CRYSTAL FORM OF PYRIDOPYRIMIDINE DERIVATIVE, PREPARATION METHOD THEREFOR AND USE THEREOF SPECIFICATION CROSS-REFERENCE TO RELATED APPLICATIONS

[0001] The present disclosure claims the benefit and priority of Chinese Patent Application No. CN202410040440.3, filed with the China National Intellectual Property Administration on January 10, 2024, and Chinese Patent Application No. CN202510018660.0, filed with the China National Intellectual Property Administration on January 06, 2025, the entire contents of which are incorporated herein by reference. TECHNICAL FIELD

[0002] The present disclosure provides a crystal form of a pyridopyrimidine derivative, a preparation method therefor, and a use thereof, and specifically discloses a crystal form of a compound of formula (I), a preparation method therefor, and a use thereof. BACKGROUND

[0003] Mutations in NRAS, HRAS, and KRAS within the RAS family account for nearly one-quarter of all human cancers, making them one of the most common cancer-related gene mutations. These mutations cover almost all cancer types and cause 1 million deaths globally each year. Among them, KRAS is the most common oncogene (accounting for 85% of all RAS mutations) and is present in 90% of pancreatic cancers, 30-40% of colon cancers, and 15- 20% of lung cancers (mostly non-small cell lung cancer). Depending on the specific mutations present, G12C, G12D, and G12R are the most common KRAS mutations in patients. In addition to these, there are G12A, G12S, G12V, and the like.

[0004] The KRAS protein belongs to the small GTPase family, possesses GTPase hydrolytic activity, and participates in various cell signaling pathways related to cell survival and growth. KRAS is in an activated state when bound to GTP (guanosine triphosphate), which can activate a series of downstream pathways. After the bound GTP is hydrolyzed into GDP (guanosine diphosphate), KRAS enters an inactivated state. Under normal conditions, once KRAS is activated, it is immediately inactivated under the action of its own hydrolase. However, oncogenic mutated KRAS exhibits impaired GTPase activity, making it more prone to remaining in the GTP-bound state. This leads to continuous activation of the KRAS protein, which in turn continuously activates downstream pathways such as the RAF / MEK / ERK and PI3K / Akt pathways, causing continuous cell proliferation, differentiation, tumor formation, and promoting tumor progression. KRAS G12D mutations account for 20%-50% of all KRAS-mutated tumors; however, no KRAS G12D drug has been marketed to date. Therefore, researching small-molecule oral inhibitors capable of inhibiting KRAS G12D is of extremely high value for the treatment of malignant tumors. SUMMARY

For any specific crystal form, there may be an error in the peak positions, and the measurement error of the 2<com:Math><semantics>θ<annotation encoding="application / x-tex">\theta< / annotation>< / semantics>< / com:Math> values may be <com:Math><semantics>±0.2∘<annotation encoding="application / x-tex">\pm 0.2^{\circ}< / annotation>< / semantics>< / com:Math>, <com:Math><semantics>±0.15∘<annotation encoding="application / x-tex">\pm 0.15^{\circ}< / annotation>< / semantics>< / com:Math>, and <com:Math><semantics>±0.1∘<annotation encoding="application / x-tex">\pm 0.1^{\circ}< / annotation>< / semantics>< / com:Math>. Therefore, when identifying each crystal form, such error should be taken into account, and those within the error are also encompassed within the scope of the present disclosure.

[0044] Unless otherwise specified, a solid wedge bond ( ) and a dashed wedge bond ( ) are used to indicate the absolute configuration of a stereocenter, and a straight solid bond ( ) and a straight dashed bond ( ) are used to indicate the relative configuration of a stereocenter. [0045] The present disclosure will be specifically described below by way of examples, which are not meant to limit the present disclosure in any way.

[0046] The structures of the compounds of the present disclosure can be confirmed by conventional methods well known to those skilled in the art; if the present disclosure involves the absolute configuration of a compound, the absolute configuration can be confirmed by conventional technical means in the art. For example, by single-crystal X-ray diffraction (SXRD), diffraction intensity data of a grown single crystal are collected using a Bruker D8 Venture diffractometer, with a CuK<com:Math><semantics>α<annotation encoding="application / x-tex">\alpha< / annotation>< / semantics>< / com:Math> radiation light source and a <com:Math><semantics>φ / ω<annotation encoding="application / x-tex">\varphi / \omega< / annotation>< / semantics>< / com:Math> scan mode; after collecting the relevant data, the crystal structure is further resolved by a direct method (Shelxs97) to confirm the absolute configuration.

