkles and pigmentation, and promoting a youthful appearance through enhanced fibroblast activity and gene regulation.

Abstract

This invention discloses an engineered exosome comprising (a) a KGF peptide fused to a first anchoring peptide, (b) an EGF peptide fused to a second anchoring peptide, and (c) an FGF-2 peptide fused to a third anchoring peptide, wherein (a), (b), and (c) are anchored to the membrane of the exosome via the first, second, and third anchoring peptides, respectively, and wherein the KGF peptide, the EGF peptide, and the FGF-2 peptide are exposed on the outer surface of the exosome membrane. The composition, nucleic acid construct, and use of the exosome for skin rejuvenation are also disclosed.
