[0001] ASSEMBLY AND METHODS FOR SPRAY DRYING

[0002] TECHNICAL FIELD OF THE INVENTION

[0003] The present invention is in the technical field of spray drying. In particular, the present invention is related to an assembly that can be installed into a support structure to form a spray drying apparatus. The present invention is also related to methods for producing the assembly, methods for producing the spray drying apparatus by installing the assembly into the support structure, and methods using the spray-drying apparatus comprising the installed assembly to produce a spray-dried product. Further, the present invention relates to methods of removing the assembly from the support structure and installing a further assembly, such that further spray drying with a different product can be undertaken.

[0004] BACKGROUND OF THE INVENTION

[0005] Spray-drying is a technology that is widely used for powder production in a large number of industries, including the chemical, food and pharmaceutical industries. Particularly in the pharmaceutical industries there is a need for aseptic spray drying in which the entire spray drying process is conducted under aseptic processing conditions, so that sterile powders suitable for medical use can be produced.

[0006] Aseptic spray drying is an enabling platform for, e.g., new modalities by allowing particle engineering needed to formulate complex injectables such as those for high load subcutaneous delivery. However, current aseptic spray drying technology requires complex facilities and large equipment requiring high capital investment and / or complex in loco sterilization procedures leading to high operational costs. This complexity can also exacerbate issues such as cost and supply disruption when problems arise, and batches of product are lost.

[0007] Single-use bioreactors have been developed for aseptic processing for use in biomanufacturing; the first single-use bioreactor was launched in 1996 by Wave Biotechnology” (Subramanian G. “Continuous Biomanufacturing: Innovative Technologies and Methods”, Wiley VCH Verlag GmbH & Co. KGaA: Weinheim, Germany, 2017: 151-152). Further, in the context of spray-drying, WO 2019 / 043007 A1 (Capsugel Belgium NV) describes the use of a single-use drying chamber and other single-use components, each of which can be pre-sterilized and sterile packaged for use. After performing spray-drying, all or some of these components can be replaced by removing a new, unused component from sterile packaging and installing the new component in place of the used component. However, these options fail to address completely the problems of the complexity and cost of the equipment and its operation. For example, with the single-use components of WO 2019 / 043007 A1 it is still necessary to remove these from their packaging prior to use and expose them to the external environment of the apparatus. This means that the environment of the apparatus must still be carefully cleaned and controlled to ensure that the components are not contaminated during their installation.

[0008] As a result, although single-use components have been widely adopted for biological and / or aseptic processing in other technologies, the size and complexity of the spray drying equipment means that they have had limited application for aseptic spray drying. (In particularly cleaning during the processing of biologies can be very complex). Further, it is even more complex to handle potent compounds using current aseptic spray drying techniques because of the conflicting requirements in under / over pressure of the chamber vs the environment arising from the requirements of sterility and personnel health and safety, respectively.

[0010] SUMMARY OF THE INVENTION

[0011] In a first aspect the present invention provides an assembly for installation into a support structure comprising a spray drying chamber and a product separator, or a section thereof or frame therefor, to form a spray drying apparatus for producing a spray-dried product and an outlet gas using a liquid feed and a drying gas, the assembly comprising:

[0012] - a gas distributor and an atomizer, the gas distributor and the atomizer being for attachment to the spray drying chamber of the support structure;

[0013] - one or more inlet conduits comprising a first inlet conduit for supplying the drying gas to the gas distributor;

[0014] - an outlet part for attachment to the product separator, or section thereof or frame therefor, of the support structure;

[0015] - one or more outlet conduits comprising a first outlet conduit comprised in the outlet part for conducting the outlet gas away from the spray drying apparatus; and

[0016] - a container arranged between the gas distributor, the atomizer, and the outlet part and connected directly or indirectly thereto, the container comprising one or more container walls that define at least a chamber part and a product separator part, wherein the chamber part is for lining the spray drying chamber of the support structure and the product separator part is for lining the product separator or section thereof of the support structure, or for attachment to the frame of the support structure.

[0017] In particular, the assembly of the invention can be used to produce particles by spray drying solutions or suspensions of ingredients (especially pharmaceutical ingredients and / or potent (toxic or potentially toxic) ingredients, which require particular care in production and handling) with the assurance of sterility and / or containment. The assembly can be installed on the support structure to form a spray drying apparatus, and can be used to produce the particles in a closed or partially closed system where any exchange of fluids (liquids, gases, powder streams) with the ancillary equipment (e.g. gas feed sources) and / or the environment in which the assembly and apparatus are situated, is mediated by isolation devices, so that the requirements for a sterile product and / or environmental control are met. These features additionally avoid the need for complex in situ cleaning and sterilization of equipment between processing runs. Therefore, equipment can be readily re-used by removing a used assembly, and installing a second assembly, ensuring less down-time between processing runs. Accordingly, in a second aspect the invention provides a method of producing an assembly as described above, the method comprising connecting the container directly or indirectly to the gas distributor, the atomizer, and the outlet part to produce the assembly. Optionally, the method may comprise sterilizing the assembly.