[0047] In the present disclosure, the term "therapeutically effective amount" means an amount of a drug or agent that (i) treats a specific disease, condition, or disorder, or (ii) alleviates, ameliorates, or eliminates one or more symptoms of a specific disease, condition, or disorder. The amount of the crystal form A of the compound of formula (I) of the present disclosure or the pharmaceutical composition thereof that constitutes a "therapeutically effective amount" varies depending on the drug or agent, the disease state and its severity, the mode of administration, and the age of the subject to be treated, but can be routinely determined by a person skilled in the art based on their own knowledge and the contents of the present disclosure.

[0048] The term "treating" or "treatment" means administering a drug or agent to ameliorate or eliminate a disease or one or more symptoms associated with said disease, and includes:

[0049] (i) inhibiting the disease or disease state, i.e., arresting its development; and

[0050] (ii) relieving the disease or disease state, i.e., causing regression of the disease or disease state.

[0051] In the present disclosure, the term "subject" includes humans and animals, for example, mammals (such as primates, cattle, horses, pigs, dogs, cats, mice, rats, rabbits, goats, sheep, and avian species, and the like).

[0052] The term "pharmaceutically acceptable excipient" refers to those excipients that do not cause significant irritation to an organism and do not impair the biological activity and properties of the active substance. Conventional pharmaceutical excipients include fillers, absorbents, wetting agents, binders, disintegrants, lubricants, carriers, diluents, isotonicity adjusters, pH adjusters, and the like. Suitable excipients are well known to those skilled in the art, such as carbohydrates, waxes, water-soluble and / or water-swellable polymers, hydrophilic or hydrophobic materials, gelatin, oils, solvents, water, and the like.

[0053] Typical routes for administering the crystal form A of the compound of formula (I) of the present disclosure or the pharmaceutical composition thereof include, but are not limited to, oral, rectal, topical, inhalation, parenteral, sublingual, intravaginal, intranasal, intraocular, intraperitoneal, intramuscular, subcutaneous, and intravenous administration.

[0054] The pharmaceutical composition of the present disclosure can be manufactured by methods well known in the art, such as conventional mixing methods, dissolving methods, granulating methods, emulsifying methods, lyophilizing methods, and the like.

[0055] In some embodiments, the pharmaceutical composition may be in an oral form. For oral administration, the pharmaceutical composition can be formulated by mixing the active substance with pharmaceutically acceptable excipients well known in the art. These excipients enable the crystal form A of the compound of formula (I) of the present disclosure or the pharmaceutical composition thereof to be formulated into tablets, pills, lozenges, dragees, capsules, liquids, gels, syrups, slurries, suspensions, and the like, for oral administration to a patient.

[0056] Solid oral compositions can be prepared by conventional mixing, filling, or tableting methods. For example, they can be obtained by mixing the active compound with a solid excipient, optionally grinding the resulting mixture, adding other suitable excipients if necessary, and then processing the mixture into granules to obtain cores of tablets or dragees.

[0057] The therapeutically effective amount of the crystal form A of the compound of formula (I) contained in the pharmaceutical composition of the present disclosure is selected from 0.001 mg / kg body weight to 1000 mg / kg body weight, for example, 0.01 mg / kg body weight to 500 mg / kg body weight, in the form of a single dose or divided doses.

