Abstract
Description
Technical Field
[0001] This invention relates to the field of cosmetics, and more particularly to a composition and cosmetics containing the composition. Background Technology
[0002] Advanced glycation end products (AGEs) are stable compounds produced by a series of non-enzymatic reactions, including condensation, rearrangement, cleavage, and oxidative modification, between reducing sugar carbonyl groups and amino acid groups in macromolecules such as proteins, lipids, and nucleic acids. With age, AGEs accumulate in human tissues, and their levels are one of the markers of physiological aging. AGEs accumulation in the skin causes cross-linking damage to proteins and binding to specific receptors, leading to various skin problems such as dullness, wrinkles, dark brown spots, decreased collagen elasticity, and even promoting the production of matrix metalloproteinases (MMPs) that break down collagen and elastin, as well as increasing the secretion of cytokines that can induce inflammatory responses. These factors ultimately lead to accelerated skin aging. Most existing anti-glycation products are oral preparations. For their effective anti-glycation components to reach the skin and exert their anti-glycation effects, they must pass through the digestive and circulatory systems, resulting in a long onset time and limited functionality.
[0003] Antioxidant is short for anti-free radical. The human body is constantly exposed to the external environment; factors such as respiration, air pollution, light pollution, and work stress cause the production of free radicals in the body. Skin aging is largely related to an excess of free radicals. Using antioxidant cosmetics can effectively remove excess free radicals, thereby delaying skin aging and maintaining skin elasticity. However, existing antioxidant products often have two drawbacks. First, they typically use a single antioxidant ingredient, resulting in a limited and insufficiently comprehensive antioxidant pathway, leading to generally weak overall antioxidant capacity and less than ideal antioxidant effects. Second, some ingredients pose an allergy risk; for example, pure vitamin C and retinol can easily irritate the skin and cause allergies, meaning the safety of these products is not ideal.
[0004] Therefore, how to balance anti-glycation, antioxidant, and safety properties in cosmetics is an urgent problem to be solved. Summary of the Invention

[0017] The technical solutions of this disclosure will be clearly and completely described below with reference to the embodiments thereof. Obviously, the described embodiments are only a part of the embodiments of this disclosure, and not all of the embodiments. Based on the embodiments of this disclosure, all other embodiments obtained by those skilled in the art without creative effort are within the scope of protection of this disclosure.
[0018] Throughout this specification, unless otherwise specified, the terminology used herein shall be understood as having the meaning as commonly used in the art. Therefore, unless otherwise defined, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art. In the event of any conflict, this specification shall prevail.
[0019] It should be noted that, in this disclosure, the term "comprising" or any other variation thereof is intended to cover non-exclusive inclusion, such that a method or apparatus that comprises a list of elements includes not only the elements expressly stated, but also other elements not expressly listed, or elements inherent to implementing the method or apparatus. Without further limitations, an element defined by the phrase "containing..." does not exclude the presence of other related elements in the method or apparatus that includes that element.
[0020] While existing cosmetics can maintain skin health through hydration, anti-oxidation, and sun protection, they cannot regulate the levels of advanced glycation end products (AGEs) in the body, meaning they cannot alleviate aging problems caused by AGEs. Most existing anti-glycation products are oral formulations; for effective anti-glycation ingredients to reach the skin and exert their anti-glycation effects, they must pass through the digestive and circulatory systems, resulting in a long onset time and limited functionality.
[0021] Furthermore, the mainstream anti-glycation ingredients currently available are mainly carnosine, pro-xylane, alpha-lipoic acid, grape seed extract, flavonoids, polyphenols, ferulic acid, and glucosamine. The mainstream antioxidant ingredients are mainly astaxanthin, anthocyanins, tea polyphenols, coenzyme Q10, grape seed extract, lutein, and resveratrol. These ingredients can easily irritate the skin, leading to allergies, meaning the safety of these products is not ideal.
[0022] On the other hand, existing antioxidant products usually use a single antioxidant ingredient, which makes the antioxidant pathway relatively simple and not comprehensive enough. As a result, the overall antioxidant capacity of the products is generally average and the antioxidant effect is not ideal.

[0056] The antioxidant, anti-glycation, and soothing properties of the compositions described in the examples and comparative examples were tested as follows. The reagents and instruments used were as described below.
[0057] Reagents: chloromethyl derivative of 2',7'-dichlorodihydrofluorescein diacetate (H2DCFDA) (5-(and 6-)chloromethyl-2',7'-dichlorodihydrofluorescein diacetate, acetyl ester, abbreviated as CM-H2DCFDA, used as an indicator of oxidative stress); bovine serum albumin, abbreviated as BSA (Sigma, M26HS122527); glucose (Sigma, SLBR0902V); phosphate buffered saline (PBS) (Solepro, P1010); aminoguanidine hydrochloride (Aladdin, G1909194); sodium dodecyl sulfate (SLS).
