Abstract
Description
The present invention relates to a cosmetic composition for moisturizing the skin comprising mineral components and quinoa extract. This application claims the benefit of the filing date of Korean Patent Application No. 10-2014-0037143 filed with the Korean Intellectual Property Office on March 28, 2014, the entire contents of which are incorporated herein. The skin performs a crucial role as a barrier that protects the individual from the external environment. This barrier function serves to defend against various external stimuli (chemicals, air pollutants, dry environments, ultraviolet rays, etc.) and prevents the excessive loss of body moisture through the skin; this protective function can be maintained only when the stratum corneum, composed of keratinocytes, is properly formed. The stratum corneum (horney layer), the outermost layer of the epidermis, is formed from keratinocytes and consists of fully differentiated keratinocytes and a surrounding lipid layer. Keratinocytes are characteristic cells that rise to the surface of the skin as basal cells, which continuously proliferate in the lowest layer of the epidermis, undergo gradual changes in form and function. After a certain period, old keratinocytes are shed from the skin and new keratinocytes take over their function. This repetitive series of changes is called “epidermal cell differentiation” or “keratinization.” Furthermore, during the keratinization process, keratinocytes produce natural moisturizing factors (NMF) and intercellular lipids (ceramide, cholesterol, fatty acids) to form the stratum corneum, which gives the stratum corneum firmness and flexibility, thereby enabling it to function as a skin barrier. The function of this stratum corneum can easily be lost due to lifestyle factors such as excessive washing or bathing, environmental factors such as dry air pollutants, and endogenous conditions like atopic or senile skin. In fact, due to the increasing variety of factors in modern times, there has been a recent trend of increasing complaints regarding dry skin symptoms and the resulting disorders. It is known that the retention of moisture in each layer through an increase in Natural Moisturing Factors (NFFs), which are composed of specific amino acids, is crucial as a mechanism for the expression of moisturizing action that promotes moisture retention in skin tissues. Since the amino acids constituting the main components of the Natural Moisturizing Factor are supplied through the breakdown of a protein called Filggrin produced by epidermal keratinocytes, the ability to promote Filggrin production plays an important role in the expression of moisturizing action. In addition, the types of keratin expressed in keratinocytes are specific depending on the degree of cell differentiation; K5 and K14 are primarily expressed in basal keratinocytes, and as keratinocytes leave the basal layer and move to the upper spinous layer, different keratin pairs, K10 and K1, begin to be expressed instead of the K5 and K14 keratin pairs. Loricrin is expressed during terminal differentiation, and since it binds to the cell membrane in the upper granular layer to form a complete protein, it is used as a marker to track the terminal differentiation process of keratinocytes. Therefore, in order to prevent skin aging and maintain skin health against external environmental changes such as dry air, ultraviolet rays, and various pollutants, and to form a stratum corneum that strengthens the skin barrier to prevent moisture loss from skin tissue while promoting moisture retention in skin tissue, there is a need to develop a skin moisturizing substance that promotes the expression of filaggrin and loricrin. The present invention aims to provide a cosmetic composition that promotes the expression of filaggrin and loricrin in order to form a stratum corneum to strengthen the skin barrier, thereby preventing moisture loss in skin tissue while promoting moisture retention in skin tissue. The present invention provides a cosmetic composition for moisturizing the skin comprising one or more mineral components selected from the group consisting of calcium, manganese, magnesium, and zinc; and quinoa extract. The skin moisturizing cosmetic composition of the present invention includes quinoa extract together with calcium, manganese, magnesium, or zinc among mineral components, and when applied to the human body, it promotes the expression of natural moisturizing factors, thereby promoting moisture retention in skin tissue and strengthening the skin barrier, resulting in an excellent skin moisturizing effect that retains moisture within the skin. Figure 1 shows the results of filaggrin gene expression for the composition treatment group according to Comparative Example 1 (mineral treatment group), the composition treatment group according to Comparative Example 2 (quinoa treatment group), and the composition treatment group according to Example 1 (mineral + quinoa treatment group) as relative values to the untreated group. Figure 2 shows the Loricrin gene expression results relative to the untreated group for the composition treatment group according to Comparative Example 1 (mineral treatment group), the composition treatment group according to Comparative Example 2 (quinoa treatment