[0058] A person skilled in the art recognizes that the measurement data of XRPD peak positions and / or intensities for a given crystal form of the same compound will vary within an error range. The <com:Math><semantics>2θ<annotation encoding="application / x-tex">2\theta< / annotation>< / semantics>< / com:Math> values in the present disclosure encompass an appropriate error range, which is generally represented by "<com:Math><semantics>±<annotation encoding="application / x-tex">\pm< / annotation>< / semantics>< / com:Math>". For example, a <com:Math><semantics>2θ<annotation encoding="application / x-tex">2\theta< / annotation>< / semantics>< / com:Math> value represented by a specific angle value <com:Math><semantics>±0.20∘<annotation encoding="application / x-tex">\pm 0.20^{\circ}< / annotation>< / semantics>< / com:Math> in the present disclosure means that the specific angle value therein has an error fluctuation range of <com:Math><semantics>±0.20∘<annotation encoding="application / x-tex">\pm 0.20^{\circ}< / annotation>< / semantics>< / com:Math>; that is, <com:Math><semantics>8.09∘±0.2∘<annotation encoding="application / x-tex">8.09^{\circ} \pm 0.2^{\circ}< / annotation>< / semantics>< / com:Math> indicates that 20 is within the range of 7.89° to 8.29°. Depending on sample preparation techniques, calibration techniques applied to instruments, human operational variations, and the like, a person skilled in the art recognizes that an appropriate error range for XRPD diffraction angles can be <com:Math><semantics>±0.20∘<annotation encoding="application / x-tex">\pm 0.20^{\circ}< / annotation>< / semantics>< / com:Math>, <com:Math><semantics>±0.15∘<annotation encoding="application / x-tex">\pm 0.15^{\circ}< / annotation>< / semantics>< / com:Math>, <com:Math><semantics>±0.10∘<annotation encoding="application / x-tex">\pm 0.10^{\circ}< / annotation>< / semantics>< / com:Math>, <com:Math><semantics>±0.05∘<annotation encoding="application / x-tex">\pm 0.05^{\circ}< / annotation>< / semantics>< / com:Math>, or less, and peak intensities allow for a certain degree of variability. When used to describe an XRPD pattern, the term "substantially the same" or "substantially as shown in..." refers to a pattern including at least 50%, at least 60%, at least 70%, at least 80%, at least 90%, at least 95%, or at least 99% of diffraction peaks whose diffraction angles are within a standard deviation range of <com:Math><semantics>±0.2∘2θ<annotation encoding="application / x-tex">\pm 0.2^{\circ} 2\theta< / annotation>< / semantics>< / com:Math>.

[0059] Since a person skilled in the art recognizes that the measurement data of a DSC pattern for a given crystal form of the same compound will vary within an error tolerance, the peak onset values and single peak maximum values (expressed in degrees Celsius) allow for an appropriate error range. Generally, the error range is represented by "±". For the same crystal form of the same compound, the error in thermal transition temperature and melting point is typically within <com:Math><semantics>±3.00<annotation encoding="application / x-tex">\pm 3.00< / annotation>< / semantics>< / com:Math>°C in consecutive analyses. For example, a peak value of "204.65±3.00°C" indicates that it is within the range of 201.65°C to 207.65°C. Depending on sample preparation techniques, calibration techniques applied to instruments, human operational variations, and the like, a person skilled in the art recognizes that an appropriate error range for a single peak maximum value can be <com:Math><semantics>±3.0∘<annotation encoding="application / x-tex">\pm 3.0^{\circ}< / annotation>< / semantics>< / com:Math>C, <com:Math><semantics>±2.0∘<annotation encoding="application / x-tex">\pm 2.0^{\circ}< / annotation>< / semantics>< / com:Math>C, or less.

[0060] The term "optional" or "optionally" means that the subsequently described event or circumstance may or may not occur, and the description includes instances where the event or circumstance occurs and instances where it does not.

[0108] T The present disclosure is described in detail below by way of examples, which are not meant to impose any adverse limitation on the present disclosure. The present disclosure has been described in detail herein, including specific embodiments thereof disclosed therein. It will be apparent to a person skilled in the art that various changes and modifications can be made to the specific embodiments of the present disclosure without departing from the spirit and scope of the present disclosure.

[0109] Example 1: Synthesis of the compound of formula (I) [Image disponible dans le document PDF, Image available in the PDF document]

[0110] Step 1: Preparation of intermediate 2-2

[0111] Intermediate 2-1 (50 g, 418.09 mmol) was dissolved in dichloromethane (500 mL), and triethylamine (84.61 g, 836.17 mmol) was added thereto, followed by the addition of di- tert-butyl dicarbonate (100.37 g, 459.90 mmol). The reaction was carried out at 25°C for 16 hours. Water (100 mL) was added to the reaction solution, and the phases were separated. The organic phase was dried over anhydrous sodium sulfate, filtered, and concentrated. The crude product was purified by silica gel column chromatography (petroleum ether / ethyl acetate = <com:Math><semantics>100 / 1<annotation encoding="application / x-tex">100 / 1< / annotation>< / semantics>< / com:Math> to <com:Math><semantics>50 / 1<annotation encoding="application / x-tex">50 / 1< / annotation>< / semantics>< / com:Math>) to obtain intermediate 2-2. MS: m / z = <com:Math><semantics>206.1<annotation encoding="application / x-tex">206.1< / annotation>< / semantics>< / com:Math> [M+Na]<com:Sup>+< / com:Sup>.

