Description

FIELD OF THE INVENTION The present invention relates to the field of pharmaceutical chemistry, specifically, relates to a salt crystal form of plinabulin, the preparation methods therefor and the use thereof. BACKGROUND OF THE INVENTION Plinabulin, ((3Z,6Z)-3-[(5-tert-butyl-1H-imidazol-4-yl)methylene]-6-(benzylidene)-2,5-piperazinedione), is a synthetic analogue of phenylahistin, a diketopiperazine isolated from marine and terrestrial Aspergillus species, whose structure is shown below. () M ™ 'AA-''11 O / V" Structurally, plinabulin differs from colchicine and combretastatin-like analogues thereof (e.g., combretastatin phosphate), and binds at or near the colchicine-binding site on tubulin monomers. Previous studies have demonstrated that, compared with colchicine, plinabulin induces depolymerization of tubulin in vascular endothelial cells and monolayer permeability at low concentrations, and triggers apoptosis in Jurkat leukemia cells. Studies of plinabulin as a single agent in patients with advanced malignant tumors (lung cancer, prostate cancer and colon cancer) have exhibited favorable pharmacokinetic, pharmacodynamic and safety profiles. Nevertheless, there remains a need to provide a salt crystal form of plinabulin with appropriate physicochemical properties to improve its solubility in solvents.