[0058] Instruments: Microplate reader (BioTek, Epoch); Vortex oscillator (Shanghai Qiqian Electronic Technology, MZX-2500).
[0059] Test Example 1: Antioxidant Properties Test of the Composition
[0060] Ultraviolet radiation is the most significant exogenous environmental factor causing skin damage and aging. It easily induces reactions such as inflammation, oxidative stress (when the production of free radicals exceeds the body's ability to eliminate them, oxidative stress occurs and can severely damage cell structure), and DNA damage. It can even disrupt the active structures of collagen and elastin, manifesting as redness, pigmentation, sagging, and fine lines. Zebrafish naturally contain oxygen free radicals (ROS). A universal oxidative stress indicator (CM-H2DCFDA) is used to react with ROS in zebrafish, and the fluorescence value of the resulting fluorescent product is detected. Therefore, changes in zebrafish ROS can be used to evaluate the antioxidant capacity of a sample.
[0061] Wild-type AB strain zebrafish, 2 days post-fertilization (2 dpf), were used as experimental subjects. The zebrafish were raised and bred according to the group standard T / HPCIA003—2022 "General Technical Specifications for Cosmetic Zebrafish Experiments" issued by the Guangzhou Development Zone Huangpu Cosmetics Industry Association. Antioxidant activity was tested following the procedures outlined in "Establishment and Application of Antioxidant Screening Model Based on Zebrafish Juveniles" (Liu Haibo et al., China Food Hygiene Journal, 2022, Vol. 34, No. 5, pp. 871-877).
[0062] Zebrafish were divided into three groups: one normal control group, one model control group, and eight sample groups (corresponding to Examples 1-4 and Comparative Examples 1-4, respectively), with 15 fish in each group. The following steps were performed, and the normal control group, model control group, and sample groups were collectively referred to as the experimental groups. Zebrafish from each experimental group were placed in a 6-well plate, with each well corresponding to one experimental group. 3 mL of feeding solution was added to the normal control group and the model control group, while 3 mL of feeding solution containing 0.5% of one of the compositions from Examples 1-4 and Comparative Examples 1-4 was added to each of the eight sample groups.
[0063] Zebrafish in each experimental group were incubated at 28℃ in the dark for 2 hours. Except for the normal control group, zebrafish in the other experimental groups were irradiated under ultraviolet light three times, each time for 15 minutes, with an interval of 30 minutes. Zebrafish in each experimental group were incubated at 28℃ in the dark for 48 hours. CM-H2DCFDA was added to both the model control group and each experimental group. Ten zebrafish were randomly selected from each experimental group and transferred to a 96-well microplate, one zebrafish per well, with 100 μL of culture medium per well. After incubation at 28℃ in the dark for 20 hours, the ROS fluorescence value (S) of zebrafish was detected using a microplate reader, and the average value of each experimental group was calculated. Antioxidant activity was calculated according to the following formula (1), where S represents the average value of the ROS fluorescence value of zebrafish, and the statistical P-value of antioxidant activity was calculated (P-value < 0.05 was considered statistically significant, the same below). The results are shown in Table 2 below.
[0064]
[0065] Table 2. Antioxidant activity (%) of the composition samples from Examples 1-4 and Comparative Examples 1-4.
[0066]
[0067] As shown in Table 2, the antioxidant properties of the samples in Examples 1 to 4 were significantly higher than those of Comparative Examples 1 to 4.
[0068] Test Example 2: Anti-glycation test of the composition
[0069] The compositions of Examples 1 to 4 were serially diluted with water to obtain sample solutions with contents of 6.25%, 12.5%, 25%, 50% and 100% (v / v) for each composition. The solutions were then filtered through a 0.22 μm filter membrane for sterilization and used for later use.
[0070] A mixed solution containing 80 mg / mL bovine serum albumin and 240 mg / mL glucose was prepared using 0.1 mol / L phosphate buffered saline (PBS), filtered through a 0.22 μm filter membrane, and used as the saccharification reaction solution.
[0071] Reaction systems W to Z were prepared according to the volume ratios of the saccharification reaction solution, sample solution, and 0.1 mol / L phosphate buffered solution (PBS) shown in Table 3. The final concentrations of bovine serum albumin (BSA) and glucose in reaction systems W to Z were 40 mg / mL and 120 mg / mL, respectively, with the contents of each composition being 3.125%, 6.25%, 12.5%, 25%, and 50%.