[0112] Step 2: Preparation of intermediate 2-3

[0113] Intermediate 2-2 (25 g, 136.43 mmol) was dissolved in anhydrous tetrahydrofuran (350 mL), and 3,7-dipropyl-3,7-diazabicyclo[3.3.1]nonane (37.31 g, 177.36 mmol) was added thereto. The mixture was cooled to -65°C, and sec-butyllithium (1.3 M, 157.42 mL) was slowly added dropwise. After reacting for 1 hour, methyl chloroformate (15.73 g, 166.44 mmol) was added dropwise, and the reaction was carried out at -65°C for 2 hours. The reaction mixture was quenched by the dropwise addition of saturated ammonium chloride (20) mL), and extracted with ethyl acetate (300 mL * 2). The organic phases were combined, washed with saturated brine (150 mL), dried over anhydrous sodium sulfate, filtered, and concentrated. The crude product was purified by silica gel column chromatography (PE / EA <com:Math><semantics>=50 / 1<annotation encoding="application / x-tex">= 50 / 1< / annotation>< / semantics>< / com:Math> to 25 / 1) to obtain intermediate 2-3. MS: m / z = 186.0 [M-tBu+H]<com:Sup>+< / com:Sup>.

[0114] Step 3: Preparation of intermediate 2-4

[0115] Intermediate 2-3 (12 g, 49.73 mmol) was dissolved in tetrahydrofuran (120 mL), and 3-chloro-2-chloromethylpropene (24.87 g, 198.94 mmol) was added thereto. The mixture was cooled to -40 °C, and lithium bis(trimethylsilyl)amide (1 M, 99.47 mL) was slowly added dropwise. The temperature was gradually raised to 20°C, and the reaction was carried out for 2 hours. The reaction mixture was quenched by the dropwise addition of saturated ammonium chloride (20 mL), and extracted with ethyl acetate (150 mL * 2). The organic phases were combined, washed with saturated brine (80 mL), dried over anhydrous sodium sulfate, filtered, and concentrated. The crude product was purified by silica gel column chromatography (petroleum ether / ethyl acetate = <com:Math><semantics>20 / 1<annotation encoding="application / x-tex">20 / 1< / annotation>< / semantics>< / com:Math> to <com:Math><semantics>10 / 1<annotation encoding="application / x-tex">10 / 1< / annotation>< / semantics>< / com:Math>) to obtain a relative configuration mixture of intermediate <com:Math><semantics>2−4<annotation encoding="application / x-tex">2-4< / annotation>< / semantics>< / com:Math>. TLC thin-layer chromatography (petroleum ether:acetone = <com:Math><semantics>5:1<annotation encoding="application / x-tex">5:1< / annotation>< / semantics>< / com:Math>) was developed twice, wherein the Rf value of 2-4 was 0.5, and the Rf value of its isomer was 0.55. The retention time of 2-4 on LCMS (chromatographic column: Agilent Poroshell 120 EC-C18 2.7 micrometers 3.0 * 30 mm; mobile phase: A: water (0.037% formic acid) - B: acetonitrile (0.0187% formic acid); gradient: B: 5%-95%) was 0.776 min, and the retention time of its isomer was 0.801 min. [0116] Step 4: Preparation of intermediate 2-5

[0117] Intermediate 2-4 (3.6 g, 10.92 mmol) was dissolved in hydrogen chloride / ethyl acetate (4 M, 27.29 mL), and the reaction was carried out at 20°C for 2 hours. The reaction mixture was concentrated under reduced pressure to obtain a crude hydrochloride salt of intermediate 2-5. MS: <com:Math><semantics>m / z=230.1[M+H]+<annotation encoding="application / x-tex">m / z = 230.1 [M+H]^+< / annotation>< / semantics>< / com:Math>.

