Description

Technical Field

[0001] This invention relates to a method for determining the fingerprint spectrum of a traditional Chinese medicine composition. It belongs to the field of traditional Chinese medicine detection technology. Background Technology

[0002] Traditional Chinese medicine (TCM) fingerprinting refers to the chromatograms or spectra obtained by appropriate analysis of certain TCM materials or preparations after proper processing. It is a comprehensive and quantifiable identification method based on systematic research into the chemical components of TCM, primarily used to evaluate the authenticity, quality, and stability of TCM materials and semi-finished TCM preparations. Its prominent characteristics are "holistic" and "fuzzy" properties.

[0003] Traditional Chinese medicine (TCM) and its preparations are complex multi-component systems. Therefore, evaluating their quality requires appropriate detection methods that provide rich identification information. However, current methods such as microscopic identification, physicochemical identification, and content determination are insufficient to address this issue. Establishing a fingerprint spectrum for TCM can comprehensively reflect the types and quantities of chemical components contained in TCM and its preparations, thereby providing a holistic description and evaluation of drug quality. This aligns perfectly with the holistic theory of TCM. Furthermore, further research into spectroscopic efficacy can truly integrate the quality of TCM with its efficacy, helping to elucidate the mechanisms of action of TCM. In conclusion, the research and establishment of TCM fingerprint spectra are of great significance for improving the quality of TCM and promoting its modernization.

[0004] Currently, fingerprinting technology for traditional Chinese medicine (TCM) involves numerous methods, including chromatographic methods such as thin-layer chromatography (TLCS), high-performance liquid chromatography (HPLC), gas chromatography (GC), and high-performance capillary electrophoresis (HPCE), as well as spectroscopic methods such as ultraviolet (UV) spectroscopy, infrared spectroscopy (IR), mass spectrometry (MS), nuclear magnetic resonance (NMR), and X-ray diffraction. Chromatographic methods are the mainstream, especially HPLC, TLCS, and GC, which have become three recognized routine analytical techniques. HPLC is favored for its high separation efficiency, high selectivity, high detection sensitivity, fast analysis speed, and wide application range; most TCM components can be analyzed and detected on HPLC, and there is considerable experience in its application. Therefore, HPLC has become the preferred method for TCM fingerprinting technology. With the application of coupled techniques such as HPLC-MS and GC-MS, TCM fingerprinting technology is becoming increasingly sophisticated.

[0016] The feasibility of the fingerprint spectral determination method for the traditional Chinese medicine composition of the present invention was evaluated from multiple aspects using the sample prepared in Example 1. The evaluation method is as follows: 1. Instruments and Materials 1.1 Instruments Waters ACQUITY UPLC H-CLASS ultra-high performance liquid chromatograph (PDA detector, Empower 3.0 chromatography workstation), circulating multi-purpose vacuum pump (Zhengzhou Changcheng Science & Industry Trade Co., Ltd.), KQ250DB ultrasonic cleaner (Kunshan Ultrasonic Instrument Co., Ltd.), and Swiss METTLER TOLEDOAL electronic analytical balance.

[0017] 1.2 Materials 1.2.1 Reference Standard Information 1.2.2 Sample Information The traditional Chinese medicine composition of this invention (Shijiazhuang Yiling Pharmaceutical Co., Ltd.) 1.2.3 Reagents Acetonitrile (chromatographic grade, Merck, batch number JB091230), methanol (chromatographic grade, Merck, batch number l1061607948), phosphoric acid (analytical grade, Tianjin Kemeo Chemical Reagent Co., Ltd., batch number 20190518). 2. Selection of chromatographic conditions 2.1 Selection of detection wavelength The traditional Chinese medicine composition sample was subjected to full-wavelength determination. Chromatograms were extracted at different wavelengths (220nm, 254nm, 280nm, 300nm, and 330nm). After careful comparison, the most chromatographic peaks were extracted at a wavelength of 254nm, which can more comprehensively reflect the chromatographic information of the sample. Therefore, 254nm was selected as the detection wavelength.

