Description

SKIN MOISTURIZING COMPOSITION INCLUDING BOTANIC ACTIVE INGREDIENTSFIELD

[0001] The present disclosure relates to compositions providing moisturization and soothing efficacy without leaving the skin upon which the compositions are applied overly oily or greasy. More specifically, the present disclosure relates to compositions comprising a combination of oats, portulaca extract, and meadow foam oil suitable for providing improved moisturization and soothing efficacy.BACKGROUND

[0002] Current emollients, including mineral oil and petrolatum, are derived from petroleum. These emollients are hydrophobic and have a greasy feel. Mineral oil and petrolatum are typically applied to the skin in the form of creams or lotions in order to provide moisturization to skin. Creams or lotions that contain mineral oil or petrolatum also typically contain surfactants to enable the formation of stable oil in water emulsions. However, these emollients also tend to leave the skin feeling more greasy than natural emollients, such as oils derived from plants, while natural emollients fail to moisturize and sooth as well as petroleum-derived emollients. Thus, there is an ongoing need to identify ingredients and formulations which provide moisturization and / or soothing benefits without being overly greasy.SUMMARY

[0003] This Summary is provided to introduce a selection of concepts in a simplified form that are further described below in the Detailed Description. This Summary is not intended to identify key features or essential features of the claimed subject matter, nor is it intended to be used as an aid in determining the scope of the claimed subject matter.

[0007] The various implementations and examples will be described in detail with reference to the accompanying drawings. Wherever possible, the same reference numbers will be used throughout the drawings to refer to the same or like parts. References made throughout this disclosure relating to specific examples and implementations are provided solely for illustrative purposes but, unless indicated to the contrary, are not meant to limit all examples.

[0008] Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which the invention belongs. Also, all publications, patent applications, patents, and other references mentioned herein are incorporated by reference.

[0009] As used herein, “atopic dermatitis” (AD) , also known as eczema, is a chronic relapsing pruritic inflammation of the skin that can compromise quality of life. AD affects 10-20%of children and 1-3%of adults worldwide with increasing prevalence in highly industrialized countries. AD is characterized by an impaired epidermal barrier, dysregulation of innate and adaptive immunity, and a high susceptibility to bacterial colonization and infection. Hence, lubricants (ointments &creams) and topical or oral corticosteroids, which act on the immune system by blocking the production of substances that trigger allergic and inflammatory actions, remain the first-line treatments. Similarly, antihistamine can also be administrated.

[0010] As described herein, current emollient options leave a desire for skin care products with excellent moisturization efficacy for skin moisturization, with natural ingredients and without  leaving the skin greasy and oily. Due to worse oxidation stability and higher cost of natural oil, lower dosages of natural oils are conventionally used to provide limited moisture efficacy. Other considerations include consumer preferences toward natural or naturally derived components. Accordingly, the present disclosure provides a skin-moisturizing composition, and method of treating skin comprising applying the skin-moisturizing composition, that includes a combination of oats, portulaca extract, and meadow foam oil are effective in increasing natural moisture factor and skin barrier-associate proteins, provide moisture efficacy verified by in invitro testing, and provides soothing efficacy by inhibiting the secretion of inflammatory factors.

[0011] Unless otherwise indicated, percentages used to express amounts of ingredients are percentage by weight (referred to as “weight %, ” “wt%” , “%by weight” or “% (W / W) ” ) . Similarly, weight ratios used to express relative proportions of ingredients are also determined using percentage by weight (i.e., weight ratios are calculated by dividing the percentage by weight of one ingredient by another) . Unless stated otherwise, all ranges are inclusive of the endpoints, e.g., “from 4 to 9” includes the endpoints 4 and 9.

[0012] As used herein, “cosmetically acceptable” means that the ingredients the term describes are suitable for use in contact with tissues (e.g., the skin or hair) without undue toxicity, incompatibility, instability, irritation, allergic response, or the like.

[0013] As used herein, the term “safe and effective amount” means an amount sufficient to induce the desired effect, but low enough to avoid serious side effects. The safe and effective amount of the compound, extract, or composition will vary with, e.g., the age, health and environmental exposure of the end user, the duration and nature of the treatment, the specific extract, ingredient, or composition employed, the particular carrier utilized, and like factors.

[0014] As used herein, the term “about” refers to within 5%weight, within 4%weight, within 3%weight, within 2.5%weight, within 2%weight, or within 1%weight of a disclosed value.

[0015] One aspect of the present disclosure pertains to a combination of botanic active ingredients having excellent moisturization efficacy. In vitro testing on a three-dimensional (3D) epidermal skin model was utilized to evaluate the moisture efficacy of the active ingredients’ combination based on a blank testing base comprising at least one acrylates polymer and preservative. Additionally, another aspect of the present disclosure pertains to a skin care product composition comprising the combination of botanic active ingredients that is suitable for skin hygiene, protection, rehabilitation and regeneration, and maintenance of skin’s wellbeing.