[0018] In a third aspect, the invention provides a process for producing a spray drying apparatus, the process comprising installing an assembly as described above into a support structure comprising a spray drying chamber and a product separator or section thereof, or frame therefor, wherein the process comprises:

[0020] - connecting the outlet part directly or indirectly to the product separator or section thereof, or frame therefor;

[0024] In a fifth aspect the present invention provides a method of spray drying comprising using a spray drying apparatus according to the fourth aspect to produce the spray-dried product and the outlet gas from the liquid feed and the drying gas.

As indicated above, spray drying is a well-established technology, in which a spray dried product (a powder) is formed by atomizing a liquid feed to create droplets, and the droplets are dried using a drying gas in a spray drying chamber. As such, suitable gas distributors and atomizers for use in the present invention are known and are commercially available. The gas distributor and atomizer of the assembly are usually together as one part, with the atomizer being part of the gas distributor, so that attachment of the gas distributor to the container also results in the atomizer being attached to the container. (For example, the one part may be a gas distributor with an imbedded atomizer e.g., as shown in Figures 6A to 6C.)

As described above, it must also be possible for the container to be attached either directly or indirectly to the gas distributor, the atomizer and the outlet part(s) in a manner that again prevents the escape of the components of the spray-drying process, and the ingress of contaminants such as microorganisms.

[0076] The container may be made from synthetic or natural materials such metallic materials and polymeric materials including plastic or rubber. In particular, the flexible portions of the container walls may be made from polymeric material. Suitable examples are flexible pharmaceutical grade materials (e.g., LDPE, polypropylene (PP) or polyamide (PA) having a typical thickness of between 50 to 350 pm (more typically 50.8 to 305 pm representing 2 to 12 mil (British Imperial)), or semi-rigid I rigid pharmaceutical grade materials e.g., HDPE, or a mix between the two. (It is noted that some of these materials may be slightly permeable to gases e.g., water vapour, and atmospheric gases such as oxygen, carbon dioxide and nitrogen, but this does not prevent their use in the invention since they are sufficiently impermeable to prevent any of the components of the spray-drying process from escaping from the assembly and / or to avoid contaminants, e.g., microorganisms, from the outside environment affecting the aseptic environment inside the assembly.)

As indicated above, the choice of material will be impacted by the range of process variables for the intended spray drying method and possible sterilization method. The connecting conduit may be positioned on a section of the chamber part that is configured to line a side wall of the spray drying chamber (2a, 2b) of the support structure (39), i.e. , a wall that is not diametrically opposite the atomizer. This enhances the safety of the spray drying of such components and also avoids the need for the additional cleaning requirements that are necessary with such products.

Claims

CLAIMS1 . An assembly (38) for installation into a support structure (39) comprising a spray drying chamber (2a, 2b) and a product separator (35a, 35b) or a section thereof, or a frame therefor (47a, 47b, 47c, 47d), to form a spray drying apparatus for producing a spray-dried product and an outlet gas using a liquid feed and a drying gas, the assembly comprising:- a gas distributor (1 ) and an atomizer (22), the gas distributor and the atomizer being for attachment to the spray drying chamber (2a, 2b) of the support structure;- one or more inlet conduits (23, 24, 25) comprising a first inlet conduit (24) for supplying the drying gas to the gas distributor;- an outlet part (26) for attachment to the product separator or section thereof or frame therefor of the support structure;- one or more outlet conduits (5) comprising a first outlet conduit comprised in the outlet part for conducting the outlet gas away from the spray drying apparatus; and- a container (27) arranged between the gas distributor, the atomizer, and the outlet part and connected directly or indirectly thereto, the container comprising one or more container walls (28, 29) that define at least a chamber part (3) and a product separator part (31 ), wherein the chamber part is for lining the spray drying chamber of the support structure and the product separator part is for lining the product separator or section thereof of the support structure, or for attachment to the frame of the support structure.

2. The assembly according to claim 1 , wherein the one or more inlet conduits comprise a second inlet conduit (23) for supplying the liquid feed to the atomizer (22).

3. The assembly according to claim 1 , wherein the assembly comprises a feed reactor and a feed conduit for delivering feed from the feed reactor to the atomizer (22).

