By designing a multifunctional protein-DNA complex fluorescent probe, an ACP complex is formed using a DNA probe modified with avidin and biotin-labeled AP site 3' double mismatch. This complex binds to phenylboronic acid-modified AVD and enters the cell nucleus, solving the sensitivity and specificity problems of APE1 detection and imaging in existing technologies. This enables efficient and accurate detection and imaging of APE1 in the nucleus of living cells.
