Abstract

This invention relates to an extracellular vesicle derived from the Indian gooseberry plant, its preparation method, and its uses. The preparation method includes the following steps: (1) Sample processing: Fresh Indian gooseberry fruit is taken, washed, and the pulp and seeds are removed. The fruit is homogenized using a juicer to obtain juice, which is then centrifuged to remove impurities; (2) Tangential flow filtration: The supernatant is taken and filtered sequentially through a filter membrane. The supernatant is then concentrated using an ultra-concentrated filtration device with an MWCO ultrafiltration column; (3) Final concentration and purification: The sample is concentrated again using an MWCO ultrafiltration column, or the extracellular vesicles derived from the Indian gooseberry plant are separated using particle size chromatography. The extracellular vesicles derived from the Indian gooseberry plant prepared by this invention have antioxidant, anti-inflammatory, and photoprotective effects.

Claims

1. A method for preparing plant-derived extracellular vesicles from Indian gooseberry (Phyllanthus emblica L.), comprising the following steps: Step S1: washing Indian gooseberry fruits to remove pomace and seeds, homogenizing and centrifuging, and collecting the supernatant; and Step S2: filtering the supernatant with a filter membrane to remove particulate impurities, and then concentrating the supernatant using an ultra-concentrating filtration device with an ultrafiltration column to obtain the plant-derived extracellular vesicles from Indian gooseberry, wherein the centrifugation process involves first centrifuging the homogenized Indian gooseberry fruits at 3000 rpm for 30 minutes, and then increasing the speed to 9000 rpm for 4 hours to obtain the supernatant; and the ultrafiltration column is a molecular weight cutoff (MWCO) ultrafiltration column with a molecular weight cutoff value of 750 kDa.

2. The preparation method as described in claim 1, wherein in step S1, the centrifugation is performed at 4°C.

3. The preparation method as described in claim 1, wherein in step S2, the supernatant is filtered sequentially through 0.45µm and 0.22µm filter membranes to remove particulate impurities.

4. An extracellular vesicle of Indian gooseberry (Phyllanthus emblica L.) plant origin, prepared by the method described in any one of claims 1 to 3.

5. The Indian gooseberry plant-derived extracellular vesicles as described in claim 4, comprising the active ingredients of β-glucosamine, gallic acid, rutin, and ellagic acid.