4. The assembly according to claim 3, wherein the one or more inlet conduits comprises one or more feed reactor inlet conduits for supplying reactants to the feed reactor.

5. The assembly according to any preceding claim, wherein the one or more inlet conduits comprises a third inlet conduit (25) for supplying an atomizing gas to the atomizer (22).

6. The assembly according to any preceding claim, wherein the one or more outlet conduits comprises a second outlet conduit for removing the spray dried product from the assembly.

7. The assembly according to any preceding claim, comprising a plurality of isolation devices (7, 8, 9, 10), wherein at least one of the plurality of isolation devices is positioned on at least one of the one or more inlet conduits (23, 24, 25) and at least one other of the plurality of isolation devices is positioned on at least one of the one or more outlet conduits (5), wherein the plurality of isolation devices are to prevent contaminants from entering the assembly and / or to prevent the spray dried product escaping from the assembly during spray-drying.

8. The assembly according to claim 7, wherein at least one of the plurality of isolation devices (7, 8, 9, 10) is positioned on each of the one or more inlet conduits (23, 24, 25) and on each of the one or more outlet conduits (5).

9. The assembly according to claim 7 or claim 8, wherein the plurality of isolation devices (7, 8, 9, 10) are capable of isolating the internal environment of the assembly from the external environment.

10. The assembly according to any of claims 7 to 9, wherein the at least one isolation device is independently selected from a filter, or a valve.

11. The assembly according to claim 10, wherein the filter is a sterile filter and / or wherein the valve is a split-butterfly valve.

12. The assembly according to any of claims 7 to 11 , wherein the plurality of isolation devices comprises an isolation device (7) positioned on the first inlet conduit (24), which isolation device comprises an insulating layer (45) to reduce heat loss from the drying gas.

13. The assembly according to claim 12, wherein the insulating layer (45) comprises an envelope for the isolation device (7) for circulating a heated fluid (44) around the device.

14. The assembly according to any of claims 7 to 13, wherein the plurality of isolation devices comprises an isolation device (7) positioned on the first inlet conduit (24), which isolation device is part of the gas distributor (1 ).

15. The assembly according to any of claims 7 to 14, wherein the plurality of isolation devices (7, 8, 9, 10) comprises a plurality of filters, optionally a plurality of sterile filters.

16. The assembly according to any preceding claim, wherein the assembly has been sterilized such that the internal environment of the assembly is sterile.

17. The assembly according to any preceding claim, wherein the support structure comprises the product separator, and the product separator (35a, 35b) is a cyclone or an electrostatic separator.

18. The assembly according to claim 17, wherein the product separator (35a, 35b) is a cyclone.

19. The assembly according to any of claims 1 to 16, wherein the support structure comprises a frame (47a, 47b, 47c, 47d) for the product separator and wherein the product separator part (31 ) of the container comprises the product separator for attachment to the frame.

20. The assembly according to claim 19, wherein the product separator is a cyclone.

21. The assembly according to any of claims 1 to 16, wherein the support structure comprises: (a) a frame for a first section of the product separator; and (b) a second section of the product separator, and wherein the product separator part of the container comprises: (c) the first section of the product separator for attachment to (a); and (d) a segment for lining (b).

22. The assembly according to claim 21 wherein the one or more container walls that define (c) of the product separator part are rigid and the one or more container walls that define (d) of the product separator part are flexible.

23. The assembly according to claim 21 or claim 22, wherein the product separator is a cyclone and the first section is a cyclone barrel and the second section is a cyclone cone.

The assembly according to any preceding claim, wherein the container (27) comprises a connecting conduit (37) connecting the chamber part (3) to the product separator part (31 ), optionally wherein the connecting conduit is positioned on a section of the chamber part that is configured to line a side wall of the spray drying chamber (2a, 2b) of the support structure (39).

25. The assembly according to any preceding claim, wherein the one or more container walls comprise one or more flexible portions.

26. The assembly according to claim 25, wherein the one or more flexible portions are formed of a polymeric material.

The assembly according to claim 25 or claim 26, wherein the one or more flexible portions enable the assembly to exist in at least two arrangements, the at least two arrangements comprising an installed form as installed on the support structure and a collapsed form in which the dimensions of the container (27) are reduced as compared to the installed form.

28. The assembly according to claim 27, wherein at least a part of the container (27) is in the collapsed form.

29. The assembly according to any of claims 25 to 28, wherein the product separator part (31 ) comprises a bag (36) for collecting the spray-dried product.

