本发明属于基因工程技术领域,公开了一种大片段DNA组装方法及其应用。本发明将大片段DNA通过PCR方式转变为小片段DNA,然后将小片段DNA交替整合到含有两种不同抗性基因(氯霉素抗性基因、四环素抗性基因)的pBR322质粒上,接着再将这些新构建的质粒依次转化到底盘细胞中,实现大片段DNA的组装。本发明大片段DNA组装方法可方便、高效地对酿酒酵母染色体进行组装,为人工设计合成酿酒酵母长染色体提供了基础。同时,采用本发明组装方法能够在枯草芽孢杆菌体内组装一条完整的PNP降解途径,并成功构建了高效PNP矿化工程菌,该方法构建的工程菌具有与野生菌株相似的生长趋势,外源基因能够稳定表达,实现PNP矿化功效,为农业环境的修复提供一种可行方案。
