[0004] This invention provides certain novel compounds that are glucocorticoid receptor agonists. This invention further provides certain novel compounds that are prodrugs for glucocorticoid receptor agonists. Furthermore, this invention provides certain novel compounds that are glucocorticoid receptor agonists and can be used to treat autoimmune and inflammatory diseases, such as atopic dermatitis, inflammatory bowel disease, rheumatoid arthritis, systemic lupus erythematosus, and lupus nephritis.

[0438] Process 17
[0439]
[0440] In process 17, under conditions known to those skilled in the art, the compound of structure 1, where R is H, reacts with the compound of structure 2, where Pg is a suitable nitrogen protecting group such as tert-butoxycarbonyl, and where G is:
[0441]
[0442] We obtained a compound of formula I, in which R is hydrogen.
[0443] More specifically, as shown in process 17A below, under conditions known to those skilled in the art, a compound of structure 1a, wherein R is H, reacts with a compound of structure 2, wherein Pg and G are as defined above, to give compounds of formula Ib and Ic.
[0444] Process 17A
[0445]
[0446] For example, about 1 equivalent of compound 1a (where R is H) and about 1 equivalent of compound 2 (where Pg and G are as defined in process 17) are suspended in a suitable organic solvent, such as acetonitrile. The suspension is cooled to about -10°C to about -25°C and then treated with about 5 equivalents of a suitable acid, such as perchloric acid (70% aqueous solution) or trifluoroacetic acid. The reaction mixture is then heated to room temperature and stirred for about 1 to 8 hours. Additional organic solvents, such as acetonitrile and dimethylformamide, may be added, and the mixture is stirred again for about 2 hours. The reaction is then quenched under standard conditions, such as with a saturated aqueous solution of sodium bicarbonate, and the products are separated using standard techniques well known in the art, such as extraction with a suitable organic solvent, such as dichloromethane:isopropanol, drying the organic extract with magnesium sulfate, filtering, and concentrating under vacuum to give a crude product mixture. The crude mixture can be purified using techniques well known in the art to separate the products of formula Ib and formula Ic, such as chromatography, including normal-phase chromatography, for example with a suitable eluent (e.g., DCM containing MeOH), and reversed-phase chromatography with a suitable eluent (e.g., 2:1 10 mM ammonium bicarbonate aqueous solution + 5% methanol:acetonitrile).

[0469] Alternative preparation in Example 5
[0470] All solids handling steps were performed using disposable glove bags, including filling flasks with 16α-hydroxyprednisolone, solids filtration, and solids transfer.
[0471] At -25°C, an aqueous perchloric acid solution (31 mL, 330 mmol, 10.6 mol / L) was added dropwise to a suspension of (25 g, 67 mmol, 2-fluoro-3-formyl-4-methoxyphenoxy)methyl)phenyl)carbamate (25 g, 65 mmol) in ACN (1000 mL) in a 2-L round-bottom flask. The mixture was stirred at -20°C while the reaction was monitored by LCMS. Internal temperature monitoring showed a small amount of exothermic reaction upon the addition of perchloric acid. After the addition of dry ice, the internal temperature was maintained below -19°C. The mixture was maintained below -19°C for 2 h and then heated to -10°C. The mixture was divided in two and added dropwise to two separate beakers (beaker 1 and beaker 2), each containing 76 mL of a 2 L aqueous solution of 5 M NaOH. The solid formed after the addition of perchloric acid and complete quenching was tested (pH 10). Each addition of viscous solid was transferred and combined into a third beaker (beaker 3) containing DCM containing 10% MeOH. The mixture was stirred until the viscous solid was completely dissolved. The remaining solution (beakers 1 and 2) was then filtered through filter paper to collect the residual solid. The separated solid was rinsed with water and transferred to beaker 3. Beaker 3 was stirred until all the solid was dissolved. The solution was dried with Na₂SO₄ for 2 hours, filtered, and evaporated to obtain a brown foam. The filtrate containing the collected solid was extracted with three portions of DCM containing 10% IPA. The combined organic extracts were dried with Na₂SO₄ overnight with stirring, then filtered and evaporated to obtain another crude residue. The crude product was dissolved in 150 mL of DCM containing 1% MeOH and purified by normal phase chromatography, eluting with DCM containing 0–5% MeOH to obtain a solid. It was diluted with 600 mL of EtOH and rotated on a rotary evaporator at 45 °C for 5 minutes. The water bath was heated to 60 °C. When the water bath reaches 60°C, turn off the water bath and rotate the flask until the water bath reaches room temperature. Remove the flask from the rotary evaporator, cap it, and leave it at room temperature overnight.
[0472] The solid was collected by vacuum filtration in a glove bag. The solid was washed with EtOH and dried on a filter for 5 h. The solid was redissolved in 1.5 L of DCM containing 20% EtOH. Additional DCM was added to obtain a nearly clear solution (some haze remained). Solvent exchange was achieved by rotary evaporation at 450 mbar and 45 °C. After distillation was stopped, the pressure was reduced to 100 mbar, allowing the EtOH to decrease to approximately 600 mL. The mixture was removed from the rotary evaporator and allowed to stand at room temperature for 5 h. The solid was collected by vacuum filtration and washed twice with additional EtOH. The filter cake was dried in a vacuum oven at room temperature for 48 h to give the title compound (42 g, 38% yield). MS m / z 634.6 (M+H).
