Microsatellite genetic marker set of cricetulus griseus and screening method and application thereof

Abstract

The invention discloses a microsatellite genetic marker set of Cricetulus griseus (or Cricetulus barabensis griseus), as well as its screening method and application, belonging to the technical field of molecular genetic markers. The invention extracts genomic DNA of Cricetulus griseus, fragments the DNA by acoustic focusing, and recovers the DNA fragments; constructs a microsatellite-enriched genomic library of Cricetulus griseus, picks positive clone plasmids from it for sequencing, screens out 10 microsatellite markers whose nucleotide sequences are shown in SEQ ID NO.24-SEQ ID NO.33, and designs 10 pairs of primers for genetic detection. In specific application, the 10 pairs of primers are used to perform PCR amplification on genomic DNA of different populations or different individuals within a population of Cricetulus griseus, the amplified products are subjected to electrophoresis detection, and the genotype of each individual is determined according to the detection data and maps; the method of the invention has good stability, high repeatability, strong comparability and simple operation.