30. The assembly according to claim 29, wherein the bag (36) is formed as an extension of the one or more flexible portions of the container walls defining the product separator part (31 ).31 . The assembly according to any of claims 25 to 30, wherein the one or more container walls that define the chamber part (3), the product separator part (31 ), and optionally the connecting conduit (39), comprise a single flexible portion.

32. The assembly according to any preceding claim, wherein the atomizer (22) is connected to or is part of the gas distributor (1 ).

33. The assembly according to any preceding claim, wherein the gas distributor (1 ) and atomizer (22) are formed as a single unit.

34. The assembly according to claim 33, wherein the gas distributor (1 ), the atomizer (22), the first inlet conduit (23), and optionally the second inlet conduit (24) are formed as a single unit.

35. The assembly according to any preceding claim, wherein the assembly comprises a heat shield (46) connected to the gas distributor (1 ) to protect the chamber part (3) of the container from heat from the drying gas entering the spray drying chamber from the gas distributor.

36. The assembly according to any preceding claim, wherein the chamber part (3) of the container is sealed to the gas distributor (1 ) and the atomizer (22), or wherein the chamber part is sealed to the heat shield of claim 35.

37. The assembly according to any preceding claim, wherein the product separator part (31 ) is sealed to the outlet part (26).

38. The assembly according to any preceding claim, wherein the assembly comprises one or more fixings (14, 15, 17, 18, 19) for fixing the assembly to the support structure.

39. The assembly according to any preceding claim, wherein the one or more container walls are formed of a single layer or are formed of two or more layers.

40. The assembly according to any preceding claim, which is provided in a sterile package.

41. A method of producing an assembly (38) according to any of claims 1 to 40, the method comprising connecting the container (27) directly or indirectly to the gas distributor (1 ), the atomizer (22) and the outlet part (26), to produce the assembly.

42. The method according to claim 41 , comprising sterilizing the assembly.

43. The method according to claim 42, comprising sterilizing the assembly with gamma irradiation or in an autoclave.

44. The method according to any of claims 41 to claim 43, comprising packaging the assembly.

A process for producing a spray drying apparatus, the process comprising installing an assembly (38) according to any of claims 1 to 41 into a support structure (39) comprising a spray drying chamber (2a, 2b) and a product separator (35a, 35b) or section thereof, or frame (47a, 47b, 47c, 47d) therefor, wherein the process comprises:- connecting the gas distributor (1 ) and the atomizer (22) directly or indirectly to the spray drying chamber (2a, 2b);- connecting the outlet part (26) directly or indirectly to the product separator (35a, 35b) or section thereof, or frame therefor;- fitting the chamber part (3) of the container into the spray drying chamber (2a, 2b); and- fitting the product separator part (31 ) into the product separator (35a, 35b) or section thereof, or attaching the product separator part to the frame.

46. The process according to claim 45, comprising:- connecting the first inlet conduit (24) to a drying gas source; and- connecting the first outlet conduit (5) to a gas exhaust unit or to a gas recycling unit.

47. The process according to claim 45 or claim 46, comprising:- connecting the second inlet conduit (23) to a liquid feed source; or- connecting the one or more feed reactor inlet conduits to one or more reactant sources.

48. The process according to any of claims 45 to 47, comprising:- connecting the second outlet conduit to a product collection part.

49. The process according to any of claims 45 to 48, comprising:- connecting the third inlet conduit (25) to an atomizing gas source.

50. The process according to any one of claims 45 to 49, wherein fitting the chamber part (3) into the spray-drying chamber (2a, 2b) and / or fitting the product separator part (31 ) into the product separator (35a, 35b) or section thereof comprises inflating the chamber part and / or the product separator part such that they form a lining for the spray drying chamber and / or the product separator or section thereof, respectively.51 . The process according to any one of claims 45 to 50, comprising securing the chamber part (3) to the spray drying chamber (2a, 2b) and securing theproduct separator part (31 ) to the product separator (35a, 35b) or section thereof, or to the frame.

52. A spray drying apparatus comprising an assembly (38) according to any of claims 1 to 40 installed into a support structure (39) comprising a spray drying chamber (2a, 2b) and a product separator (35a, 35b), or section thereof, or frame (47a, 47b, 47c, 47d) therefor.

53. The spray drying apparatus according to claim 52, wherein the spray drying apparatus is produced by the process of any one of claims 45 to 51 .

54. A method of spray drying comprising using a spray drying apparatus according to claim 52 or claim 53 to produce the spray-dried product and the outlet gas from the liquid feed and the drying gas.

55. A method of reusing the spray drying apparatus used in the method of claim 54, the method comprising uninstalling the assembly (38) after use of the spray drying apparatus, and installing a further assembly according to any of claims 1 to 40 into the support structure.
