Abstract
Description
As used herein, when referring to the amino acid sequence of PD-1 protein (NCBI GenBank: NM_005018), it includes the full length of PD-1 protein, or the extracellular fragment PD-1 ECD of PD-1, or a fragment comprising PD-1 ECD, and it also includes a fusion protein of the full length of PD-1 protein or a fusion protein of PD-1 ECD, such as a fragment fused to an Fc protein fragment of mouse or human IgG (mFc or hFc). However, those skilled in the art will appreciate that in the amino acid sequence of the PD-1 protein, mutations or variations (including but not limited to, substitutions, deletions, and / or additions) can be naturally produced or artificially introduced without affecting biological functions thereof. Therefore, in the present disclosure, the term “PD-1 protein” shall include all such sequences, including their natural or artificial variants. In addition, when describing a sequence fragment of the PD-1 protein, it also includes the corresponding sequence fragments in their natural or artificial variants. As used herein, when referring to the amino acid sequence of lymphocyte-activation gene 3 (LAG3), it includes the full length of the LAG3 protein, or the extracellular fragment LAG3 ECD of LAG3, or a fragment comprising LAG3 ECD, and it also includes a fusion protein of the full length of the LAG3 protein or a fusion protein of LAG3 ECD, such as a fragment fused to an Fc protein fragment of mouse or human IgG (mFc or hFc). However, those skilled in the art will appreciate that in the amino acid sequence of the LAG3 protein, mutations or variations (including but not limited to, substitutions, deletions, and / or additions) can be naturally produced or artificially introduced without affecting biological functions thereof. Therefore, in the present disclosure, the term “LAG3 protein” shall include all such sequences, including their natural or artificial variants. In addition, when describing a sequence fragment of the LAG3 protein, it also includes the corresponding sequence fragments in their natural or artificial variants. As used herein, when referring to the amino acid sequence of a CD47 protein (NCBI GenBank: NP_001768.1), it includes the full length of the CD47 protein, or the extracellular fragment CD47 ECD of CD47, or a fragment comprising CD47 ECD, and it also includes a fusion protein of the full length of the CD47 protein or a fusion protein of CD47 ECD, such as a fragment fused to an Fc protein fragment of mouse or human IgG (mFc or hFc). However, those skilled in the art will appreciate that in the amino acid sequence of the CD47 protein, mutations or variations (including but not limited to, substitutions, deletions, and / or additions) can be naturally produced or artificially introduced without affecting biological functions thereof. Therefore, in the present disclosure, the term “CD47 protein” shall include all such sequences, including their natural or artificial variants. In addition, when the sequence fragment of the CD47 protein is described, the CD47 protein also includes the corresponding sequence fragments in natural or artificial variants thereof. In the present disclosure, the term “ADCP” refers to antibodydependent cellular phagocytosis. The Fc fragment of an antibody that binds to a cell surface antigen binds to the Fc receptor of a phagocytically active cell, such as a macrophage, which in turn mediates phagocytosis of the antibody-bound cells by phagocytic cells. In the present disclosure, the terms “first” (e.g., first protein functional region, first antibody, or first linker), “second” (e.g., second protein functional region, second antibody, or second linker), “third” (e.g., third linker), and “fourth” (e.g., fourth linker) are used for distinguishing or clarity in expression and do not carry typical sequential meanings, unless otherwise specified. Treatment of Raji cells with IFN-y: On day 5 of macrophage induction, Raji cells (purchased from the Chinese Academy of Sciences) were conventionally passaged and treated with IFN-y (sinobiological, Cat: 11725-HNAS-100, at a final concentration of 100 ng / mL), and the cells were cultured in an incubator for 2 days. CFSE staining treatment of Raji cells: On the day of the experiment, the Raji cells that had been treated with IFN-y for 2 days were conventionally collected and counted; CFSE (Biolegend, Cat: 423801) was diluted with serum-free RPMI-1640 to a final concentration of 0.25 gM for staining and labeling; the cells were incubated in a 37 °C, 5% CO 2 incubator for labeling for 15 min and centrifuged; after the supernatant was discarded, the cells were resuspended in 10 mL of RPMI-1640 containing 10% FBS; and the cells were incubated in a 37 °C, 5% CO2 incubator for 10 min to restore the state, centrifuged, resuspended, and counted. According to the groups (the negative control was macrophages + Raji cells; the isotype control was the negative control + hIgG4 at a final concentration of 10 nM; the Bs-PL022B group was the negative control + Bs-PL022B at a final concentration of 30 nM; the 6F7 H1L1(hG4) group was the negative control + 6F7 H1L1(hG4) at a final concentration of 1 / 10 nM; and the 6F7 H1L1(hG4) + Bs-PL022B group was the negative control + 6F7 H1L1(hG4) at a final concentration of 1 / 10 nM + Bs-PL022B at a final concentration of 30 nM), macrophages and CFSE-labeled Raji cells that had been treated with IFN-y for 2 days were plated in a V-bottom 96-well plate, and different antibodies were added. The cells were cultured in a 37 °C, 5% CO2 incubator for 2 h (Raji cells:macrophages = 3:1). After washing, APC anti-mouse / human CD11b (BioLegend, Cat: 101212) diluted at 1:100 was added at 100 gL / well, and the mixture was incubated on ice for 30 min in the dark. After washing, 200 gL of 1% PBSA was added to resuspend the cells, and then the phagocytosis rate (CD11b+CFSE+%) was detected by flow cytometry. The results are shown in FIG. 1. The results show that compared with the isotype control (hIgG4), the anti-CD47 monoclonal antibody 6F7 H1L1(hG4) and the anti-LAG3-anti-PD-1 bispecific antibody Bs-PL022B in combination with the anti-CD47 monoclonal antibody 6F7 H1L1(hG4) can both effectively promote phagocytosis of IFN—Y—treated Raji cells by macrophages, and the anti-LAG3-anti-PD-1 bispecific antibody in combination with the anti-CD47 monoclonal antibody has stronger pro-phagocytic activity than the anti-CD47 monoclonal antibody.