[0018] 2.2 Selection of Mobile Phase System This study investigated the chromatographic peaks in methanol-water, methanol-0.1% formic acid aqueous solution, methanol-0.1% phosphoric acid aqueous solution, acetonitrile-water, and acetonitrile-0.1% formic acid aqueous solution. The results showed that the peak shapes were asymmetrical, and some peaks could not achieve baseline separation. The acetonitrile-0.1% phosphoric acid aqueous solution was investigated, and under this condition, the peak shapes were better and baseline separation was achieved. The results showed that the acetonitrile-0.1% phosphoric acid system had the best peak resolution; therefore, the mobile phase system was: acetonitrile-acid-water (0.1% phosphoric acid aqueous solution), flow rate: 0.3 mL / min. The gradient elution table is shown in Table 2. The chromatographic column was determined to be ACQUITY UPLC HSS T3 C18 1.8 μm, 2.1*100 mm; the following parameters were set as initial conditions: wavelength 254 nm; column temperature: 30℃; injection volume: 1 μL. .

[0019] 2.3 Selection of Flow Rate The effects of different flow rates (0.2 ml / min, 0.3 ml / min, and 0.4 ml / min) on the chromatograms were investigated. At a flow rate of 0.2 ml / min, compounds with longer elution times and larger peak areas exhibited poor peak symmetry. At a flow rate of 0.4 ml / min, most peaks failed to reach baseline separation. Considering all factors, the chromatograms at a flow rate of 0.3 ml / min showed better peak shapes and most peaks achieved near-baseline separation. Therefore, a flow rate of 0.3 ml / min was selected.

[0020] 2.4 Column Temperature Selection The chromatographic column was investigated at different column temperatures. At a column temperature of 25℃, the chromatographic peak shape was tailed. At 30℃, the chromatographic peak was excellent. At a column temperature of 35℃, the chromatographic peak with a larger peak area had poor symmetry. After comprehensive comparison, the column temperature of 30℃ was selected.

[0021] 2.5 Selection of Chromatographic Column Different chromatographic columns were investigated: ACQUITY UPLC HSS T3 C18 1.8μm, 2.1*100mm; CORTECS UPLC C18 1.6μm, 2.1*100mm; and ACQUITY UPLC BEH C18 1.7μm, 2.1*100mm. Based on a comprehensive comparison of the number and shape of chromatographic peaks, ACQUITY UPLC HSS T3 C18 1.8μm, 2.1*100mm was selected.

[0022] 2.6 Fingerprint chromatograms of reference standard and test sample Under the established chromatographic conditions, the reference solution and the test solution were injected separately. Fourteen major chromatographic peaks were identified in the chromatogram of the test solution. Based on the retention times and spectral characteristics of the peaks, the following peaks were identified in the fingerprint chromatogram: Peak 1: Neochlorogenic acid; Peak 2: Chlorogenic acid; Peak 3: Cryptochlorogenic acid; Peak 5: Isoflavone A; Peak 7: Forsythoside A; Peak 10: Hesperidin; Peak 12: Baicalin; Peak 13: Arctiin; Peak 17: Aloe-emodin; Peak 18: Ammonium glycyrrhizate; Peak 19: Rhein; Peak 20: Emodin; Peak 21: Rhein; Peak 22: Emodin methyl ether. The chromatograms and spectra of the reference and test solutions are attached. Figure 1-2 .

[0051] The traditional Chinese medicine composition described in this invention is a marketed traditional Chinese medicine product. Currently, the quality control of this product relies on the individual identification of some medicinal materials. However, as this is a complex traditional Chinese medicine product, limited identification is insufficient to reflect its quality. To better ensure product quality and protect public medication safety, we studied the fingerprint spectrum of this traditional Chinese medicine composition. Fingerprint spectrum studies were conducted on seven different batches of the drug. The fingerprint spectrum analysis identified 22 representative common peaks, and 14 of these peaks were identified. The separation of each peak was good. Using fingerprint spectrum for quality control can comprehensively reflect the overall quality of the traditional Chinese medicine product, effectively avoiding the drawbacks of simply adding only the individual components that need to be tested during the market inspection process.

[0052] The fingerprint spectrum detection method described in this invention is a complete technical solution, including several technical features. For each of these features, the inventors have conducted numerous experimental method studies. Although each technical feature may seem simple, the overall technical solution composed of countless features represents the optimal solution for quality control of the formulation. In particular, the fingerprint spectrum similarity among seven batches tested using this method is greater than 95%, indicating that the product is stable and controllable.

[0053] This method is simple to operate, highly feasible, and quick, solving the problem of quality control in large compound traditional Chinese medicine preparations. The experimental results show that this method has good precision, stability, and repeatability, providing a new approach to improve the quality control of this traditional Chinese medicine composition.