[0033] Table 1

[0034] Table 2

[0035] As shown in Table 2, the placebo includes only water, acrylates provided as PemulenTM EZ-4U Polymer, sodium hydroxide, and phenoxyethanol. In contrast, the compositions of the inventive examples include includes Avena Sativa (Oat) Kernel Flour provided as Colloidal Oat, Avena Sativa Kernel Extract with potassium sorbate and glycerin as a raw material stabilizer, Avena Sativa (Oat) Kernel Oil, Portulaca Oleracea extract with butylene glycol as a solvent, and Limnanthes Alba (Meadowfoam) Seed Oil with tocopherol as anti-oxidant.

[0036] The test composition was prepared by first adding water toa vessel. The pH adjuster (sodium hydroxide) was dissolved in the water as a first premix. The Avena Sativa (Oat) Kernel Flour (Colloidal Oat) was dissolved in the water as a second premix. The suspension agent was dispersed in the water by an agitator (WERKE, IKA) with 200 rpm / min mixing for thirty minutes until fully dispersed. The combination of the pH adjuster, Avena Sativa (Oat) Kernel Flour  (Colloidal Oat Flour and suspension agent were then homogenized by a homogenizer (L4R, SILVERSON) with 3000 rpm / min for three minutes. The pH adjuster was adjusted to a weak acidic, preferably in the range of 5.2-6.0, with the first premix of sodium hydroxide mixing with 200rpm / min. The second premix, Avena Sativa Kernel Extract, Avena Sativa (Oat) Kernel Oil, Portulaca Oleracea, and Limnanthes Alba (Meadowfoam) Seed Oil were added and then homogenized for three minutes with 3000 rpm / min. Phenoxyethanol was then added and mixed for between five and ten minutes at 200 rpm / min. Where needed, compensating water was added to 100 W / W%, as there may be some water loss during the process, while mixing until the formulation is unform. The formulation is then discharged in bulk, for example into a storage container.

[0037] Testing

[0038] Table 3 illustrates efficacy results of different combinations of ingredients in various examples, including a Placebo, Comparative Examples CE1-4 and Inventive Examples E1-E2. More particularly, Table 3 illustrates different combinations of ingredients and in-vitro testing results associated with both moisturization and soothing properties.

[0039] Table 3

[0040] Throughout the testing of different combinations, a P-value < 0.05 was considered as significant difference, while a P-value < 0.01 was considered as extremely significant difference. Table 3 illustrates that inventive examples E1 and E2 include each of Avena Sativa (Oat) Kernel Flour (Colloidal Oat, from LaCrosse Milling Co) , Avena Sativa Kernel Extract ( from Symrise) , Avena Sativa (Oat) Kernel Oil (Avena Lipid, from New  Holland Extraction) , Limnanthes Alba (Meadowfoam) Seed Oil (Cropure Meadowfoam-LQ- (JP) , from Croda Japan KK) , and Portulaca Oleracea (Portulaca Extract, from Hyundai Bioland Co) , in varying weight %. In comparison, the placebo control contains none of these ingredients, comparative example CE1 contains only Meadowform and Portulaca Extract, CE2 contains only Avena Sativa Kernel Extract ( from Symrise) and Portulaca Extract, CE3 contains only Meadowform and Portulaca Extract in different weight %than CE1, and CE4 contains only Avena Sativa (Oat) Kernel Flour (Colloidal Oat, from LaCrosse Milling Co) , Avena Sativa Kernel Extract ( from Symrise) , and Avena Sativa (Oat) Kernel Oil (Avena Lipid, from New Holland Extraction) without the Limnanthes Alba (Meadowfoam) Seed Oil (Cropure Meadowfoam-LQ- (JP) , from Croda Japan KK) and Portulaca Oleracea (Portulaca Extract, from Hyundai Bioland Co) .

[0041] Moisturization In Vitro Test method:

[0042] The in vitro test was conducted in agency on a 3D skin model. The in-vitro test was carried out according to the barrier-associated proteins, such as Filaggrin and Loricrin, and natural moisturizing factor (Pyrrolidone carboxylic acid, Urocanic acid) content based on the model. The 3D epidermal skin model was used as the research object. The hydration efficacy of the samples to be tested was evaluated by detecting the changes in barrier-associated proteins (Filaggrin, Loricrin) and natural moisturizing factor (Pyrrolidone carboxylic acid, Urocanic acid) content based on model after the sample was applied.

[0043] Moisturization Test material:

[0044] In a stable combination of botanic active ingredients, including the oil solubility ingredients and water solubility ingredients for the efficacy test, a simple base contains at least one acrylates polymer. Based on this, one preservative was built. The base stabilizes the combination during the test period and is used as the placebo to verify the test results of botanic active ingredients combination. Botanic active ingredients were added into the base with proper dosage.

