ANTIBODY, ANTIGEN-BINDING FRAGMENT AND MEDICAL USE THEREOFFIELD OF THE INVENTION

[0001] The present invention relates to the field of therapeutic monoclonal antibodies, in particular antibodies specific to claudin-6, and to use thereof in the treatment of diseases.BACKGROUND ART

[0002] Claudin-6 (CLDN6) is a type 2 membrane protein containing 220 amino acids. It belongs to Claudin family, which has 27 distinct proteins characterized by 4 transmembrane domains and 2 extracellular loops. Various claudin (CLDN) family members are located at tight junctions between epithelial cells. Recently, Claudin-6 has also been implicated in some critical intracellular signaling pathways and promoting tumor progression in cancers. Claudin-6 is expressed at elevated levels in multiple human cancers including ovarian and endometrial malignancies, with little or no detectable expression in normal adult tissue. In both ovarian and endometrial cancer patients, high level of Claudin-6 expression in malignant tissue is an independent prognostic marker for poorer progression-free and overall survival. The composite data demonstrating high cancer versus normal tissue expression in adults makes Claudin-6 an attractive target for the development of therapeutics against cancer.

[0003] Anti-CLDN6 antibodies have been described in WO2022096700A1, WO2021006328A1, WO2020168059A1, WO2023285560A1. There are several bispecific antibodies and antibody-drug conjugates targeting CLDN6 moved into clinical trials, such as AMG-794, SAIL66, TORL-1-23, and DS9606a.

[0004] There is a high degree of homology between CLDN6 and its family members, such as CLDN9, CLDN4 and CLDN3. It is challenging to develop CLDN6-specific antibodies. Due to the low number of available CLDN6-specific monoclonal antibodies, there is a need in the art for better anti-CLDN6 antibodies that have efficient internalization or other functional properties not possessed by the known antibody. There is also a need for additional diagnostic tools for detecting CLDN6 expressions in CLDN6-related disease conditions. The instant invention is directed to addressing these and other needs.SUMMARY OF THE INVENTION

[0005] An object of the present invention is to provide a novel antibody or an antigen-binding fragment of the antibody (hereinafter, also referred to as an antibody, etc. ) which binds to claudin-6, a molecule comprising the antibody, and a pharmaceutical composition having cytotoxic activity, etc. which includes the antibody, etc. or the molecule as an active ingredient.

[0006] The present inventors have conducted extensive research to achieve this object and have realized the present invention by developing a novel anti-claudin-6 antibody and a molecule comprising this antibody.

[0007] In one aspect, the present invention provides an antibody or antigen-binding fragment thereof that specifically binds to CLDN6, wherein, the antibody comprises:

[0122] EXAMPLE 3 Characterization of anti-claudin-6 recombinant antibody cell internalization in PA-1 cells by indirect killing assay
