Claims

This description relates to anti-PTK7 / B7H3 antibodies and antibody-drug conjugates derived therefrom, wherein the antibodies bind specifically to at least two different antigens, PTK7 and B7H3. Pharmaceutical compositions comprising such antibodies and antibody-drug conjugates, methods of use, and methods of preparation of such antibodies and antibody-drug conjugates are also described.

1. An anti-PTK7 / B7H3 antibody or antigen-binding fragment thereof, comprising: a first antigen-binding domain that binds specifically to PTK7; and a second antigen-binding domain that binds specifically to B7H3.

30. A nucleic acid comprising a polynucleotide encoding the anti-PTK7 / B7H3 antibody or antigen-binding fragment thereof of any of claims 1-28.

31. A vector comprising the nucleic acid of claim 30.

32. A cell comprising the vector according to claim 31.

34. A cell comprising the nucleic acid of claim 30.

35. A method for producing an anti-PTK7 / B7H3 antibody or an antigen-binding fragment thereof, the method comprising (a) growing the cell of any of claims 32-34 under conditions sufficient for the cell to produce the anti-PTK7 / B7H3 antibody or the antigen-binding fragment thereof; and (b) collecting the anti-PTK7 / B7H3 antibody or the antigen-binding fragment thereof produced by the cell.

36. A composition comprising: a first vector encoding VH1, a second vector encoding VH2, and a third vector encoding VL1 / VL2, of the anti-PTK7 / B7H3 antibody or antigen-binding fragment thereof of any of claims 1-28.

37. A cell comprising the composition of claim 36.

38. A cell according to claim 37, which is a CHO-S cell.

39. A method for producing an anti-PTK7 / B7H3 antibody or an antigen-binding fragment thereof, the method comprising (a) growing the cell of claim 37 or 38 under conditions sufficient for the cell to produce the anti-PTK7 / B7H3 antibody or the antigen-binding fragment thereof; and (b) collecting the anti-PTK7 / B7H3 antibody or the antigen-binding fragment thereof produced by the cell.

40. An anti-PTK7 / B7H3 antibody-drug conjugate (ADC) comprising a therapeutic agent covalently linked to the anti-PTK7 / B7H3 antibody or antigen-binding fragment thereof of any of claims 1-28.

68. A pharmaceutical composition comprising a pharmaceutically acceptable carrier and (a) the anti-PTK7 / B7H3 antibody or antigen-binding fragment thereof of any of claims 1-28, and / or (b) the anti-PTK7 / B7H3 antibody-drug conjugate of any of claims 40-48.