[0045] The primary equipment utilized during the in vitro testing included a carbon dioxide incubator, a fluorescence microscope, a corneometer CM 825 skin moisture tester, and high-performance liquid chromatography (HPLC) .

[0046] Moisturization In-Vitro Test Results:

[0047] Regarding natural moisture factor content evaluation, natural moisturizing factors such as pyrrolidone carboxylic acid (PCA) and urocanic acid (UCA) may effectively absorb and lock water in the skin. To evaluate whether the skin care product compositions CE1-CE4 and E1-E2 had enhanced the skin hydration function, in vitro testing determined whether the content of PCA and UCA was detected.

[0048] Relative to the placebo and comparative examples CE1-CE4, the in-vitro test results demonstrate the inventive examples E1-E2 of the skin care product compositions of the present disclosure provide dramatically increased moisture efficacy by increasing the PCA and UCA. For example, the increase in PCA of the comparative examples CE1-CE4 demonstrated a range of 114.36 –195.50, while E1 demonstrated an increase of 208.30 and E2 demonstrated an increase of 233.04. As another example, the increase in UCA of the comparative examples CE1-CE4 demonstrated a range of 78.85 –198.21, while E1 demonstrated an increase of 166.67 and E2 demonstrated an increase of 230.47. Thus, the inventive examples E1 and E2 demonstrate significantly improved natural moisturization over the comparative examples of CE1 –CE3 that do not include each of the each of Avena Sativa (Oat) Kernel Flour (Colloidal Oat, from LaCrosse Milling Co) , Avena Sativa Kernel Extract ( from Symrise) , and Avena Sativa (Oat) Kernel Oil (Avena Lipid, from New Holland Extraction) and over the comparative example of CE4 that does not include Limnanthes Alba (Meadowfoam) Seed Oil (Cropure Meadowfoam-LQ- (JP) , from Croda Japan KK) and Portulaca Oleracea (Portulaca Extract, from Hyundai Bioland Co) .

[0049] Regarding skin barrier associated protein content evaluation, a barrier-associated protein Filaggrin (FLG) was a structural component of the skin barrier that is also hydrolyzed by Caspase-13 to form natural moisturizing factors and plays a role in moisturizing. Loricin (LOR) also plays a key role in forming a skin barrier. In some instances, a decrease of skin barrier function-associated proteins may lead to weak skin barrier function, which in turn leads to increased transdermal water loss. Detecting the content of FLG and LOR is used to evaluate whether the compositions had enhanced the skin barrier function.

[0050] As compared to the placebo, the in vitro test results demonstrate the compositions of the skin care product composition of the present disclosure provide dramatically increased moisture efficacy by increasing the FLG and LOR. For example, the increase in FLG of the comparative examples CE1 –CE4 demonstrated a range of no significant increase –34.00, while E1 demonstrated an increase of 36.00 and E2 demonstrated an increase of 165.00. As another examples, the increase in LOR of the comparative examples CE1 –CE4 demonstrated a range of 33.00 –141.00, while E1 demonstrated an increase of 155.00 and E2 demonstrated an increase of 167.00. Thus, the inventive examples E1 and E2 demonstrate significantly improved skin barrier function over the comparative examples of CE1 –CE3 that do not include each of the each of Avena Sativa (Oat) Kernel Flour (Colloidal Oat, from LaCrosse Milling Co) , Avena Sativa Kernel Extract ( from Symrise) , and Avena Sativa (Oat) Kernel Oil (Avena Lipid, from New Holland Extraction) and over the comparative example of CE4 that fails to include  Limnanthes Alba (Meadowfoam) Seed Oil (Cropure Meadowfoam-LQ- (JP) , from Croda Japan KK) and Portulaca Oleracea (Portulaca Extract, from Hyundai Bioland Co) .

[0051] Moisturization Clinical Testing

[0052] Skin care product compositions, in the form of lotions and creams, containing the inventive examples E1 and E2 were further subjected to clinical testing to determine moisture efficacy by evaluating the skin hydration and trans-epidermal water loss (TEWL) of skin on thirty adults. Table 4 below illustrates the results of the clinical testing.

[0053] Table 4

[0054] As shown in Table 4, when compared to the baseline value (T0) , the increasing rates of skin hydration and decreasing rates were statistically significant, as were the measured values at follow-up time in intra-group and inter-group (P-value <0.001) . For example, at T1, one hour after application, skin hydration significantly increased and skin TEWL significantly decreased for each of the skin care product containing E1 and the skin care product containing E2. At each of T24 hours, twenty-four hours after application, and T48, forty-eight hours after application, skin hydration remained increased and skin TEWL remained decreased relative to T0 for each of the skin care product containing E1 and the skin care product containing E2, indicating both short term, i.e., after one hour, efficacy and longer term, i.e., twenty-four and forty-eight hour, efficacy.

