[0004] A bispecific antibody is an artificial protein that can simultaneously bind to two different types of antigens or two different epitopes. This dual specificity opens up a wide range of applications, including redirecting T cells to tumor cells, dual targeting of different disease mediators, and delivering payloads to targeted sites. The approval of catumaxomab (anti-EpCAM and anti-CD3) and blinatumomab (anti-CD19 and anti-CD3) has become a major milestone in the development of bispecific antibodies.

[0006] This disclosure relates to anti-EGFR / HER3 antibodies or antigen-binding fragments thereof, wherein the antibodies or antigen-binding fragments thereof specifically bind to EGFR and HER3. In some embodiments, the antibodies or antigen-binding fragments thereof have identical light chain variable regions. In some embodiments, the antibodies or antigen-binding fragments thereof have a common light chain. The disclosure also relates to antibody drug conjugates derived from these anti-EGFR / HER3 antibodies.

[0361] Anti-EGFR / HER3 bispecific antibodies were buffer exchanged into pH 6.0 (3 mg / mL histidine, 80 mg / mL sucrose, and 0.2 mg / mL Tween 80) . The antibodies were kept in sealed Eppendorf tubes at 40 ±2℃, 60%±5%RH and 4±3℃, for 7 days, and their thermal stability were evaluated. Alternatively, the bispecific antibodies were loaded into protein A column and eluted with a buffer (0.1mol / L HAc) at pH 3.5. Half of the antibodies were added 2M Tris buffer to make the pH into 7.5 immediately. The remain half were kept at pH 3.5 for 6 hours and then adjust the pH into 7.5. The diluted antibodies were kept in sealed Eppendorftubes at pH 3.5±0.1, 25±2℃ (hereinafter referred to as pH 3.5) for 6 hours to test stability at low pH.

[0441] The pharmacokinetic clearance rates of the anti-EGFR / HER3 ADC were determined in C57BL / 6 mice. Specifically, the mice were placed into four groups (6 mice per group) , and administered with 9F3-3E1-CPT2 (G1, 3 mg / kg; G2, 10 mg / kg) or Ref4-CPT2 (G3, 4 mg / kg; G4, 12 mg / kg) by intravenous injection. Blood samples were collected 3 days before administration and 15 minutes, 1 day, 3 day, 7 days, 10 days, 14 days and 21 days after administration.