group), and the composition treatment group according to Example 1 (mineral + quinoa treatment group). In this specification, the term "skin" refers to tissue covering the surface of an animal's body, and is a broad concept that includes not only tissue covering the surface of the body such as the face or body, but also the scalp and hair. In this specification, "mineral component" refers to elements other than oxygen (O), carbon (C), hydrogen (H), and nitrogen (N), which are the main components of organic matter and are necessary for physiological activities such as growth and maintenance of the human body. In this specification, the term "extract" is a broad concept that includes all substances obtained by extracting components of natural products, regardless of the extraction method, extraction solvent, extracted components, or form of the extract. In this specification, quinoa refers to the flowers, leaves, fruits, and seeds of the plant of the genus Chenopodium (scientific name: Chenopodium quinoa) of the family Amaranthaceae. In one embodiment of the present invention, the quinoa extract may be prepared according to a conventional method known in the art, that is, using a conventional solvent under conditions of conventional temperature and pressure. Preferably, quinoa flowers, leaves, fruits, and seeds are dried, then extracted with an extraction solvent selected from the group consisting of water, anhydrous or hydrated lower alcohols having 1 to 4 carbon atoms, acetone, ethyl acetate, butyl acetate, and 1,3-butylene glycol, and then filtered and concentrated. In one embodiment of the present specification, the quinoa extract is 0.001% by weight or more and 5% by weight or less with respect to the total weight of the skin moisturizing cosmetic. When the content of the quinoa extract is applied to the skin together with mineral components, it promotes an increase in the expression of filaggrin by the mineral components, thereby promoting skin moisturization, and also promotes an increase in the expression of loricrin by the mineral components, thereby promoting the strengthening of the skin barrier. If the quinoa extract is less than 0.001% by weight with respect to the total weight of the skin moisturizing cosmetic, it is difficult to expect an effect of promoting the expression of filaggrin and loricrin, and if it exceeds 5% by weight, there are difficulties in stability or formulation preparation. In one embodiment of the present specification, the mineral component is 0.001% by weight or more and 1% by weight or less with respect to the total weight of the skin moisturizing cosmetic. When the content of the mineral component is applied to the skin together with quinoa extract, it increases the expression of filaggrin by the mineral component, thereby promoting skin moisturization, and also promotes the increase in the expression of loricrin by the mineral component, thereby strengthening the skin barrier. If the content is less than 0.001% by weight with respect to the total weight of the skin moisturizing cosmetic, it is difficult to expect the effect of promoting filaggrin and loricrin expression, and if it exceeds 1% by weight, there are difficulties in stability or formulation preparation. In one embodiment of the present specification, the mineral component is one or more selected from the group consisting of calcium, manganese, magnesium and zinc. In one embodiment of the present specification, the mineral component includes all of calcium, manganese, magnesium, and zinc. When the mineral component includes all of calcium, manganese, magnesium, and zinc, the skin moisturizing effect and skin barrier strengthening effect are superior compared to when it includes calcium, manganese, magnesium, and zinc separately. In one embodiment of the present specification, the weight ratio of calcium : manganese : magnesium : zinc of the mineral components is 5 to 20 : 1 to 5 : 5 to 20 : 0.5 to 5. In one embodiment of the present specification, the cosmetic composition for skin moisturization has the ability to promote keratinocyte differentiation. The mineral components and quinoa extract of the composition promote the expression of skin moisturizing factor genes, thereby providing an excellent effect of promoting keratinocyte differentiation. In one embodiment of the present specification, the cosmetic composition for moisturizing the skin increases skin moisturizing factors. In one embodiment of the present specification, the skin moisturizing factors are filaggrin and loricrin. In one embodiment of the present specification, the skin moisturizing cosmetic composition is the food, cosmetic, or pharmaceutical composition. The structure and effects of the present invention will be explained in more detail below with reference to test examples and embodiments. However, these test examples and experimental examples are provided for illustrative purposes only to aid in understanding the present invention, and the scope and range of the present invention are not limited by the following embodiments. <Example 1> Quinoa extract was prepared by adding 1 liter of a 1:1 (volume:volume) mixed solvent of water and ethanol to 0.2 kg of quinoa fruit and seeds, extracting the mixture, filtering it through a membrane filter, and then concentrating it.