[0055] Moisturization Consumer Use Testing:

[0056] Skin care product compositions containing the inventive examples E1 and E2 were further subjected to consumer use testing to determine moisture efficacy by evaluating the skin hydration and TEWL of skin on adults. Questionnaires were used to investigate the consumer’s agreement to evaluate the sensory aspects of skin care products containing E1 and E2. Table 5 below illustrates the results of the consumer use testing.

[0057] Table 5

[0058] As shown in Table 5, 108 adults responded regarding the skin care product containing E1 and 109 adults responded regarding the skin care product containing E2. Of the 108 adults that responded regarding the skin care product containing E1, 92%agreed the skin care product was non-greasy and 94%agreed the skin care product was non-sticky. Of the 109 adults that responded regarding the skin care product containing E2, 87%agreed the skin care product was non-greasy and 82%agreed the skin care product was non-sticky.

[0059] Accordingly, based on the in-vitro testing, clinical testing, and consumer use testing, skin care products containing the inventive examples E1 and E2 are demonstrated to provide significant skin hydration efficacy as well as significant moisturization efficacy, without a greasy or sticky feel to the skin.

[0060] Soothing In Vitro Test Method:

[0061] The in vitro test was conducted in agency on a 3D skin model. The in-vitro test was carried out according to the Pro-Inflammatory Factor (IL-1α) and Inflammatory Mediator (PGE2) Content based on Model. In this test, sodium lauryl sulfate (SLS) was used to stimulate the 3D epidermal skin model to construct the impaired skin model in vitro. The soothing efficacy of the samples to be tested was evaluated by detecting the changes in pro-inflammatory factor (IL-1α) and inflammatory mediator (PGE2) content of skin model after the sample was applied.

[0062] Soothing Test Material

[0063] To build a stable botanic active ingredients combination, including the oil solubility ingredients and water solubility ingredients for the efficacy test, a simple base containing at least one acrylates polymer and one preservative was built. The base provided stabilization for the botanic active ingredients combination during the test period and served as the placebo to verify the test results of the botanic active ingredients combination.

[0064] The primary equipment utilized during the in vitro testing included a carbon dioxide incubator and a microplate reader.

[0065] Soothing In-Vitro Test Results:

[0066] IL-1α is a pro-inflammatory factor and a non-hydrophobic polypeptide. It could only be released outside the cell after the cell membrane was damaged. Prostaglandin PGE2 is a metabolite of arachidonic acid, which was mainly synthesized by cyclooxygenase. PGE2 can dilate blood vessels, cause erythema, and induce clinical symptoms of inflammatory pain.

[0067] Compared to the placebo, the in-vitro test results demonstrate the compositions of the present disclosure provide soothing efficacy by decreasing the IL-1α and PGE2 content. As illustrated in Table 3, the content of IL-1α and PGE2 in the inventive examples E1 and E2 each  decreased relative to the comparative examples CE1-CE4 after the action of the analyte, particularly seen in E1. For example, the decrease in IL-1α of the comparative examples CE1 -CE4 demonstrated a range of 39.05%-59.41%, while E1 demonstrated a decrease of 37.40%and E2 demonstrated a decrease of 54.14%. As another example, the decrease in the decrease in IL-1αof the comparative examples CE1 -CE4 demonstrated a range of 39.05%-59.41%, while E1 demonstrated a decrease of 37.40%and E2 demonstrated a decrease of 54.14%. of the comparative examples CE1 -CE4 demonstrated a range of 25.23%-60.96%, while E1 demonstrated a decrease of 26.37%and E2 demonstrated a decrease of 47.87%. Accordingly, the decreasing content of IL-1α and PGE2 indicates the combination of botanic active ingredients have a soothing effect.

[0068] Soothing Consumer Use Testing:

[0069] Skin care product compositions containing the inventive examples E1 and E2 were further subjected to consumer use testing to determine soothing efficacy on skin. Questionnaires were used to investigate the consumer’s agreement to evaluate the sensory aspects of skin care products containing E1 and E2. Table 6 below illustrates the results of the consumer use testing.

[0070] Table 6

[0071] As illustrated in Table 6, 108 adults responded regarding the skin care product containing E1 and 109 adults responded regarding the skin care product containing E2. Of the 108 adults that responded regarding the skin care product containing E1, 94%agreed after one week that the skin care product soothed baby’s delicate / fragile skin, 92%agreed the skin care product was non-greasy, and 94%agreed the skin care product was non-sticky. Of the 109 adults that responded regarding the skin care product containing E2, 94%agreed after one week that the skin care product soothed baby’s delicate / fragile skin, 87%agreed the skin care product was non-greasy, and 82%agreed the skin care product was non-sticky.