[0118] Step 5: Preparation of intermediate 2-6

[0119] The hydrochloride salt of intermediate 2-5 (2.9 g) was dissolved in methanol (100 mL), and potassium carbonate (4.52 g, 32.69 mmol) was added thereto. The reaction was carried out at 20°C for 2 hours. Dichloromethane (80 mL) was added to the reaction mixture, and the mixture was filtered and concentrated. The crude product was purified by silica gel column chromatography (petroleum ether / ethyl acetate = <com:Math><semantics>10 / 1<annotation encoding="application / x-tex">10 / 1< / annotation>< / semantics>< / com:Math> to <com:Math><semantics>5 / 1<annotation encoding="application / x-tex">5 / 1< / annotation>< / semantics>< / com:Math>) to obtain intermediate 2- 6. The reaction was carried out at 0°C for 2 hours. The reaction mixture was diluted by the dropwise addition of ethyl acetate (10 mL), and water (0.63 mL) was added dropwise. The mixture was dried over anhydrous sodium sulfate, filtered, and concentrated to obtain crude intermediate 2- 7. MS: <com:Math><semantics>m / z=166.1[M+H]+<annotation encoding="application / x-tex">m / z = 166.1 [M+H]^+< / annotation>< / semantics>< / com:Math>.

[0122] Step 7: Preparation of intermediate 1-2

[0123] Intermediate 1-1 (25 g, 99.03 mmol) was dissolved in dichloromethane (200 mL), and the mixture was cooled to -30°C. Subsequently, N,N-diisopropylethylamine (38.40 g, 297.08) mmol) and 1-1B (21.02 g, 99.03 mmol) were sequentially added thereto, and the reaction was carried out at -30°C for 2 hours. The reaction mixture was directly concentrated under reduced pressure to obtain crude intermediate 1-2. MS: <com:Math><semantics>m / z=428.1[M+1]+<annotation encoding="application / x-tex">m / z = 428.1 [M+1]^+< / annotation>< / semantics>< / com:Math>.

[0124] Step 8: Preparation of intermediate 1-3A

[0163] Table 11. Main reagents [Image disponible dans le document PDF, Image available in the PDF document]

[0164] II. Experimental method

[0165] Cell culture medium for experiment: Fetal bovine serum at a final concentration of 10% and a penicillin / streptomycin solution at a final concentration of 1% were added into a DMEM cell culture medium, and the mixture was stored at 4°C for future use.

[0166] GP2D cells that had reached 80% cell confluence were digested with trypsin, centrifuged, resuspended, and counted. A cell suspension was prepared using the culture medium. The cell suspension was added into a 96-well plate at an amount of 180 µL / well (1000 cells / well). The plate was placed in a cell incubator containing 5% CO<com:Sub>2< / com:Sub> and cultured at 37°C overnight.

[0167] The test compound was diluted with a DMSO solution to 200 µM, and then further diluted with the complete culture medium to 10 µM. Taking 10 µM as an initial concentration, a 5-fold serial dilution was performed to obtain 9 concentrations. After the cells were cultured overnight, the diluted mixture solutions were respectively transferred to the corresponding cell plates. The final concentration of the compound took 1 µM as an initial concentration, and a 5-fold serial dilution was performed to obtain 9 concentrations. The mixtures were mixed well, and placed in a cell incubator containing 5% CO<com:Sub>2< / com:Sub> and cultured at 37°C for 5 days.

[0168] After the compound was incubated for the above-mentioned time, the 96-well cell culture plate was taken out. After 50 µL of the supernatant was discarded, 80 µL of 3D- CellTiter Glo was added thereto, and the mixture was incubated with shaking at room temperature for 30 minutes. The plate was read on an Envision plate reader using the *US LUM-US LUM 96 (cps) program.

[0169] The measured signal values of each group were normalized by subtracting the signal value of a cell-free control well. Data were calculated according to the following formula for the cell viability after the compound treatment: % Inhibition rate = (1 - (RFU compound - AVER(RFU negative control)) / (AVER(RFU positive control) - AVER(RFU negative control))) x 100%. Positive control: cells treated with 0.5% DMSO; negative control: cell-free wells containing only the complete culture medium. Prism 8.3.0 was used for plotting and calculating the IC<com:Sub>50< / com:Sub> value of the compound. The calculation formula for the IC<com:Sub>50< / com:Sub> value is log(inhibitor) vs. response (three parameters): <com:Math><semantics>Y=Bottom+(Top−Bottom) / (1+10(X−Bottom))<annotation encoding="application / x-tex">Y = Bottom + (Top - Bottom) / (1 + 10^{(X - Bottom)})< / annotation>< / semantics>< / com:Math> <com:Math><semantics>LogIC50<annotation encoding="application / x-tex">LogIC_{50}< / annotation>< / semantics>< / com:Math>).