[0072] Table 3. AGEs Inhibition Test Reaction System
[0073]
[0074]
[0075] After mixing all systems thoroughly, they were incubated at 55°C for 40 h. PBS was used instead of the sample (reaction system Y) as the negative control, PBS was used instead of the saccharification reaction solution as the control system (reaction system X), PBS was used as the blank control (reaction system Z), and 1% aminoguanidine hydrochloride was used instead of the sample solution as the positive control. After the reaction, the incubated solution was cooled to room temperature, and 200 μL of the reaction solution was added to a 96-well plate. The fluorescence intensity of each reaction system was detected using a fluorescence microplate reader at an excitation wavelength of 370 nm and an emission wavelength of 440 nm. The AGEs inhibition rate was calculated according to the following formula (2), where F W F X F Y F Z The fluorescence intensities of W, X, Y, and Z represent the reaction system, respectively. If, after reacting with the glycosylation reaction solution at a certain concentration, the AGEs inhibition rate is >0% compared to the negative control group, and the P-value is <0.05, then the sample has the ability to inhibit AGEs formation. In other words, the higher the inhibition rate, the stronger the AGEs inhibition effect.
[0076]
[0077] The inhibition rates of AGEs by samples of different concentrations in Examples 1-4 are shown in Tables 4-7 below.
[0078] Table 4. Inhibition rate of AGEs by the samples in Example 1
[0079]
[0080] Table 5. Inhibition rate of AGEs in Example 2

[0082]
[0083] Table 6 Inhibition rate of AGEs in Example 3
[0084]
[0085] Table 7 Inhibition rate of AGEs in Example 4
[0086]
[0087] Furthermore, the AGES inhibition rate (%) of comparative samples 1-4 at a concentration of 25% was detected through the above operations, as shown in Table 8 below.
[0088] Table 8. AGES inhibition rate (%) of comparative samples 1–4 at a concentration of 25%.
[0089]
[0090] As shown in Tables 4 to 7, compared with the negative control group, the inhibition rate of AGEs by the samples of Examples 1 to 4 increased with the increase of sample concentration, and the anti-glycation effect also increased, but the growth curves were different. Among them, at a sample concentration of 25%, Example 4 had the highest inhibition rate of 30.66%, while Example 1 had the lowest inhibition rate of 19.58%.
[0091] As shown in Table 8, at a sample concentration of 25%, Comparative Examples 1 to 4 did not contain formulations A, B, C, and D, respectively, but the total amount of formulations contained was the same as in each example, and their AGEs inhibition rates were all lower than those in Example 1.
[0092] Test Example 3: Soothing effect test of the composition
[0093] Sodium dodecyl sulfate (SLS) may trigger a stimulating response in the body. When this stimulant enters the zebrafish, it induces an inflammatory response, causing neutrophils to mount an immune response, migrate to the skin epidermis, and aggregate. The soothing effect of the sample was determined by detecting changes in the number of neutrophils in the skin of transgenic zebrafish strain MPX with green fluorescent neutrophils before and after treatment.
[0094] Transgenic neutrophil-producing green fluorescent zebrafish (MPX) 2 days after fertilization (2dpf) were used as experimental subjects. The zebrafish were raised and bred in accordance with the standards of our laboratory (accreditation number: 001458) which meets the accreditation requirements of the International Committee for Assessment and Accreditation of Laboratory Animals (AAALAC).
[0095] Zebrafish were divided into three groups: one normal control group, one model control group, and four sample groups (corresponding to Examples 1, 2, 3, and 4, respectively), with 15 fish in each group. The following steps were performed: the normal control group received no treatment, the model control group received only SLS treatment, and the sample groups received SLS treatment along with the compositions from each example. The normal control group, model control group, and sample groups were collectively referred to as the experimental groups. Zebrafish from each experimental group were placed in a 6-well plate, with each well corresponding to one experimental group. For each experimental group, SLS was administered in water to establish a zebrafish skin inflammation model. The normal control group and model control group received 3 mL of culture medium, while the four sample groups received 3 mL of culture medium containing 0.5% of the composition from Example 1, 3 mL of culture medium containing 0.5% of the composition from Example 2, 3 mL of culture medium containing 0.5% of the composition from Example 3, and 3 mL of culture medium containing 0.5% of the composition from Example 4, respectively. The zebrafish in each experimental group were incubated at 28°C in the dark for 18 hours. Ten zebrafish were selected from each experimental group and photographed under a fluorescence microscope. The data were analyzed and collected using image processing software. The number of neutrophils (N) in the zebrafish skin was analyzed. The statistical P-values of soothing and calming effects were calculated according to the following formula (3). The results are shown in Table 9 below.
